Sewage of Marseilles' main outfall permanently pollutes a large coastal area centered around Cortiou, south of the city. In order to study the impact of that urban pollution on the zooplankton, more than 200 samples were collected between 1977 and 1981, according to several sampling strategies.Quantitatively, the study area showed a rather poor zooplankton. The more important populations were encountered near Cortiou, the non-perturbated reference point with lowest abundance of organisms. Sampling sites located near the outfall are sometimes azoic. Qualitatively, the observed communities are not characteristic of a heavily polluted environment, but correspond to an impoverished neritic community. In the more polluted area, the community is organized around the copepods Clausocalanus sp., Paracalanus sp. and Oithona helgolandica, and a group of less important species (Oithona nana and the cladoceran Evadne spinifera). Centropagidae, Coryceidae, Onceidae, but also Chaetognathians, Fritilliarins and the meroplanktonic larvae are more frequently encountered in clean water. Community structure is higher during the cold months than summer. The latter period frequently shows a disorganized zooplankton. In most situations, the copepod Acartia clausi plays a minor role in the structural definition of the communities.The variations observed seem largely independent of the parameters reflecting pollution intensity. Stress integration, in relation with the anterior community history (intensity of contact with polluted water, trophic potential of the area) seem to be the main regulator factors.
Impact d'une pollution urbaine sur la partie zooplanctonique d'un systeme neitique (Marseille - Cortiou)
Resumé Le rejet permanent du grand émissaire de Marseille (5 m3, sec–1) perturbe considérablement, par son importance, le système néritique du secteur de Cortiou. Afin d'approcher l'impact de cette pollution sur la partie zooplanctonique du système, plus de 200 prélèvements, concernant l'hydrologie et le plancton, ont of é\'t é réalises entre 1977 et 1981, selon différentes strategiés d'échantillonnage (suivi de masse d'eau, radiales, réseaux).Quantitativement on observe, sur l'ensemble de la zone étudiée, une abondance générale en zooplancton moyenne, voire faible. Les effectifs les plus importants se rencontrent cependant dans la cuvette de Cortiou, alors que le point de référence considéré comme non perturbé présente les effectifs les plus faibles. Les stations situées face à l'égout sont parfois azoïques.Qualitativement les peuplements ne paraissent pas trés caractéristiques d'un secteur pollué mais correspondent plutôt à un appauvrissement du peuplement néritique. Dans le secteur le plus pollué, la composition spécifique varie au cours du temps autour d'une communauté composée d'un groupe important avec Clausocalanus sp., Paracalanus sp., Oithona helgolandica et d'un groups de taxons moins fréquents représentés par des larves méroplanctoniques, Oithona nana et Evadne spinifera, tandis que les coryceidés, onceidés, Centropages typicus, chaetognathes et fritillaires se retrouvent plus fréquemment en eaux propres. La structure des populations est plus importante en période froide qu'en période chaude, période durant laquelle la communauté planctonique est fortement désorganisée. Paradoxalement Acartia clausi joue un rôle assez secondaire dans la définition structurelle de la communauté.Les fluctuations observées paraissent cependant peu liées à des paramètres reflétant l'intensité de la pollution. L'intégration du stress, en relation avec l'histoire antérieure de la communauté (intensité et durée du contact avec la nappe de dilution, potentialités trophiques) semblent ainsi prépondérantes.

Dispersal and mating features strongly influence the evolutionary dynamics and the spatial genetic structure (SGS) of marine populations. For the first time in a marine invertebrate, we examined individual reproductive success, by conducting larval paternity assignments after a natural spawning event, combined with a small‐scale SGS analysis within a population of the gorgonian Paramuricea clavata. Thirty four percent of the larvae were sired by male colonies surrounding the brooding female colonies, revealing that the bulk of the mating was accomplished by males from outside the studied area. Male success increased with male height and decreased with increasing male to female distance. The parentage analyses, with a strong level of self‐recruitment (25%), unveiled the occurrence of a complex family structure at a small spatial scale, consistent with the limited larval dispersal of this species. However, no evidence of small scale SGS was revealed despite this family structure. Furthermore, temporal genetic structure was not observed, which appears to be related to the rather large effective population size. The low level of inbreeding found suggests a pattern of random mating in this species, which disagrees with expectations that limited larval dispersal should lead to biparental inbreeding. Surface brooding and investment in sexual reproduction in P. clavata contribute to multiple paternity (on average 6.4 fathers were assigned per brood), which enhance genetic diversity of the brood. Several factors may have contributed to the lack of biparental inbreeding in our study such as (i) the lack of sperm limitation at a small scale, (ii) multiple paternity, and (iii) the large effective population size. Thus, our results indicate that limited larval dispersal and complex family structure do not necessarily lead to biparental inbreeding and SGS. In the framework of conservation purposes, our results suggested that colony size, proximity among colonies and the population size should be taken into consideration for restoration projects.  相似文献

The production of biopharmaceutical proteins in plants requires efficient downstream processing steps that remove impurities such as host cell proteins (HCPs) and adventitious endotoxins produced by bacteria during transient expression. We therefore strived to develop effective routines for endotoxin removal from plant extracts and the subsequent use of the extracts to generate antibodies detecting a broad set of HCPs. At first, we depleted the superabundant protein ribulose‐1,5‐bisphosphate carboxylase/oxygenase (RuBisCO) for which PEG precipitation achieved the best results, preventing a dominant immune reaction against this protein. We found that a mixture of sera from rabbits immunized with pre‐depleted or post‐depleted extracts detected more HCPs than the individual sera used alone. We also developed a powerful endotoxin removal procedure using Polymyxin B for extracts from wild type plants or a combination of fiber‐flow filtration and EndoTrap Blue for tobacco plants infiltrated with Agrobacterium tumefaciens. The antibodies we generated will be useful for quality and performance assessment in future process development and the methods we present can easily be transferred to other expression systems rendering them useful in the field of plant molecular farming.  相似文献

Five cheese-ripening yeasts (Geotrichum candidum, Saccharomyces cerevisiae, Kluyveromyces lactis, Yarrowia lipolytica and Debaryomyces hansenii) were compared with respect to their ability to generate volatile aroma compounds. K. lactis produced a variety of esters - ethylacetate (EA) being the major one - and relatively limited amounts of volatile sulphur compounds (VSCs). Conversely, G. candidum produced significant amounts of VSCs [with the thioester S-methyl thioacetate (MTA) being the most prevalent] and lower quantities of non-sulphur volatile compounds than K. lactis. We suspect that K. lactis is able to produce and/or accumulate acetyl CoA - a common precursor of MTA and EA - but that it produces limited amounts of methanethiol (MTL); both acetyl CoA and MTL are precursors for MTA synthesis. When supplemented with exogenous MTL, MTA production greatly increased in K. lactis cultures whereas it was unchanged in G. candidum cultures, suggesting that MTL is a limiting factor for MTA synthesis in K. lactis but not in G. candidum. Our results are discussed with respect to L-methionine catabolism.  相似文献

The effect of the carbon source and l-methionine on the ability of Geotrichum candidum to produce volatile sulphur compounds (VSC) was studied. This yeast was cultivated in a synthetic medium supplemented with various carbon sources and l-methionine at different concentrations. Both glycerol and glucose significantly increased VSC production by G. candidum. Unlike the effect on the l-methionine- and 4-methylthio-2-oxobutyric acid-demethiolating activities, the supply of a carbon source had a dramatic effect on the activity of aminotransferase, a key enzyme in l-methionine catabolism. An increase in the initial concentration of l-methionine resulted in a rise in the production of sulphur compounds (VSC, 4-methylthio-2-oxobutyric acid) but had limited effect on l-methionine-catabolising enzyme activities. Evidence for the existence of a dual effect of the carbon source and l-methionine on VSC biosynthesis was obtained in this study.  相似文献

Gut fluorescence, feeding and egg production rates of zooplankton assemblages were measured in a shallow, eutrophic brackish-water pond for 24 days. Brachionus plicatilis, Hexarthra intermedia and Apocyclops panamensis successively developed and exhibited differences in food selectivity. Rotifers selected small particles but also had a preference for larger particles (15–21 μm, Equivalent Spherical Diameter, ESD). B. plicatilis appeared less selective than H. intermedia, which fed mostly on particles <6 μm. A. panamensis adults showed a selectivity for 6–21 μm ESD particles. Laboratory experiments suggested that A. panamensis adults were able to shift from seston to carnivorous feeding, depending on the availability of these food resources. Measurements of gut fluorescence and grazing gave comparable ingestion rates. Rotifers displayed the highest ingestion rates (up to 486% body C d-1). Despite high total phytoplankton and seston biomasses, the high ingestion rates and selective grazing of rotifers induced auto food-limitation phenomena and caused major changes in seston abundance and size structure. Grazing impact was less important when A. panamensis dominated. This revised version was published online in July 2006 with corrections to the Cover Date.  相似文献

Seasonal variations of zooplankton communities and their interactions with phytoplankton and environmental parameters were studied over a twelve‐month period in Lake Guiers (Senegal, West Africa). Zooplankton was most abundant during the warm and rainy season corresponding to the flood event (July to October) with peaks for rotifers and cyclopoid copepods in July–August and September–October, respectively. Filamentous cyanobacteria were the dominant phytoplankton during the warm season (August to November), while diatoms were dominant during the cool season (December to April). Temperature increase and river flooding both played an important role in these successions, favoring intense development of cyanobacteria and cyclopoid copepods. Trophic interactions also seem to be an important factor in structuring of the plankton community. Overall, the plankton composition displayed a trend toward a eutrophication state. Our study confirms the existence of seasonality in zooplankton communities and the importance of using biological indicators such as phyto‐ and zooplankton to monitor water quality, in particular in tropical freshwater ecosystems. The size, composition and distribution of particles in the watercolumn were surveyed in a shallow area (1 m depth) of a tropicallagoon (Cte d'Ivoire) during a sequence of wind-induced resuspension.Water samples were collected hourly near the surface duringone tidal cycle. Three characteristic periods were distinguished:a calm period with low wind speed (average 1.2 m s–1 awindy period with wind speed >3 m–1 s (range between4 and 6 m s–1) inducing sediment resuspension and a relaxationperiod during the decrease of wind velocity. From the analysisof several parameters (particle size and volume, bacteria. pico-and nanophytoplankton, ciliates and detritus), sediment resuspensioncaused a regular injection of particles from the bed. The finestparticles (1.5–6 µm: chlorophytes such as Chiorellaspp., picocyanobacteria such as Synechococcus) were the firstto be affected by wind-induced turbulence, whereas large particles(6–12 µm: diatoms. cyanobacteria such as Lyngbiaspp.) were dispersed into the water column at the highest windspeed. The fate of the different seston components differedaccording to their size. Therefore, wind-induced resuspensioncould greatly influence the food web organization through thequantity, quality and size of edible particles available ata given time.  相似文献

Résumé La présente étude porte sur deux séries de données récoltées en baie de Biétri (lagune Ebrié, Côte d'Ivoire). La première concerne un programme d'échantillonnage à basse fréquence (octobre 1985–mars 1986), au cours duquel ont été mises en évidence les conditions hydroclimatiques nécessaires à la suppression dans ce milieu du gradient de densité (augmentation de la salinité de surface, refroidissement des eaux lagunaires consécutif à une remontée côtière). La seconde se rapporte à un échantillonnage à haute fréquence (janvier–février 1987), sur le même site, afin de définir les mécanismes et les conséquences écologiques de la déstratification. Lorsque les conditions hydroclimatiques sont réunies, la déstratification proprement dite se réalise à l'occasion d'une séquence de vive-eau, lors d'une basse basse-mer exceptionnelle, par l'accélération des déplacements d'eau de surface dans la baie, traduite par un nombre de Richardson minimal. Cette déstratification permet une homogénéisation des profils verticaux des concentrations en sels nutritifs, mais surtout, elle entraîne une disparition de 90% de la charge en N-NH4 et de 60% de celle en P-PO4. Une modification de l'environnement chimique des eaux de l'hypolimnion lors de la réoxygénation est suggérée pour expliquer cette brutale et transitoire disparition. A l'opposé, la répartition verticale des biomasses semble peu affectée par la suppression du gradient de densité, la topographie des fonds interdisant l'établissement d'importants mouvements convectifs. Ainsi, les conséquences biologiques de la déstratification dans ce milieu estuarien eutrophe sont peu marquées, et sans comparaison avec les modifications hydrodynamiques et hydrochimiques observées à cette occasion.
Two sets of data were collected in the Biétri bay (Ebrié lagoon, Côte d'Ivoire). The first one concerns a program with a low frequency sampling (October 1985–March 1986), during which hydroclimatic conditions necessary to the suppression of the density gradient (increasing surface salinity, cooling of surface waters in relation with a coastal upwelling) were defined. The second one refers to a high frequency program (January–February 1987), in the same area, in order to describe the mechanisms and the ecological consequences of the destratification. When hydroclimatic conditions are favorable, destratification occurs during a spring tide period, corresponding to an exceptional low low waters, induced by a higher tidal current velocity in the bay, evidenced by a low Richardson number. That destratification allows an homogenization of vertical profiles of nutrient concentrations, but leads mainly to the disappearance of 90% of N-NH4 and 60% of the P-PO4 loads. A modification of the chemical environment in the hypolimnion during its re-oxygenation is proposed to explain this sudden and transitory disappearance. On the contrary, the vertical distribution of biomass seems to be little affected by the gradient suppression, the bottom topography preventing from important convective movements. Thus, the biological consequences of the destratification in this eutrophic estuarine area are not as important as the hydrodynamical and hydrochemical modifications observed during this phenomenon.

Calpains are Ca(2+)-dependent cysteine proteases known to be important for the regulation of cell functions and which aberrant activation causes cell death in a number of degenerative disorders. To provide a tool for monitoring the status of calpain activity in vivo under physiological and pathological conditions, we created a mouse model that expresses ubiquitously a fluorescent reporter consisting of eCFP and eYFP separated by a linker cleavable by the ubiquitous calpains. We named this mouse CAFI for calpain activity monitored by FRET imaging. Our validation studies demonstrated that the level of calpain activity correlates with a decrease in FRET (fluorescence resonance energy transfer) between the two fluorescent proteins. Using this model, we observed a small level of activity after denervation and fasting, a high level of activity during muscle regeneration and ischemia, and local activity in damaged myofibers after exercise. Finally, we crossed the CAFI mouse with the alpha-sarcoglycan-deficient model, demonstrating an increase of calpain activity at the steady state. Altogether, our results present evidence that CAFI mice could be a valuable tool in which to follow calpain activity at physiological levels and in disease states.  相似文献
