Ovaries were surgically removed from female goats (Toggenburg, Nubian and Saanen breeds). Oocytes were collected by follicular aspiration or after ovaries were minced, then matured in mTCM-199 with 100 mug LH + 0.5 mug FSH + 1.0 mug estradiol 17-beta/ml for 27 h prior to in vitro fertilization (17). Although more oocytes were made available by mincing than by aspiration, higher proportions of aspirated oocytes were fertilized and developed to morulae. Proportions that fertilized and reached morulae were 82 102 (80.4%) and 50 102 (49.0%) versus 77 126 (61.1%) and 27 126 (21.4%) for oocytes obtained by aspiration and after ovarian mincing, respectively (P<0.05). Proportions of inseminated ova undergoing cleavage and continuing development to the morula stage differed significantly (P<0.05) among 5 co-culture treatment groups, with higher proportions of cleavage (23 27 , 85.2%) and morulae (14 27 , 51.9%) obtained by co-culture on caprine cumulus cells (cCC). Some oocytes reached the blastocyst stage (4 54 , 7.4%) following oocyte collection by aspiration and culture on caprine oviduct epithelial cells (cOEC). After 4- and 8-cell stage embryos obtained by aspiration and culture on cCC were transferred pregnancy resulted. Twin male kids (developed from different embryos) were born on August 6, 1993, and have developed into normal bucks. Conditions reported here provided an adequate environment for support of oocyte maturation, fertilization and early embryonic development in vitro (IVMFC) with normal development after embryo transfer. 相似文献

本文报道海拔3417m和4280m地区世居藏族和移居汉族青少年运动状态下心肺功能的对比研究。结果显示:3417m和4280m世居藏族的最大氧耗量、无氧阈值及最大心输出量都明显大于汉族,血氧饱和度(Sao2)随运动负荷的增加而降低。海拔3417m藏、汉族的△Sao2分别为7.46%和10.03%,4280m处为8.57%和13.75%,最大心率随海拔升高而下降。研究提示,藏族青少年有较高的最大有氧能力,反映了他们对低氧环境的适应优势。 相似文献

Abstract Filamentous hemagglutinin (FHA), a 220-kDa protein located on the surface of Bordetella pertussis , is one of the major cell adhesins of this bacterium. We have produced three hybridoma cell lines that express monoclonal antibodies (mAbs) against FHA: X3C, X3E and X4B. The anti-FHA mAbs X3C and X3E reacted with 220-kDa FHA protein bands on Western blots. The mAb X4B, which reacted with FHA in ELISA, did not bind to FHA in a Western blot assay. All three mAbs seemed to be directed to the same epitope or to epitopes in close proximity as suggested by competition ELISAs. All three mAbs were able to inhibit the adherence of Chinese hamster ovary cells to purified FHA, and they could also inhibit the FHA-mediated agglutination of goose red blood cells. The attachment of B. pertussis to epithelial cell monolayers was inhibited by the mAb X3C. These antibodies are very useful probes to identify the presence of FHA in bordetellae species and in clinical reagents such as pertussis vaccines, and to characterize the functional domains of this important bacterial adhesin. 相似文献

Prostaglandin E1 receptor sites were measured in homogenates of NG108-15 neuroblastoma-glioma hybrid cells after exposure of intact cells to PGE1. Scatchard analysis of competitive binding studies showed that incubation of NG108-15 cells in the presence of 2.5 microM PGE1 for 16 h resulted in a loss of PGE1 receptors and an increase in the dissociation constant of the remaining receptors. Thus, cells challenged with PGE1 not only lose adenylate cyclase activity, but also lose PGE1 receptors and decreased the affinity of the remaining receptors for PGE1. 相似文献

Background
Many musculoskeltal injuries in the workplace have been attributed to the repetitive loading of muscle and soft tissues. It is not disputed that muscular fatigue is a risk factor for musculoskeltal injury, however the disparity between gender with respect to muscular fatigability and rate of recovery is not well understood. Current health and safety guidelines do not account for sex differences in fatiguability and may be predisposing one gender to greater risk. The purpose of this study was to quantify the sex differences in fatigue development and recovery rate of lower and upper body musculature after repeated bouts of sustained isometric contractions.Methods
Twenty-seven healthy males (n = 12) and females (n = 15) underwent bilateral localized fatigue of either the knee extensors (male: n = 8; female: n = 8), elbow flexors (male: n = 8; female: n = 10), or both muscle groups. The fatigue protocol consisted of ten 30-second sub-maximal isometric contractions. The changes in maximum voluntary contraction (MVC), electrically evoked twitches, and motor unit activation (MUA) were assessed along with the ability to control the sustained contractions (SLP) during the fatigue protocol using a mixed four-factor repeated measures ANOVA (gender × side × muscle × time) design with significance set at p < 0.05.Results
There was a significant loss of MVC, MUA, and evoked twitch amplitude from pre- to post-fatigue in both the arms and legs. Males had greater relative loss of isometric force, a higher rate of fatigue development, and were less capable of maintaining the fatiguing contractions in the legs when compared to the females.Conclusion
The nature of the induced fatigue was a combination of central and peripheral fatigue that did not fully recover over a 45-minute period. The results appear to reflect sex differences that are peripheral, and partially support the muscle mass hypothesis for explaining differences in muscular fatigue.9.
The long-term goal of this project was to find alternative uses for bottom ash (BA) and composted dairy manure (CM), by-products of coal combustion and livestock production, respectively. The study discussed in this paper focused on potential water quality impacts associated with using blended BA and CM as a soil amendment. The constituents of BA and CM include heavy metals and other chemicals that, while essential nutrients for plant growth, also pose a potential threat to water quality. Four blends (BA:CM, v/v) namely, B1 (100%:0%), B2 (70%:30%), B3 (50%:50%) and B4 (0%:100%), were subjected to flow-through water table management and two blends, B2 (70%:30%) and B3 (50%:50%), were subjected to constant head water table management using de-ionized water. Leachate and standing water from saturated and flooded blends of BA and CM were examined for total solids (TS), volatile solids (VS), COD, pH, total Kjeldahl nitrogen (TKN), NO(3)-N, total P, total K as well as selected metals over a 5 and 7 week period for flow-through and constant head watertables, respectively. The results showed that higher CM content resulted in higher TS, VS, TKN, P and K concentrations in the leachate and standing water. Concentrations of these constituents were higher in leachate than in the standing water. Even though, marked reductions of most chemicals in the leachate and standing water were realized within one to three weeks, initially high concentrations of chemicals in leachate and standing water from these particular blends made them unsuitable as soil amendment material. Based upon these results, it was concluded that additional column studies of BA and CM blends with reduced CM content (5%, 10% and 20%) should be performed to further assess the feasibility of BA and CM blends as an environmentally safe soil amendment material. 相似文献

Relationships in the Drosophila obscura species group, inferred from mitochondrial cytochrome oxidase II sequences 总被引:2,自引:0,他引:2
We compare the sequences for the mitochondrial cytochrome oxidase II gene
of 13 species of the Drosophila obscura group. The survey includes six
members of the D. affinis subgroup, four of the D. pseudoobscura subgroup,
and three of the D. obscura subgroup. In all species, the gene is 688
nucleotides in length, encoding a protein of 229 amino acids plus the first
position T of the stop codon. The sequences show the typical
high-transition bias for closely related species, but that bias is
essentially eliminated for species pairs of > 5% sequence divergence.
The phylogenetic relationships in the species group are inferred using both
neighbor-joining and maximum parsimony. The two procedures give comparable
results, showing that the D. affinis and D. pseudoobscura subgroups are
monophyletic groupings that appear to have closer affinities to one another
than either has to the D. obscura subgroup. We use transversion distances
to estimate times of divergence, on the basis of three different estimates
of the time of separation of the D. obscura species group from the D.
melanogaster group. If that event occurred 35 Mya, then we can estimate the
origin of the nearctic forms at approximately 22 Mya and the separation of
the D. affinis and D. pseudoobscura subgroups at approximately 17 Mya.
