Pycnogenol® (PYC), a patented combination of bioflavonoids extracted from the bark of French maritime pine (Pinus maritima), scavenges free radicals and promotes cellular health. The protective capacity of PYC against ethanol toxicity of neurons has not previously been explored. The present study demonstrates that in postnatal day 9 (P9) rat cerebellar granule cells the antioxidants vitamin E (VE) and PYC (1) dose dependently block cell death following 400, 800, and 1600 mg/dL ethanol exposure (2) inhibit the ethanol‐induced activation of caspase‐3 in the same model system; and (3) reduce neuronal membrane disruption as assayed by phosphatidylserine translocation to the cell surface. These results suggest that both PYC and VE have the potential to act as therapeutic agents, antagonizing the induction of neuronal cell death by ethanol exposure. © 2004 Wiley Periodicals, Inc. The development of the highly sensitive cytochemical bioassay for ACTH has permitted the measurement of plasma ACTH levels during the insulin hypoglycaemia test (I.H.T.) in patients treated with corticosteroids and corticotrophin. The ACTH, corticosteroid, and growth hormone (GH) responses in the I.H.T. were measured in three groups of 12 rheumatoid arthritis patients. One group was receiving long-term corticotrophin treatment, the second was undergoing long-term corticosteroid treatment, and the third had never received systemic hormone therapy. The increments in plasma ACTH, corticosteroids, and GH were diminished in the corticosteroid-treated group, as were increments in plasma GH and ACTH in the corticotrophin-treated group; but in this group the corticosteroid increment was normal. Examination of the area under the curve of the ACTH response showed that the total amount of ACTH secreted was normal though the rate of secretion was reduced. Catalytically defective rare variants of Sialic acid Acetyl Esterase (SIAE) have previously been linked to autoimmunity. Studies presented here confirm that the M89V SIAE protein and all other products of common variant alleles of SIAE are catalytically normal. Although overexpressing transfected non-lymphoid cells secrete small amounts of SIAE that can associate with the cell surface, normal human lymphocytes do not exhibit cell surface SIAE, supporting genetic evidence in mice that indicates that this protein functions in a lymphocyte intrinsic manner. Analyses of the plasma proteome also indicate that SIAE is not secreted in vivo. A re-analysis exclusively of catalytically defective rare variant alleles of SIAE in subjects in which this gene was completely sequenced confirmed an association of SIAE with autoimmunity. A subset of catalytically defective rare variant SIAE alleles has previously been typed in a large genotyping study comparing a diverse group of disease subjects and controls; our re-analysis of this data shows that catalytically defective alleles are enriched in disease subjects. These data suggest that SIAE may be associated with autoimmunity and that further study of catalytically defective rare variant SIAE alleles in terms of autoimmune disease susceptibility is strongly warranted. 相似文献

Antimycotic Compounds from the Plant Pathogen Rhizoctonia solani and its Antagonist Trichoderma harzianum 总被引:5,自引:0,他引:5
The soilborne rhizosphere-competent fungal biocontrol agent Trichoderma harzianum isolate Th008 secreted trichodermin (MW = 292) and a small peptide (MW = 876) in culture. These compounds were antagonistic in culture to the mycelial growth of the soilborne fungal pathogen Rhizoctonia solani isolate 2B-12, which is highly virulent to soybean ( Glycine max )seedlings. When 100mg of dried autoclaved mycelial mat of R. solani was added to 200 ml liquid cultures of T. harzianum , the quantity of antimycotic compounds secreted by the latter was 3.5 times greater than that of the antagonist alone. R. solani secreted a coumarin derivative (MW = 313) in liquid culture, which inhibited the mycelial growth of T. harzianum ; however, inhibition of the growth of the antagonist required a greater concentration than that for the antimycotic compounds produced by the antagonist against the pathogen. The inclusion of 100 mg of dried autoclaved mycelial mat of T. harzianum in a 200 ml liquid culture of R. solani did not affect the quantity of the antimycotic compound produced by the pathogen. 相似文献

The conjugative transposon Tn919, originally isolated in Streptococcus sanguis FC1, is capable of low-frequency transfer (10−7 and 10−8 per recipient) on membrane filters to a wide number of streptococcal recipients including the industrially important lactic streptococci. The introduction of pMG600 (Lac+ Lax−; a lactose plasmid capable of conjugative transfer at high frequencies and which, in certain hosts, confers an unusual clumping phenotype) into a Streptococcus lactis CH919 donor, generating S. lactis CH001, resulted in a significant improvement in the transfer frequency of Tn919 to S. lactis CK50 (1.25 × 10−4 per recipient). In addition, these matings could be performed on agar surfaces, allowing the recovery of a greater number of recipients than with filter matings. Tn919 also transferred at high frequency to S. lactis subsp. diacetylactis 18-16S but not to Streptococcus cremoris strains. Insertion in 18-16S transconjugants generated from filter matings with an S. lactis CH919 donor was random, occurring at different sites on the chromosome and also in plasmid DNA. Thus, the conditions necessary for the practical exploitation of Tn919 in the targeting and cloning of genes from a member of the lactic streptococci, namely, high-frequency delivery and random insertion in host DNA, were achieved. 相似文献

The cardiotonic activities of pumiliotoxins, pyrethroids and sodium and calcium channel activators were assessed in vitro with spontaneously beating guinea pig atria. The ability of these compounds to stimulate phosphoinositide turnover was assessed in guinea pig cerebral cortical synaptoneurosomes. The activity of pumiliotoxins for both cardiotonic activity and phosphoinositide breakdown was strongly dependent on the structure and configuration of the side chain and there was a correlation between structure and activity in the two systems. Pyrethroids that had cardiotonic activity also induced phosphoinositide breakdown. Other sodium channel and calcium channel activators that induced phosphoinositide breakdown were also cardiotonic. It is suggested that phosphoinositide breakdown leading to inositol phosphates and diacylglycerols may represent a mechanism underlying the cardiotonic effects of certain agents. A phorbol ester, phorbol 12-myristate 13-acetate, that mimics the activation of protein kinase C elicited by diacylglycerols, had cardiotonic activity. 相似文献
