The potential clinical utility of genetic markers associated with response to fluoropyrimidine treatment in colorectal cancer patients remains controversial despite extensive study. Our aim was to test the clinical validity of both novel and previously identified markers of adverse events in a broad clinical setting. We have conducted an observational pharmacogenetic study of early adverse events in a cohort study of 254 colorectal cancer patients treated with 5-fluorouracil or capecitabine. Sixteen variants of nine key folate (pharmacodynamic) and drug metabolising (pharmacokinetic) enzymes have been analysed as individual markers and/or signatures of markers. We found a significant association between TYMP S471L (rs11479) and early dose modifications and/or severe adverse events (adjusted OR = 2.02 [1.03; 4.00], p = 0.042, adjusted OR = 2.70 [1.23; 5.92], p = 0.01 respectively). There was also a significant association between these phenotypes and a signature of DPYD mutations (Adjusted OR = 3.96 [1.17; 13.33], p = 0.03, adjusted OR = 6.76 [1.99; 22.96], p = 0.002 respectively). We did not identify any significant associations between the individual candidate pharmacodynamic markers and toxicity. If a predictive test for early adverse events analysed the TYMP and DPYD variants as a signature, the sensitivity would be 45.5 %, with a positive predictive value of just 33.9 % and thus poor clinical validity. Most studies to date have been under-powered to consider multiple pharmacokinetic and pharmacodynamic variants simultaneously but this and similar individualised data sets could be pooled in meta-analyses to resolve uncertainties about the potential clinical utility of these markers.  相似文献

Unlike pathogenic fungi, the budding yeast Saccharomyces cerevisiae is not efficient at using heme as a nutritional source of iron. Here we report that for this yeast, heme uptake is induced under conditions of heme starvation. Heme synthesis requires oxygen, and yeast grown anaerobically exhibited an increased uptake of hemin. Similarly, a strain lacking aminolevulinate synthase exhibited a sixfold increase in hemin uptake when grown without 2-aminolevulinic acid. We used microarray analysis of cells grown under reduced oxygen tension or reduced intracellular heme conditions to identify candidate genes involved in heme uptake. Surprisingly, overexpression of PUG1 (protoporphyrin uptake gene 1) resulted in reduced utilization of exogenous heme by a heme-deficient strain and, conversely, increased the utilization of protoporphyrin IX. Pug1p was localized to the plasma membrane by indirect immunofluorescence and subcellular fractionation. Strains overexpressing PUG1 exhibited decreased accumulation of [(55)Fe]hemin but increased accumulation of protoporphyrin IX compared to the wild-type strain. To measure the effect of PUG1 overexpression on intracellular heme pools, we used a CYC1-lacZ reporter, which is activated in the presence of heme, and we monitored the activity of a heme-containing metalloreductase, Fre1p, expressed from a constitutive promoter. The data from these experiments were consistent with a role for Pug1p in inducible protoporphyrin IX influx and heme efflux.  相似文献

Hemoglobin DDavis (Hb DD), an autosomal codominant in chickens, the αD-globin chain of Hb M of primitive cells and Hb D of definitive erythrocytes. Erythropoiesis and Hb synthesis was investigated in normal, heterozygous, and homozygous Hb DD mutant embryos (stages 15–44) and adults. The time of appearance, morphology, relationships to developmental changes, and number of primitive and definitive cells were determined. Primitive hemoglobins between stages 17 and 44 showed four components, P1, P2, E, and M (or MD), on high-resolution isoelectric focusing gels. Comparison of P1P2 ratios in the four phenotypes indicated that homozygous Hb DD embryos had an increased proportion of Hb P2 relative to Hb P1 between stages 17 and 35. This difference coincided with an increase in the number of large primitive cells. In all phenotypes the proportions of primitive hemoglobins decreased after stage 25 and they were not detected after stage 40. Basophilic definitive erythroblasts were present in cell suspensions from all phenotypes between stages 24 and 25. Hb A, the major Hb and Hb D, the minor Hb, of definitive cells of embryos and adults were detected by isoelectric focusing of lysates by stage 29. Definitive cells from late embryos of all phenotypes had higher proportions of Hb D (or Hb DD) than did red cells from corresponding adult birds. Heterozygous Hb DD embroys and adults had both Hb D and Hb DD. Hb DD comprises about 30% of the total minor Hb rather than 50% expected for heterozygosity at a single locus. In this respect heterozygous Hb DD chick embryos and adult birds are similar to certain heterozygous α-chain variants in humans. A minor Hb, H, found in lysates of later embryos disappears in lysates of normal chicks 65 days after hatching, but was present in the circulation of homozygous Hb DD chicks until at least 195 days after hatching. Additionally, several minor Hb components which may be asymmetrical hybrids or derived precursors of Hb A and Hb D (or Hb DD) were observed. This study provides the precise developmental stages when the switchover of erythroid cell populations and hemoglobins in the chick embryo occurs. This is the first investigation of an α-globin chain mutant which is synthesized during all stages of red cell development and may be a useful animal model for the study of hemoglobinopathies in vertebrates.  相似文献

Miscanthus x giganteus's efficacy as an energy crop relies on maintaining low greenhouse gas (GHG) emissions. As demand for Miscanthus is expected to rise to meet bioenergy targets, fertilizers and composts may be employed to increase yields, but will also increase GHG emissions. Manipulation experiments are vital to investigate the consequences of any fertilizer additions, but there is currently no way to measure whole‐plant GHG fluxes from crops taller than 2.5 m, such as Miscanthus, at the experimental plot scale. We employed a unique combination of eddy covariance (EC), soil chambers and an entirely new automated chamber system, SkyBeam, to measure high frequency (ca. hourly) fluxes of carbon dioxide (CO2), methane (CH4) and nitrous oxide (N2O) from a Miscanthus crop amended with green compost. Untreated controls were also monitored in a fully replicated experimental design. Net ecosystem exchange (NEE) of CO2 was partitioned into soil respiration (Rs), gross primary productivity (GPP) and ecosystem respiration, and the crop was harvested to determine the effect of compost on crop productivity. Compost increased NEE emissions by 100% (p < .05), which was the result of a 20% increase of Rs (p < .06) and a 32% reduction in GPP (p < .05) and biomass of 37% (p < .06). Methane fluxes were small and unaffected by compost addition. N2O emissions increased 34% under compost during an emission event; otherwise, fluxes were low and often negative, even under dry conditions. Diurnal variation in N2O fluxes, with uptake during the day and emission at night was observed. These fluxes displayed a negative relationship with soil temperature and a hitherto undescribed diurnal temperature hysteresis. We conclude that compost addition negatively affected the productivity and environmental effects of Miscanthus cultivation during the first year following application.  相似文献

Coral reef management techniques such as relocation and transplantation are increasingly implemented in the context of increasing coastal development and a global decline of coral reefs over the last 30 years. A 170 m submarine desalination pipeline was constructed in 2020 to discharge wastewater from the desalination plant for Hayman Island resort, Whitsundays, Queensland, Australia. Pre-construction site assessments were conducted indicating a healthy, diverse and recovering coral community between intertidal and 12 m depth in the proposed route of the pipeline. Mitigation options included the selection of a pipeline route that minimised impact on coral, and relocation and transplantation of hard corals. Two hundred and four corals comprising 35 species from 15 genera, with estimated sizes ranging from small (less than 2 kg) to extra large (over 50 kg), were relocated from the pipeline footprint to a similar nearby site. The estimated total weight of relocated corals was 873–2850 kg. The most common species transplanted were Hump Coral (Porites lutea) (27%), Lesser Star Coral (Goniastrea aspera) (8.3%) and Starflower Coral (Astreopora ocellata) (7.8%). Individual coral survivorship and growth was monitored at zero, one, six, 12 and 24 months. After 24 months total coral survival was 77.5%. The survivorship of relocated coral exceeded the mean for global coral restoration projects and was deemed successful by the regulator. To assist benchmarking of future coral relocation projects we propose a standard of below 50% as poor, 50–60% as below average, over 60% as acceptable and over 80% coral survival at two years as excellent.  相似文献

Capsule: Diversionary feeding reduced Hen Harrier Circus cyaneus nestlings’ natural food intake by half. Red Grouse Lagopus lagopus scotica chicks constituted 0–4% of all nestling food items. Annually, this reduced annual grouse chick production by 0–6%.

Aim: To quantify proportions of diversionary and natural food (including grouse) delivered to Hen Harrier nestlings in relation to brood size, male status and natural prey abundance.

Methods: We recorded diversionary food provisioned to 25 Hen Harrier broods (2008–15) and studied the diet of 15 broods using observations from hides, nest cameras and regurgitated pellet analysis. Variation in nestling diet was analysed using compositional analysis.

Results: Hen Harriers took 76% of diversionary food provided. Depending on assessment method, average nestling diet was 44–53% diversionary food, 39–55% natural prey (including 24–45% passerines, 4–15% small mammals, 0–4% grouse chicks) and 0–9% unknown items. The amount of diversionary food consumed was not influenced by male status, brood size or natural prey abundance. The number of Red Grouse chicks delivered annually was 34–100% lower than expected under unfed conditions, however, the confidence intervals associated with these estimates were large.

Conclusion: Diversionary food influenced Hen Harrier nestling diet and reduced the number of Red Grouse chicks taken relative to modelled predictions. It may help reduce conflict between Hen Harrier conservation and Red Grouse shooting, but only if overall grouse productivity is thereby maintained or increased.  相似文献

Ranger-led law enforcement patrols are the primary, site-level response to – and the most common source of data on – illegal activity threatening wildlife in protected areas. Yet evidence that patrols effectively deter rule-breaking is limited, and common management metrics for evaluating deterrence, which use ranger-collected data, are particularly vulnerable to bias. “Differenced plots” (of the association between change in patrol effort and subsequent change in illegal activity) were recently proposed as a simple, new metric for deterrence, which, in tests with simulated patrol data, were more robust than the common alternatives. Here, we trial application of differenced plots to real patrol data collected in four protected areas, and explore methods for applying the metric in practice, using two indicators of rule-breaking: snares, and people. We find evidence which is consistent with deterrence in some but not all sites, over shorter timescales than observed hitherto: increases in patrol effort were associated with subsequent reductions in snaring in one site, and in the presence of people in two sites. However, whether pressure on wildlife had been reduced or merely displaced was unclear from differenced plots, nor could the metric confirm absence of deterrence, raising questions for future applications. Our findings suggest differenced plots can be a useful metric, particularly for exploring variation in deterrence within sites, but should be applied and interpreted with care, and further work is urgently needed to determine whether and how patrols deter illegal activity, and to evaluate the effect reliably.  相似文献

Summary Kanamycin, paromomycin, and neomycin, like streptomycin, permanently bleach Euglena gracilis. This effect, along with general toxicity, is opposed by Mg, histidine or a combination of pantothenate, nicotinic acid, and threonine. Such opposition is thought to be peripheral effects centered on uptake and transport.Certain antihistamines, notably tripelennamine, methapyrilene, and pyrilamine induce permanent bleaching. Diphenylhydramine and phenindamine induced temporary bleaching. Doxylamine, antazoline, pyrathiazine, pheniramine, prophenpyridamine, and promethazine did not bleach when tested up to inhibitory concentrations.Bleaching by streptomycin+heat was additive, not synergistic.The evidence at hand for the mode of action of ultraviolet irradiation, streptomycin antibiotics, and erythromycin suggests, as a working hypothesis, that the common factor may be interference with nucleic acid metabolism; the common factor in bleaching by antibiotics may be simultaneous provision of a molecular grouping favoring uptake and transport of the active moiety, which in turn may be rare sugars interfering with ribose and desoxyribose in the photosynthetic apparatus.New antibiotics of the streptomycin family might well be screened for bleaching activity as a possible index of damage to the 8th cranial nerve, for so far the correlation is excellent for this class of antibiotic.This paper is dedicated to Professor Dr. E. Stimulated peripheral blood mononuclear cells (PBMC) can oxidize normal lipoproteins, and sufficiently oxidized lipoproteins are cytotoxic. However, the role of lipid peroxidation in the inhibition of mitogen-stimulated PBMC proliferation by physiologic concentrations of normal lipoproteins is unclear. In the present investigation, normal low density lipoprotein (LDL) and very low density lipoprotein (VLDL) suppressed [3H]thymidine incorporation and gamma interferon production in concanavalin A-stimulated PBMC without causing cell death. This suppression was accompanied by parallel increases in lipid peroxidation products measured as thiobarbituric acid reactive substances (TBARS). In contrast, high density lipoprotein (HDL) failed to inhibit PBMC and TBARS remains low. Differences between the PBMC suppression from LDL, VLDL, and HDL were best accounted for by normalizing the lipoprotein concentrations by their total lipid content. Moreover, the antioxidants superoxide dismutase and butylated hydroxytoluene each substantially ameliorated the inhibition of PBMC caused by LDL, and reduced the levels of lipid peroxidation products that were generated. Altogether, these results suggest that reactive oxygen species generated by stimulated PMBC may cause oxidative alterations of normal lipoproteins that may, in turn, account for much of the previously reported inhibition of PBMC by normal lipoproteins.  相似文献

The three subspecies of Spotted Owl (Northern, Strix occidentalis caurina; California, S. o. occidentalis; and Mexican, S. o. lucida) are all threatened by habitat loss and range expansion of the Barred Owl (S. varia). An unaddressed threat is whether Barred Owls could be a source of novel strains of disease such as avian malaria (Plasmodium spp.) or other blood parasites potentially harmful for Spotted Owls. Although Barred Owls commonly harbor Plasmodium infections, these parasites have not been documented in the Spotted Owl. We screened 111 Spotted Owls, 44 Barred Owls, and 387 owls of nine other species for haemosporidian parasites (Leucocytozoon, Plasmodium, and Haemoproteus spp.). California Spotted Owls had the greatest number of simultaneous multi-species infections (44%). Additionally, sequencing results revealed that the Northern and California Spotted Owl subspecies together had the highest number of Leucocytozoon parasite lineages (n = 17) and unique lineages (n = 12). This high level of sequence diversity is significant because only one Leucocytozoon species (L. danilewskyi) has been accepted as valid among all owls, suggesting that L. danilewskyi is a cryptic species. Furthermore, a Plasmodium parasite was documented in a Northern Spotted Owl for the first time. West Coast Barred Owls had a lower prevalence of infection (15%) when compared to sympatric Spotted Owls (S. o. caurina 52%, S. o. occidentalis 79%) and Barred Owls from the historic range (61%). Consequently, Barred Owls on the West Coast may have a competitive advantage over the potentially immune compromised Spotted Owls.  相似文献
