Biological pest control by means of beneficial organisms is for long part of agriculture and Integrated Pest Management (IPM). A new and efficient strategy to control the most common timber pest species in churches and museums, the furniture beetle Anobium punctatum (De Geer) (Coleoptera: Anobiidae), is based on the parasitoid wasp species Spathius exarator (Linnaeus) (Hymenoptera: Braconidae). Once this braconid wasp detects its host species beneath the surface, it pierces the wood with its ovipositor to lay one single egg onto the beetle larva. After hatching, the wasp larva feeds on the beetle larva thereby killing it. Afterwards, it pupates and emerges through a self-gnawed hole as an adult wasp. The tiny, 0.5-mm-wide exit hole can easily be distinguished from the 1- to 2-mm-wide exit hole of A. punctatum. Laboratory tests revealed that female wasps have an average life span of 85 days and produce a total of 24 offspring, when nutrition is provided. Between 2012 and 2019, braconid wasps were introduced into 54 different A. punctatum infested buildings. Treatment success was monitored by examining exit holes of new beetles and wasps thereby calculating corresponding parasitism rates. After the first year of treatment, parasitism rates were significantly higher with a mean value of 0.15 when compared to untreated objects with a natural parasitism and a mean value of 0.08 (n = 54). Following treatment of three objects over a period of eight years, parasitism rates continuously increased from 0.02 up to 0.31. In a church organ, which was treated during this period, the monitoring revealed a complete suppression of pest activity. These data prove that this biological method of pest control is an efficient, sustainable and non-toxic option to manage the common furniture beetle. 相似文献

Cultured bovine capillary endothelial (BCE) cells produce low levels of collagenolytic activity and significant amounts of the serine protease plasminogen activator (PA). When grown in the presence of nanomolar quantities of the tumor promoter 12-O-tetradecanoyl phorbol-13-acetate (TPA), BCE cells produced 5-15 times more collagenolytic activity and 2-10 times more PA than untreated cells. The enhanced production of these enzymes was dependent on the dose of TPA used, with maximal response at 10(-7) to 10(-8) M. Phorbol didecanoate (PDD), an analog of TPA which is an active tumor promoter, also increased protease production. 4-O-methyl-TPA and 4α-PDD, two analogs of TPA which are inactive as tumor promoters, had no effect on protease production. Increased PA and collagenase activities were detected within 7.5 and 19 h, respectively, after the addition of TPA. The TPA-stimulated BCE cells synthesized a urokinase-type PA and a typical vertebrate collagenase. BCE cells were compared with bovine aortic endothelial (BAE) cells and bovine embryonic skin (BES) fibroblasts with respect to their production of protease in response to TPA. Under normal growth conditions, low levels of collagenolyic activity were detected in the culture fluids from BCE, BAE, and BES cells. BCE cells produced 5-13 times the basal levels of collagenolytic activity in response to TPA, whereas BAE cells and BES fibroblasts showed a minimal response to TPA. Both BCE and BAE cells exhibited relatively high basal levels of PA, the production of which was stimulated approximately threefold by the addition of TPA. The observation that BCE cells and not BAE cells produced high levels of both PA and collagenase activities in response to TPA demonstrates a significant difference between these two types of endothelial cells and suggests that the enhanced detectable activities are a property unique to bovine capillary and microvessel and endothelial cells. 相似文献

Assessment of physicochemical and pharmacological properties is now conducted at very early stages of drug discovery for the purpose of accelerating the conversion of hits and leads into qualified development candidates. In particular, in vitro absorption, distribution, metabolism and elimination (ADME) assays and in vivo drug metabolism pharmacokinetic (DMPK) studies are being conducted throughout the discovery process, from hit generation through to lead optimization, with the goal of reducing the attrition rate of these potential drug candidates as they progress through development. Because the continuing trend in drug discovery has been to access ADME information earlier and earlier in the discovery process, the need has arisen within the analytical community to introduce faster and better analytical methods to enhance the 'developability' of drug leads. Strategies for streamlined ADME assessment of drug candidates in discovery and pre-clinical development are presented within. 相似文献

Stable-isotope tracer experiments performed in vitro are evaluated for their utility in differentiating between pyruvate dehydrogenase and cytochrome oxidase deficiencies, two of several enzyme defects commonly associated with the lactic acidemias. Fibroblasts of enzyme-deficient individuals and of age-matched controls are grown in medium containing [U-13C]glucose. Direct analysis of cells and conditioned culture medium provides only minor differences in the organic acid/amino acid GC-MS profiles, making differentiation of enzyme defects difficult by this method. However, differences have been found in the glucose turnover into various cell metabolites, making differentiation of these two enzyme defects possible. The cellular pool of glutamic acid experiences 13C-enrichment in both the control and cytochrome oxidase deficient lines, but not in the pyruvate dehydrogenase-deficient line. The cellular pool of an unknown, possibly an aminopentose sugar, on the other hand, experiences 13C-enrichment in the pyruvate dehydrogenase and control lines, but not in the cytochrome oxidase line. These observations, as well as other differences in the extent of enrichment into various metabolite pools, suggest that this stable-isotope approach, in vitro, is feasible and may allow these two enzyme defects to be differentiated in a definitive manner. Such stable-isotope experiments are easy to carry out with cultured cells and are inexpensive. Applications of the technique to other genetic disorders might be appropriate. 相似文献

A stereotactic system has been designed to address the problem of achieving symmetry in complex and extensive craniofacial defects. Preliminary testing suggests that such a system, which allows for the intraoperative application of preoperative CT planning, will be useful in guiding the reconstruction of congenital or acquired bony time, is being used to investigate the correlation of intraoperative globe position following enophthalmos correction with long-term outcome, particularly as it relates to the size and location of the orbital defect, and the timing of the procedure. 相似文献

The role of craniofacial surgical techniques and immediate bone grafting in the management of complex craniofacial trauma has been reviewed. Four hundred and one patients with complex facial injuries have been treated. Two hundred and forty-one primary bone and cartilage grafts have been performed in 66 patients. Complex facial injuries should be managed by direct exposure, reduction, and fixation of all fractures utilizing interfragmentary wiring. Very comminuted or absent bone is replaced by immediate bone grafting, producing a stable skeleton without the need for external fixation devices. Associated mandibular fractures are managed with rigid internal fixation utilizing A-O technique. Results of immediate bone grafting have been excellent, and complications are rare. All deformities should be corrected, whenever possible, during the initial operation. This one-stage reconstruction of even the most complex facial injuries will prevent severe postoperative traumatic deformity and disability that may be extremely difficult or impossible to correct secondarily. 相似文献

Bromine activation of phenyl 4-O-acetyl-2,3,6-trideoxy-3-C-methyl-3-trifluoroacetamido-1-thio-alpha,beta-L-ribo-hexopyranoside and attempted coupling with an acceptor in the presence of silver silicate gave an unusual bicyclic product, 2-trifluoromethyl-(4-O-acetyl-2-bromo-2,3,6-trideoxy-3-C-methyl-alpha-L-altrohexopyrano)-[3,2,1-d,e]-2-oxazine, instead of the expected disaccharide. Detailed investigation supported by X-ray crystallographic analysis showed that a trans dibromide is an intermediate in this reaction and that the dibromide is likely formed from a glycal that is generated by elimination during the coupling step. 相似文献
