Two studies were conducted in 83 college men to determine the degree of generality of individual differences in upper body muscular strength assessed by different testing modes. In study 1 (N = 43), correlations were computed between four measures of upper body strength using the bench press movement, maximal isokinetic (0.09 rad.s-1), maximal fast (0.126 m.s-1) and slow (0.037 m.s-1) hydraulic, and one repetition maximum (1-RM) free weight bench press (BP). Compared to free weight BP, maximal strength during isokinetic and slow hydraulic BP was approximately 29% and approximately 8% larger, and fast hydraulic BP strength was approximately 63% lower (p less than 0.05). Simple linear regression of isokinetic BP on 1-RM BP yielded r = 0.79, error of prediction (SE) = 12%, and generality = 81%. The corresponding averaged values for the regression of slow and fast hydraulic BP on free weight 1-RM BP were r = 0.77, SE = 13.5%, and generality = 84%. In Study 2 (N = 40), testing included maximal isokinetic concentric and eccentric arm flexion and extension at 0.524, 1.570, and 2.094 rad.s-1. The ratio of concentric to eccentric torque at the 3 speeds averaged 0.68 (flexion) and 0.70 (extension), and eccentric torques were 32% and 30% greater than concentric torques (p less than 0.05). Fifty-five consecutive female volunteers were evaluated. Linear measurements of the breast and nipple position were determined to fixed points on the chest wall. Volume determination was done by a standard chest-wall casting technique. A highly significant difference was found between left and right breasts in the axilla-to-nipple distance and nipple-to-midline measurements. A significant difference was also found in the distance from the lowest point of the breast (patient upright) to the nipple. Analysis of the volumetric difference between right and left breasts did not achieve statistical significance. 相似文献

We hypothesized that resistance training with combined eccentric and concentric actions, and concentric action only, should yield similar changes in muscular strength. Subjects in a free weight group trained three times a week for 12 wk with eccentric and concentric actions (FW, n = 16), a second group trained with concentric-only contractions using hydraulic resistance (HY; n = 12), and a control group did not train (n = 11). Training for FW and HY included five sets of supine bench press and upright squat at an intensity of 1-6 repetition maximum (RM) plus five supplementary exercises at 5-10 RM for a total of 20 sets per session for approximately 50 min. Testing at pre-, mid-, and posttraining included 1) 1 RM bench press and squat with and 2) without prestretch using free weights; 3)isokinetic peak force and power for bench press and squat at 5 degrees/s, and isotonic peak velocity and power for bench press with 20-kg load and squat with 70-kg load; 4) hydraulic peak bench press force and power, and peak knee extension torque and power at fast and slow speeds; and 5) surface anthropometry (fatfolds and girths to estimate upper arm and thigh volume and muscle area). Changes in overall fatness, muscularity, and muscle + bone cross-sectional area of the limbs did not differ between groups (P greater than 0.05). Background
Swine is an important agricultural commodity and biomedical model. Manipulation of the pig genome provides opportunity to improve production efficiency, enhance disease resistance, and add value to swine products. Genetic engineering can also expand the utility of pigs for modeling human disease, developing clinical treatment methodologies, or donating tissues for xenotransplantation. Realizing the full potential of pig genetic engineering requires translation of the complete repertoire of genetic tools currently employed in smaller model organisms to practical use in pigs.Results
Application of transposon and recombinase technologies for manipulation of the swine genome requires characterization of their activity in pig cells. We tested four transposon systems- Sleeping Beauty, Tol2, piggyBac, and Passport in cultured porcine cells. Transposons increased the efficiency of DNA integration up to 28-fold above background and provided for precise delivery of 1 to 15 transgenes per cell. Both Cre and Flp recombinase were functional in pig cells as measured by their ability to remove a positive-negative selection cassette from 16 independent clones and over 20 independent genomic locations. We also demonstrated a Cre-dependent genetic switch capable of eliminating an intervening positive-negative selection cassette and activating GFP expression from episomal and genome-resident transposons.Conclusion
We have demonstrated for the first time that transposons and recombinases are capable of mobilizing DNA into and out of the porcine genome in a precise and efficient manner. This study provides the basis for developing transposon and recombinase based tools for genetic engineering of the swine genome. 相似文献5.

cell wall retention of the recombinant enzyme. Given the presence of five potential N-glycosylation sites in the amino acid sequence coded by celA, the effect of glycosylation on the enzymatic activity of the recombinant enzyme was investigated by expressing the recombinant
fusion proteins in both, standard and glycosylation-deficient strains of S. cerevisiae. Correct targeting of the recombinant fusion proteins was confirmed by Western immunoblot using Pir-specific antibodies,
while enzymatic activity on carboxymethyl cellulose was demonstrated on plate assays, zymographic analysis and colorimetric
assays. Hyperglycosylation of the enzyme when expressed in the standard strain of S. cerevisiae did not affect activity, and values of 1.2 U/ml were obtained in growth medium supernatants in ordinary batch cultures after
24 h. These values compare quite favorably with those described for other recombinant endoglucanases expressed in S. cerevisiae. This is one of the few reports describing the expression of Bacillus cellulases in S. cerevisiae, since yeast expressed recombinant cellulases have been mostly of fungal origin. Trypsin inhibitor was converted to hydrophobic states by covalently combining cholesteryl groups using an acylation reaction, and was immobilized in reverse micelles composed of a nonionic surfactant. Using this reverse micellar phase containing trypsin inhibitor as an affinity ligand, trypsin was selectively separated with high recoveries from a mixture of several kinds of contaminating proteins by forward and backward extraction. We assessed body composition (height, body mass, body mass index, body fat by densitometry, fat mass, fat-free mass, and lean/fat ratio) and performance (10- and 40-yd sprints, pro shuttle run, vertical jump, sit and reach, and bench press) in 77 National Collegiate Athletic Association Division III football players. Data were analyzed by position and playing status. Significant differences (p = 0.05) were found between positions for all body composition measurements and all performance tests except the sit and reach. Starters outperformed nonstarters in all performance tests except the 10-yd sprint and sit and reach (p = 0.05). Correlations (r) for percent body fat and performance tests ranged from 0.52 to 0.70, and common variance with the effects of body mass removed ranged from 8 to 23%. Percent body fat is not closely correlated with results of commonly administered performance tests in Division III football players. 相似文献
