We investigated the effects of chemical variation in thyme (Thymus vulgaris L.) on its interactions with competitors, herbivores, and herbivore predators. Four different thyme monoterpene phenotypes (chemotypes) were grown in a 4x2x2 factorial of chemotype, caging (sham half-cages vs. full cages), and competition (control vs. the grass Bromus madritensis L.). Cages reduced numbers of arthropod predators. Thyme-feeding aphids Aphis serpylli Koch passed through full cage walls to increase more than fourfold. As a result, freed from their predators, aphids had a large negative effect on thyme size and flowering. Similarly, competition from Bromus had a negative effect on thyme size and flowering. Individual effects of aphids and competition were nonadditive, however, and their combined effect was significantly less than that predicted by a multiplicative null model. Differential thyme sizes among chemotypes were not mediated by herbivores or competitors, but differential flowering was due to the effects of chemotype on aphids. We thus document differential selection by aphids among thyme chemotypes and the influence of Bromus on the strength of these negative effects of aphids.  相似文献

Aims Woody plant-browser systems represent an understudied facet of herbivory. We subjected four genotypes of trembling aspen to artificial browsing, similar to that of a large mammalian herbivore, and applied deer saliva to clipped and unclipped trees to assess: (i) the effects of artificial browsing on aspen growth and phytochemistry of leaves and stems, (ii) genotypic variation in responses and (iii) potential alterations of responses by mammalian saliva.Methods Potted aspen trees were grown outdoors on the University of Wisconsin-Madison campus. The experiment consisted of a fully-crossed, 2 × 2 × 4 randomized complete block design, with two levels of artificial browsing (unclipped and clipped), two levels of saliva application (no saliva and saliva) and four aspen genotypes. To simulate ungulate browsing damage, we removed the upper 50% of the stem of half of the trees by pinching the stem with needle-nosed pliers and then separating it by tearing. For half of the damaged trees, we immediately swabbed the wound with deer saliva. Trees in the unclipped plus saliva treatment were swabbed with saliva at the 50% height mark. To assess the effects of clipping and saliva application, we harvested all trees after 2 months and measured various growth and chemical properties. Growth measurements included height, vertical growth, mass of leaves, stems and roots, leaf number and area and bud set. Chemical parameters included defensive, nutritional and structural components of both foliage and stems.Important findings Clipping affected most of the growth parameters measured, decreasing tree height, leaf, stem, root and total tree mass and leaf area. Clipped trees had greater vertical growth, more leaves and higher specific leaf area (SLA) than unclipped trees. Deer saliva had little to no effect on plant growth response to the clipping treatment. Terminal budset was delayed by clipping and varied among genotypes but not in response to saliva application. Clipping also affected most of the phytochemical variables measured, reducing defensive compounds (phenolic glycosides and condensed tannins (CTs)) and nutrients (N), but increasing structural components (cellulose and lignin) in both leaves and stems. Saliva had very little effect on tree chemistry, causing only a slight decrease in the amount of CTs in leaves. In general, leaves contained more defensive compounds and nutrients, but much less cellulose, compared with stems. Genotypes differed for all physical and chemical indices, and in tolerance to damage as measured by vertical growth. In addition, for most of the physical and chemical variables measured, genotype interacted with the clipping treatment, suggesting that in natural stands some genotypes will resist or tolerate browsing better than others, affecting forest genetic composition and ultimately forest dynamics.  相似文献

Populus deltoides is an important industrial tree species widely planted in many areas of the world, and de novo genome sequencing of this plant has been carried out by several research groups. A dense genetic map associating genome sequences is highly desirable for reconstructing the chromosome pseudomolecules using the obtained sequence scaffold assemblies. For this purpose, an intraspecific full-sib F1 mapping pedigree was established by using the sequenced P. deltoides as the maternal parent. With this mapping pedigree, we constructed a high-density genetic map using 92 randomly selected progenies. Single nucleotide polymorphism (SNP) markers were discovered by using specific length amplified fragment sequencing (SLAF-seq). In total, 487,038 SLAFs were generated, of which 179,360 were polymorphic. A high-density genetic map was built using HighMap software, which included 11,680 Mendelian segregation markers distributing in 2851 marker bins. The established map consisted of 19 linkage groups (LGs) that equaled to the 19 haploid chromosomes in poplar genome, and spanned a total genetic length of 3494.66 cM, with an average distance of 1.23 cM per marker bin. The map presented here will be useful for anchoring the genome sequence assemblies along chromosomes, and for many other aspects of genetic studies on P. deltoides and the closely related species.  相似文献

Dioecy, the presence of separate sexes on distinct individuals, has evolved repeatedly in multiple plant lineages. However, the specific mechanisms by which sex systems evolve and their commonalities among plant species remain poorly understood. With both XY and ZW sex systems, the family Salicaceae provides a system to uncover the evolutionary forces driving sex chromosome turnovers. In this study, we performed a genome-wide association study to characterize sex determination in two Populus species, P. euphratica and P. alba. Our results reveal an XY system of sex determination on chromosome 14 of P. euphratica, and a ZW system on chromosome 19 of P. alba. We further assembled the corresponding sex-determination regions, and found that their sex chromosome turnovers may be driven by the repeated translocations of a Helitron-like transposon. During the translocation, this factor may have captured partial or intact sequences that are orthologous to a type-A cytokinin response regulator gene. Based on results from this and other recently published studies, we hypothesize that this gene may act as a master regulator of sex determination for the entire family. We propose a general model to explain how the XY and ZW sex systems in this family can be determined by the same RR gene. Our study provides new insights into the diversification of incipient sex chromosomes in flowering plants by showing how transposition and rearrangement of a single gene can control sex in both XY and ZW systems.  相似文献

Salicinoids are well-known defense compounds in salicaceous trees and careful screening at the population level is warranted to fully understand their diversity and function. European aspen, Populus tremula, is a foundation species in Eurasia and highly polymorphic in Sweden. We exhaustively surveyed 102 replicated genotypes from the Swedish Aspen collection (SwAsp) for foliar salicinoids using UHPLC-ESI-TOF/MS and identified nine novel compounds, bringing the total to 19 for this species. Salicinoid structure followed a modular architecture of a salicin skeleton with added side groups, alone or in combination. Two main moieties, 2′-cinnamoyl and 2′-acetyl, grouped the SwAsp population into four distinct chemotypes, and the relative allocation of salicinoids was remarkably constant between different environments, implying a highly channeled biosynthesis of these compounds. Slightly more than half of the SwAsp genotypes belonged to the cinnamoyl chemotype. A fraction synthesized the acetyl moiety alone (∼7%) or in combination with cinnamoyl (∼2%), and close to forty percent lacked either of the two characteristic moieties, and thus resemble P. tremuloides in their salicinoid profile. The two most abundant chemotypes were evenly distributed throughout Sweden, unlike geographical patterns reported for SwAsp phenology traits, plant defense genes, and herbivore community associations. Here we present the salicinoid characterization of the SwAsp collection as a resource for future studies of aspen chemical ecology, salicinoid biosynthesis, and genetics.  相似文献

The development of non-recombining sex chromosomes has radical effects on the evolution of discrete sexes and sexual dimorphism. Although dioecy is rare in plants, sex chromosomes have evolved repeatedly throughout the diversification of angiosperms, and many of these sex chromosomes are relatively young compared to those found in vertebrates. In this study, we designed and used a sequence capture array to identify a novel sex-linked region (SLR) in Salix nigra, a basal species in the willow clade, and demonstrated that this species has XY heterogamety. We did not detect any genetic overlap with the previously characterized ZW SLRs in willows, which map to a different chromosome. The S. nigra SLR is characterized by strong recombination suppression across a 2 MB region and an excess of low-frequency alleles, resulting in a low Tajima’s D compared to the remainder of the genome. We speculate that either a recent bottleneck in population size or factors related to positive or background selection generated this differential pattern of Tajima’s D on the X and autosomes. This discovery provides insights into factors that may influence the evolution of sex chromosomes in plants and contributes to a large number of recent observations that underscore their dynamic nature.Subject terms: Plant evolution, Quantitative trait  相似文献
