The lipid-peroxidating enzyme, 15-lipoxygenase (LO)-1 and its metabolite, 13-S-hydroxyoctadecadienoic acid (13-S-HODE), likely contribute to prostate tumorigenesis. Thus, this study evaluated adenovirus-mediated overexpression of 15-LO-1 on normal mouse prostate. Adenovirus expressing either human 15-LO-1 tagged with green fluorescent protein (GFP) or GFP alone was orthotopically injected into the dorsolateral prostates of C57BL/6 mice, three times over the course of 60 days. On day 90, pathological changes in prostate tissue were assessed by hematoxylin and eosin (H&E) staining. Expression of the proliferation marker Ki-67 was evaluated by immunohistochemistry and expression of angiogenesis markers were analyzed by an antibody array. Based on the latter study, immunoprecipitation analysis was used to measure the effect of 13-S-HODE, with or without conditioned media, on fibroblast growth factor-a and b (FGF-a and FGF-b) expression in human PrEC (normal prostate epithelial), PrSMC (normal prostate smooth muscle) and PrSC (normal prostate stromal) lines. Expression of viral 15-LO-1-GFP, but not GFP alone, resulted in the development of a prostate intraepithelial neoplasia (PIN)-like phenotype with increased expression of Ki-67. Aberrant 15-LO-1 expression also induced the angiogenic markers FGF-a and FGF-b. Human PrEC, PrSMC and PrSC lines demonstrated an increase in FGF-b expression upon stimulation with 13-S-HODE, which was further increased by the addition of conditioned media from the epithelial or smooth muscle cells. Using adenoviral mediated 15-LO-1 gene delivery, this study suggests that aberrant 15-LO-1 overexpression in normal prostate can trigger events leading to prostate epithelial and stromal cell proliferation. Thus, our findings demonstrate the effectiveness of this viral system for 15-LO-1 expression studies in tissues.  相似文献

1. As parasites can dramatically reduce the fitness of their hosts, there should be strong selection for hosts to evolve and maintain defence mechanisms against their parasites. One way in which hosts may protect themselves against parasitism is through altered behaviours, but such defences have been much less studied than other forms of parasite resistance. 2. We studied whether monarch butterflies (Danaus plexippus L.) use altered behaviours to protect themselves and their offspring against the protozoan parasite Ophryocystis elektroscirrha (McLaughlin & Myers (1970), Journal of Protozoology, 17, p. 300). In particular, we studied whether (i) monarch larvae can avoid contact with infectious parasite spores; (ii) infected larvae preferentially consume therapeutic food plants when given a choice or increase the intake of such plants in the absence of choice; and (iii) infected female butterflies preferentially lay their eggs on medicinal plants that make their offspring less sick. 3. We found that monarch larvae were unable to avoid infectious parasite spores. Larvae were also not able to preferentially feed on therapeutic food plants or increase the ingestion of such plants. However, infected female butterflies preferentially laid their eggs on food plants that reduce parasite growth in their offspring. 4. Our results suggest that animals may use altered behaviours as a protection against parasites and that such behaviours may be limited to a single stage in the host-parasite life cycle. Our results also suggest that animals may use altered behaviours to protect their offspring instead of themselves. Thus, our study indicates that an inclusive fitness approach should be adopted to study behavioural defences against parasites.  相似文献

Studies with whole cells and mitochondrial fractions revealed increased respiratory activity inAspergillus repens grown under salt stress conditions. The state 3 and state 4 respiration rates, PO ratios, and Mg2+-dependent ATPase were higher in mitochondria of stressed cells than in control cells.A. repens respires via an antimycin A-and cyanide-sensitive pathway. Oligomycin, dicyclohexylcarbodiimide (DCCD) and rotenone inhibited respiration rates less in mitochondria of stressed cells than in controls. Though 2,4-dinitrophenol (DNP), carbonyl cyanide-m-chlorophenylhydrazone (m-Cl-CCP), and carbonyl cyanide-p-trifluoromethylhydrazone (p-F3-CCP) did not stimulate Mg2+-ATPase activity, DNP enhanced the respiration rates, whereasm-Cl-CCP andp-F3-CCP decreased the respiration rates in either condition; mitochondria of stressed cells exhibited a lower degree of inhibition than controls. Addition of DNP, oligomycin, and DCCD inhibited the basal Mg2+-ATPase (ATPase activity without Mg2+ addition). Oligomycin inhibited the Mg2+-ATPase. DCCD showed less inhibition in mitochondria under stress than did the controls. Levels of some respiratory enzymes were higher in the culture grown under stress than in the controls.  相似文献

In amoebae of the cellular slime molds (mycetozoans) Acrasis rosea and Protostelium mycophaga, bundles of F-actin radiate from the endoplasm-ectoplasm interface into the pseudopodia, where G-actin is also located. We conclude that these actin bundles form a core scaffold driving pseudopod extension which is subsequently completed by filling with a more loosely organized meshwork of F-actin. Some bipolar, elongate amoebae of A. rosea also contained long bundles of F-actin that traverse the cells lengthwise and remotely resemble stress fibers. Rodlets of F-actin were scattered in the body of amoebae of A. rosea or formed star-shaped or polygonal complexes near or around contractile vacuoles, where they may play a role in contraction. In total protein extracts analyzed by SDS-PAGE and immunoblots the actins migrated like the rabbit skeletal muscle control. The relative proportion of actin in total protein extracts was 7.9% for A. rosea and 34.5% for P. mycophaga. We detected four or five isoactins in extracts of both species and we determined that the genome of each species contains approximately six actin genes. Whether they are all expressed or if posttranslational modifications occur remains to be determined. Myosin II was enriched in actomyosin extracts; its Mr was 187.8 kDa for A. rosea and 220.7 kDa for P. mycophaga. Cell models ("ghosts") contracted upon the addition of ATP. We conclude that amoebae of A. rosea and P. mycophaga, although behaving differently from those of Dictyostelium discoideum, contain the basic repertoire of molecules that enable pseudopod extension by actin polymerization and ATP-induced contraction of the cell cortex. Aspergillus repens, a salt-pan isolate, was halotolerant. When grown for 72 h (log phase) and 144 h (beginning of stationary phase) in a medium containing 2m sodium chloride, the activities of invertase, malate dehydrogenase (MDH), glucose-6-phosphate dehydrogenase (G6PDH), and glutamate dehydrogenase (GDH) were found to have increased. Control cultures grown in a medium devoid of 2m NaCl failed to show such changes. The activities of MDH, G6PDH, and GDH increased with rising concentrations of Na+ (as NaCl) when added up to 100mm in vitro. At higher concentrations they decreased. Changes in kinetic constants, Km and Vmax of these enzymes, as well as their de novo synthesis, were found to be some of the responses to NaCl stress-mediated changes.  相似文献

Na+, K+ and the ratio of Na+/K+ were higher in cells of the halotolerant Aspergillus repens grown with 2 M NaCl than without NaCl. The osmolytes, proline, glycerol, betaine and glutamate, did not affect the Na+/K+ ratio, nor the polyol content of cells under any conditions. The concentrations of polyols, consisting of glycerol, arabitol, erythritol and mannitol, changed markedly during growth, indicating that they have a crucial role in osmotic adaptation.  相似文献

Docosapentaenoic acid (DPA) is a unique fatty acid that exists in two isomeric forms (n-3 and n-6), which differ in their physiological behaviors. DPA can undergo free radical-mediated peroxidation via lipoxygenase (LOX). 15-LOX, one of the LOX isomers, has received much attention in cancer research because of its very different expression level in normal tissues compared to tumors and some bioactive fatty acid metabolites modulating the tumorigenic pathways in cancer. However, the mechanism linking 15-LOX, DPA metabolites, and their bioactivities is still unclear, and the free radicals generated in DPA peroxidation have never been characterized. In this study, we have studied radicals formed from both soybean and human cellular (PC3-15LOS cells) 15-LOX-catalyzed peroxidation of DPAs at various pH's using a combination of LC/ESR/MS with the spin trapping technique. We observed a total of three carbon-centered radicals formed in 15-LOX-DPA (n-3) stemming from its 7-, 17-, and 20-hydroperoxides, whereas only one formed from 17-hydroperoxide in DPA (n-6). A change in the reaction pH from 8.5 (15-LOX enzyme optimum) to 7.4 (physiological) and to 6.5 (tumor, acidic) not only decreased the total radical formation but also altered the preferred site of oxygenation. This pH-dependent alteration of radical formation and oxygenation pattern may have significant implications and provide a basis for our ongoing investigations of LOXs as well as fatty acids in cancer biology.  相似文献
