Cercomonads are important components of microbial food webs in soils and aquatic sediments. Here, we investigated the general morphology, behaviour, life cycle and 18S rDNA phylogeny of cercomonad cultures from a German grassland soil habitat. We describe ten new species including two new genera from 23 strains. Three Cercomonas, two Eocercomonas and three Paracercomonas species are described. Based on large phylogenetic distance and distinct morphology, we erect two novel clade B genera near the root of the cercomonad tree. Nucleocercomonas nov. gen. bears a number of characters unusual for cercomonads: Its anterior flagellum is extremely long, it mostly does not glide, and in its most frequent life stage the cell body does not attach to the substratum, but produces unattached pseudopodia. Furthermore, it has a unique nucleus with a peripheral nucleolus that attaches to the nuclear envelope opposite the basal body connection. Metabolomonas nov. gen. is extremely metabolic. It is characterized by a very high beating frequency of the anterior flagellum, fast gliding, rapid changes in shape and strong cytoplasmic streams. A new genus Brevimastigomonas is erected for the previously described species Paracercomonas anaerobica. The general morphology of cercomonad species often does not correspond with their phylogenetic position: closely related species may have a very different morphology. 相似文献

The photosynthetic apparatus of purple bacteria is generally considered a well-studied and understood system. However, recent atomic force microscopy images of flattened chromatophore vesicles from Rhodobacter sphaeroides restarted a debate about the stoichiometry and positions of the membrane proteins, with the interpretations of the observed images only partly being in agreement with earlier models. The most puzzling observation from the recent images is that the Cytochrome bc(1) complex, which is a central part of the photosynthetic apparatus, seems to be missing on the chromatophore vesicles, even when these were extracted from photosynthetically grown bacteria. From the available information on the geometry of the vesicle and of the proteins we reconstructed here a three-dimensional model vesicle at molecular resolution. Its central feature, also determining its diameter of approximately 45 nm, is an equatorial array of LH1 dimers, lined by a region of LH2 rings. This naturally puts the Cytochrome bc(1) complexes and the ATPase at the vesicle's poles. This spatial model may explain why the vesicle's endcaps with the bc(1) complexes are lost during the preparatory steps of the imaging process together with the ATPase and are therefore absent from the available images. 相似文献

Metabolic processes in biological cells are commonly either characterized at the level of individual enzymes and metabolites or at the network level. Often these two paradigms are considered as mutually exclusive because concepts from neither side are suited to describe the complete range of scales. Additionally, when modeling metabolic or regulatory cellular systems, often a large fraction of the required kinetic parameters are unknown. This even applies to such simple and extensively studied systems like the photosynthetic apparatus of purple bacteria. Using the chromatophore vesicles of Rhodobacter sphaeroides as a model system, we show that a consistent kinetic model emerges when fitting the dynamics of a molecular stochastic simulation to a set of time dependent experiments even though about two thirds of the kinetic parameters in this system are not known from experiment. Those kinetic parameters that were previously known all came out in the expected range. The simulation model was built from independent protein units composed of elementary reactions processing single metabolites. This pools-and-proteins approach naturally compiles the wealth of available molecular biological data into a systemic model and can easily be extended to describe other systems by adding new protein or nucleic acid types. The automated parameter optimization, performed with an evolutionary algorithm, reveals the sensitivity of the model to the value of each parameter and the relative importances of the experiments used. Such an analysis identifies the crucial system parameters and guides the setup of new experiments that would add most knowledge for a systemic understanding of cellular compartments. The successful combination of the molecular model and the systemic parametrization presented here on the example of the simple machinery for bacterial photosynthesis shows that it is actually possible to combine molecular and systemic modeling. This framework can now straightforwardly be applied to other currently less well characterized but biologically more relevant systems. 相似文献

Effects of the water discharge on periphyton abundance and diversity in a large river (River Danube,Hungary) 总被引:4,自引:2,他引:2
This paper examines the relevance of intermediate disturbance theory in the context of in a large river the algal flora attaching to artificial substrata. It was observed that the theory is in good agreement with most of the communities analysed, however, with different frequency intervals compared to phytoplankton. Floods provided the most significant disturbances. As a rough generalization, we can say that in case of floods following each other in 8–14 days, respectively, the disturbance is of medium frequency. In case of 8 days, high frequency is more typical while in case of 14 days, low frequency is more characteristic. Owing to the fact that riverine periphytic algal organisms are attached in different ways and with differing efficiencies, the changes in the total mass occurring as a result of changes in water discharge lead to changes in diversity. The analysis of individual numbers can help in investigating the intensity of the disturbing effect on the algal communities. Negative correlation was found between water discharge and numbers of individuals, that is, the numbers of algae increase with low water discharge and decrease with high water level in the river. This second observation is valid only within a certain range of discharges (in this case up to 100 m3 s–1), which will be specific to particular rivers and individual reaches. Below the upper limit of the range, the increased water flows carry more nutrients to the organisms but with no severe wash-off effect. Beyond the upper limit (in this case in range 100–400 m3 s–1) the numbers of individuals is roughly in inverse proportion to the wash-off effect of the current, and the community is in equilibrium. In cases when discharge surpasses 400 m3 s–1 decrease in individual numbers is general. We suggest that in the environments similar to the study area, the periphyton of the artificial substrata (e.g., pontoons) undergo similar processes. In respect of the periphyton formed on the surface of the riverbeds in large rivers with considerable changes in water level, a more complex system can be supposed where part of the periphyton is periodically dried and flooded alternately. 相似文献

Periphyton was collected from sand-blasted slides exposed in the main channel of River Danube at Gö (1669 rkm). Samples were taken from 21 May to 23 November in 1984. Three to four days after placing the substrates into the river the first traces of an observable coating appeared. The formation of the algal coating in the Danube presented certain periodical features. From time to time, an essential decrease in the number of individuals could be observed, which generally coincided with changes in the struc- ture of the community. This could be traced well, by grouping the algae according to morphological categories and following the temporal changes affecting these categories. The early phase of the colo- nization process is dominated by relatively large araphid and biraphid diatoms. In the second phase small bi- and monoraphid species are dominant. Finally, the medium size mono- and biraphid algae are dominating. 相似文献

Seasonal dynamics of all major protozoan groups were investigated in the plankton of the River Danube, upstream of Budapest (Hungary), by bi-weekly sampling over a 1-year long period. Sixty-one heterotrophic flagellate, 14 naked amoeba, 50 testate amoeba, 4 heliozoan and 83 ciliate morphospecies were identified. The estimated abundance ranges of major groups throughout the year were as follows: heterotrophic flagellates, 0.27–7.8×106 ind. l?1; naked amoebae, max. 3300 ind. l?1; testaceans, max. 1600 ind. l?1; heliozoans, max. 8500 ind. l?1; ciliates, 132–34,000 ind. l?1. In terms of biovolume, heterotrophic flagellates dominated throughout the year (max. 0.58 mm3 l?1), and ciliates only exceeded their biovolume in summer (max. 0.76 mm3 l?1). Naked amoeba and heliozoan biovolume was about one, and testacean biovolume 1–3, orders of magnitude lower than that of ciliates. In winter, flagellates, mainly chrysomonads, had the highest biomass, whilst ciliates were dominated by peritrichs. In 2005 from April to July a long spring/summer peak occurred for all protozoan groups. Beside chrysomonads typical flagellates were choanoflagellates, bicosoecids and abundant microflagellates (large chrysomonads and Collodictyon). Most abundant ciliates were oligotrichs, while Phascolodon, Urotricha, Vorticella, haptorids, Suctoria, Climacostomum and Stokesia also contributed significantly to biovolume during rapid succession processes. In October and November a second high protozoan peak occurred, with flagellate dominance, and slightly different taxonomic composition. 相似文献

Some species of purple bacteria as, e.g., Rhodobacter sphaeroides contain the protein PufX. Concurrently, the light harvesting complexes 1 (LH1) form dimers of open rings. In mutants without PufX, the LH1s are closed rings and photosynthesis breaks down, because the ubiquinone exchange at the reaction center is blocked. However, the main purpose of the LH1 is light harvesting. We therefore investigate the effects that the PufX-induced dimerization has on the absorption properties of the core complexes. Calculations with a dipole model, which compare the photosynthetic efficiency of various configurations of monomeric and dimeric core complexes, show that the dimer can absorb photons directly into the reaction centers more efficiently, but that the performance of the more sophisticated dimeric LH1 antenna degrades faster with structural perturbations. The calculations predict an optimal orientation of the reaction centers relative to the LH1 dimer, which agrees well with the experimentally found configuration. Based on experimental observations indicating that the dimeric core complexes are indeed rather rigid, we hypothesize that in PufX+ species the association between the LH1 and the reaction centers is enhanced. This mechanical stabilization of the core complexes would lead to the observed quinone blockage, when PufX is missing. 相似文献
