A novel complex mutation with the presence of both deletion and insertion in very close proximity in the same region was detected in exon 8 of the LDL receptor gene from two apparently unrelated Japanese families with familial hypercholesterolemia (FH). In this mutant LDL receptor gene, the nine bases from nucleotide (nt) 1115 to nt 1123 (AGGGTGGCT) were replaced by six different bases (CACTGA), and consequently the four amino acids from codon 351 to 354, Glu-Gly-Gly-Tyr, were replaced by three amino acids, Ala-Leu-Asn, in the conserved amino acid region of the growth factor repeat B of the LDL receptor. The nature of the amino acid substitution and data on the families suggest that this mutation is very likely to affect the LDL receptor function and cause FH. The generation of this complex mutation can be explained by the simultaneous occurrence of deletion and insertion through the formation of a hairpin-loop structure mediated by inverted repeat sequences. Thus this mutation supports the hypothesis that inverted repeat sequences influence the stability of a given gene and promote human gene mutations. 相似文献

Summary An integrated system has been constructed to instantly identify and efficiently sort the heterokaryons formed by plant protoplast fusion. The system is composed of the following functions: a) a transport system, b) an electro-manipulator, c) a cell harvester, d) a flow cytometer/cell sorter, and e) a control device. The conditions for an efficient and reproducible enrichment of the heterokaryons have been investigated by this system using the fluorescein isothiocyanate stained protoplasts preparing from Glycyrrhiza glabra cell cultures and unstained protoplasts of Abrus precatorius cell cultures which contain a large quantity of chlorophyll.Abbreviations MS

Protein synthesis during germination at 24?C and vernalizationat 4?C in winter wheat embryos were investigated with a cell-freesystem. During germination, the capacity for protein synthesisincreased in the early stage between 12 and 36 hr of imbibitionthen declined to a final low level between 48 and 72 hr. Thistransition was due to quantitative changes of the activitiesof ribosomal and supernatant fractions in the early stage andmainly to those of the supernatant fraction in the later stage.During vernalization, the capacity for protein synthesis continuedto decline over 15 to 60 days at 4?C. This transition was dueto the change in activity of the supernatant fraction; the activityof the ribosomal fraction was nearly constant. Electrophoretic analysis of in vitro products indicated thatthe high molecular weight proteins present in 12-hr embryoshad disappeared in 48-hr germinated wheat embryos and that theproducts in 24- and 36-hr embryos were types intermediate betweenthose of 12- and 48-hr embryos. The products in each vernalizedembryo resembled those in 24- and 36-hr germinated embryos.Therefore, it was concluded that the mRNA species for translationchanged during germination and vernalization in winter wheatembryos. Of 625 aphidicolin-resistant clones selected at 33.5°C from mutagenized mouse FM3A cells, 13 clones could not grow at 39.5°C. Five of these clones, chosen at random, resumed growth at 39.5°C when thymidine was added to the culture medium. In hybrids, conditional thymidine auxotrophy was a recessive trait, but aphidicolin-resistance was either a codominant or recessive one depending on the mutant clone used.Thymidylate synthetase activity in crude extracts of these mutants was completely inactivated by preincubation for 30 min at 42°C, whereas that of the parent cells was not affected by the same treatment. Thus, the temperature-sensitive growth of the mutants described here seems to be due to this heat-sensitive thymidylate synthetase. 相似文献

Large-scale cDNA analysis provides several great advantages for genome investigations in rice. Isolated and partially characterized cDNA clones have contributed not only to the construction of an RFLP linkage map and physical maps of the chromosomes but also to investigations of the mechanisms of expression of various isozymes and family genes. The ultimate aim of our large-scale cDNA analysis is to catalogue all the expressed genes of this important cereal, including tissue-specific, developmental stage-specific, and stress-specific genes. As of August 1996, the Rice Genome Research Program (RGP) has isolated and partially sequenced more than 29000 cDNA clones from various tissues and calluses in rice (Nipponbare, a japonica variety). The sequence data were translated into amino acid sequences for the 3 possible reading frames, and the similarity of these amino acid sequences to known proteins registered in PIR were examined. About 25% of the clones had significant similarities to known proteins. Some of the hit clones showed library-specific distributions, indicating that the composition of the clones in each library reflects, to some extent, the regulation of gene expression specific to differentiation, growth condition, or environmental stress. To further characterize the cDNA clones, including unknown clones, nucleotide sequence similarities of 24728 clones were analyzed and the clones were classified into around 10000 independent groups, suggesting that around a half or one third of expressed genes in rice have already been captured. These results obtained from our large-scale cDNA analysis provide useful information related to gene expression and regulation in rice. 相似文献

A series of fatty acid ethyl esters ranging from C14 to C24 was isolated from a hexane extract of the liverwort Conocephalum conicum, these esters accounted for 77% of the extract.
