Much of our current understanding about neurodegenerative diseases can be attributed to the study of inherited forms of these disorders. For example, mutations in the presenilin 1 and 2 genes have been linked to early onset familial forms of Alzheimer''s disease (FAD). Using the Drosophila central nervous system as a model we have investigated the role of presenilin in one of the earliest cellular defects associated with Alzheimer''s disease, intracellular calcium deregulation. We show that expression of either wild type or FAD-mutant presenilin in Drosophila CNS neurons has no impact on resting calcium levels but does give rise to deficits in intracellular calcium stores. Furthermore, we show that a loss-of-function mutation in calmodulin, a key regulator of intracellular calcium, can suppress presenilin-induced deficits in calcium stores. Our data support a model whereby presenilin plays a role in regulating intracellular calcium stores and demonstrate that Drosophila can be used to study the link between presenilin and calcium deregulation. 相似文献

The alpha and beta-adrenergic responses of the isolated muscle of sheep rumen were analysed by pharmacodynamic methods after administration of alpha and beta-adrenergic agonists and alpha and beta-adrenergic antagonists. It was found that phenylephrine, and in a lower degree propranolol, stimulated contractions of isolated muscle of sheep rumen while adrenaline, noradrenaline, isoprenaline, phenoxybenzamine and regitine inhibited these contractions. Propranolol abolished the dilating (atonic) effect of catecholamines on the isolated muscles of sheep rumen and previous blockade of beta-adrenergic receptors with propranolol reversed the dilating effects of catecholamines. The Drosophila neuromusculin (nrm) gene encodes an immunoglobulin-like (Ig-like) cell adhesion molecule expressed in the precursors of the embryonic peripheral nervous system (PNS), in the midline precursors of the central nervous system (CNS), and in muscles. During the initial phases of CNS axonogenesis, nrm is expressed in cells involved in the development of commissures and longitudinal tracts. Mutations which alter expression of nrm mRNAs cause aberrant development of commissures and longitudinal axon pathways. Defects in the PNS and muscles of nrm mutants are also observed. In most nrm embryos, abnormal development can be detected in a subset of abdominal segments; however, in approximately 1 of 10 nrm embryos, the defects extend to all segments. Herein, we present evidence that nrm plays an important role in early morphogenesis, possibly by mediating or facilitating inductive cell contacts and movements. 相似文献

Detailed study of the ultrastructures on the cortical lists of retiolitids occurring after the lundgreni event has been undertaken. The extremely well preserved material from the Go?dap borehole of Poland (Baltica) enables classification of clathrial lists into two types—with single and double seams. Significantly different seam development is observed between two groups of retiolitids occurring before and after the lundgreni event. The study of seams and increments on the lists enables reconstruction of the membrane arrangement of the rhabdosomes. Reconstruction of the proximal ends, the arrangement of the ancora umbrella, and the layout of the proximal ventral and lateral orifices of five genera, Neogothograptus, Holoretiolites, Plectograptus, Spinograptus, and Quattuorgraptus n. gen., is presented. The new genus Quattuorgraptus is separated from Spinograptus because of the different development of the ancora umbrella and the width of the lateral walls of the rhabdosome. 相似文献

Capsule The division coefficient is an estimate of the proportion of ringed birds migrating to different destination areas taking into account area‐specific re‐encounter probabilities. Aims To explore precision and bias of the division coefficient method by a simulation study and to compare the approach with multi‐state models. Methods In a simulation study true and estimated division coefficients were compared. The division coefficient method was mathematically compared with the multi‐state model. Results The estimated division coefficients seemed to be unbiased if the assumptions were met. The precision decreased if the bird distribution became similar in both bird groups and when difference between area‐specific re‐encounter probabilities increased. A bootstrap method to assess precision is presented. The estimates from the division coefficient method equal the maximum likelihood estimates in a multi‐state model including only one time interval. Conclusion Before applying the division coefficient method or a multi‐state model to real data a simulation study should be conducted in order to explore the behaviour of parameter estimation. The division coefficient method with the bootstrap confidence intervals is an easy alternative to a multi‐state model with one time interval when the bird distribution between destination areas (e.g. migratory connectivity) alone is of interest. 相似文献
