The effects of ethephon on stomatal resistance, water potential, osmotic potential, turgor potential, and ethylene production were determined on leaves of a drought-resistant (KS 65) and a drought-sensitive (IA 25) genotype of sorghum [Sorghum bicolor (L.) Moench] grown under wellwatered or drought-stressed conditions. With both sufficient and limited water supply, ethephon had no effect on the adaxial, abaxial, or total stomatal resistance of either genotype. For both water treatments, the adaxial stomatal resistance of the drought-sensitive genotype was higher than that of the drought-resistant genotype. Ethephon increased the amount of ethylene produced by the plants under both levels of water. For plants with sufficient water, water potentials of both genotypes were lowered by ethephon. Ethephon had no effect on the water potentials under drought or on the osmotic potentials under either water regime. With drought, the turgor potential of the drought-sensitive genotype, but not that of the drought-resistant, was increased by ethephon.  相似文献

The incidence of lethal parasites in the larvae of a Tipula paludosa population was monitored for two seasons. The proportions of larvae infected with Tipula iridescent virus (TIV) and a tachinid insect were similar to those in previously studied populations, whereas the proportions of larvae infected with Tipula nuclear polyhedrosis virus (NPV) and a spore-forming bacterium (SFB) were higher. Conservative estimates of mortality due to these four agents were 10.7% in 1977–1978 and 7.7% in 1978–1979. The mean population density and the proportion of SFB-infected larvae were lower in 1978–1979 than in 1977–1978, while the proportion of NPV-infected larvae was higher. In 1979 the proportion of NPV-infected larvae was positively correlated with population density, which was highest in the wettest part of the study area. In both seasons the proportion of SFB-infected larvae was negatively correlated with population density. Larvae infected with the NPV or the SFB became pallid at an advanced stage of infection, but, although infected larvae were found throughout the larval period, pallid larvae were only found in the later part. It is suggested that larvae become infected in an early instar, then the infections slowly develop throughout the remainder of the larval period. Five larvae were found with mixed infections; four were infected with the SFB and NPV, while the fifth was infected with the SFB and TIV.  相似文献

Water and osmotic potential energies were measured with thermocouple psychrometers, at intervals during a 4-week period, in growing leaves of bean (Phaseolus vulgaris, var. Blue Lake) and barley (Hordeum vulgare, var. Liberty) plants having roots equally split between 2 differentially salinized nutrient solutions. The osmotic potentials of plants with half their roots in saline solutions were about halfway between the osmotic potentials of plants grown in nonsaline solutions and those grown in saline solutions. By the end of the 4-week measurement period, the beans and barley were almost mature. The final dry weights of shoots of plants with half their roots in saline solutions were about halfway between the dry weights of the shoots of plants grown in nonsaline solutions and the dry weights of those in saline solutions. The results obtained showed that the degree of osmotic adjustment and the rate of growth were functions of the proportion of the root system exposed to saline conditions.  相似文献

Summary Water and osmotic potentials were measured with thermocouple psychrometers, weekly after heading, two times each day at pre-dawn and at noon, in flag leaves and grain of tall and short cultivars of winter wheat grown in the field under rain-fed conditions.Water was held with less tension in the grain than in the leaf for both tall and short cultivars. The tall cultivars had lower leaf water potentials, but higher grain water potentials, than the short cultivars. The grain osmotic potential was lower in the short cultivars compared to the tall ones. Grain yield of short cultivars (1810 kg/ha) was more than that of tall cultivars (1730 kg/ha). Apparently higher leaf water potentials of short cultivars enabled more photosynthates to move into the grain.  相似文献

A major clinical need exists for cartilage repair and regeneration. Despite many different strategies having been pursued, the identification of an optimised cell type and of pre-treatment conditions remains a challenge. This study compares the cartilage-like tissue generated by human bone marrow stromal cells (HBMSCs) and human neonatal and adult chondrocytes cultured on three-dimensional (3D) scaffolds under various conditions in vitro and in vivo with the aim of informing future cartilage repair strategies based upon tissue-engineering approaches. After 3 weeks in vitro culture, all three cell types showed cartilage-like tissue formation on 3D poly (lactide-co-glycolide) acid scaffolds only when cultured in chondrogenic medium. After 6 weeks of chondro-induction, neonatal chondrocyte constructs revealed the most cartilage-like tissue formation with a prominent superficial zone-like layer, a middle zone-like structure and the thinnest fibrous capsule. HBMSC constructs had the thickest fibrous capsule formation. Under basal culture conditions, neonatal articular chondrocytes failed to form any tissue, whereas HBMSCs and adult chondrocytes showed thick fibrous capsule formation at 6 weeks. After in vivo implantation, all groups generated more compact tissues compared with in vitro constructs. Pre-culturing in chondrogenic media for 1 week before implantation reduced fibrous tissue formation in all cell constructs at week 3. After 6 weeks, only the adult chondrocyte group pre-cultured in chondrogenic media was able to maintain a more chondrogenic/less fibrocartilaginous phenotype. Thus, pre-culture under chondrogenic conditions is required to maintain a long-term chondrogenic phenotype, with adult chondrocytes being a more promising cell source than HBMSCs for articular cartilage tissue engineering.  相似文献

Functional analysis of kinetochore assembly in Caenorhabditis elegans   总被引:7,自引:0,他引:7  
In all eukaryotes, segregation of mitotic chromosomes requires their interaction with spindle microtubules. To dissect this interaction, we use live and fixed assays in the one-cell stage Caenorhabditis elegans embryo. We compare the consequences of depleting homologues of the centromeric histone CENP-A, the kinetochore structural component CENP-C, and the chromosomal passenger protein INCENP. Depletion of either CeCENP-A or CeCENP-C results in an identical "kinetochore null" phenotype, characterized by complete failure of mitotic chromosome segregation as well as failure to recruit other kinetochore components and to assemble a mechanically stable spindle. The similarity of their depletion phenotypes, combined with a requirement for CeCENP-A to localize CeCENP-C but not vice versa, suggest that a key step in kinetochore assembly is the recruitment of CENP-C by CENP-A-containing chromatin. Parallel analysis of CeINCENP-depleted embryos revealed mitotic chromosome segregation defects different from those observed in the absence of CeCENP-A/C. Defects are observed before and during anaphase, but the chromatin separates into two equivalently sized masses. Mechanically stable spindles assemble that show defects later in anaphase and telophase. Furthermore, kinetochore assembly and the recruitment of CeINCENP to chromosomes are independent. These results suggest distinct roles for the kinetochore and the chromosomal passengers in mitotic chromosome segregation.  相似文献
