Resveratrol is a plant-derived phenol but the mechanism that regulates its biosynthesis remains unidentified. Stilbene synthase (STS) catalyzes resveratrol formation in vivo and we have proposed that inducers of resveratrol production affect STS expression through an unidentified epigenetic mechanism. To investigate the role of DNA methylation in resveratrol biosynthesis, we treated both rolB transgenic and empty vector control Vitis amurensis cell cultures with the DNA demethylation agent, 5-azacytidine. Treated cells had increased resveratrol production through activation of VaSTS10 expression. The lowest levels of cytosine methylation were at the 5′- and 3′-ends of the VaSTS1 protein-coding sequence. Cytosine methylation decreased mostly at the 5′- and 3′-ends of VaSTS10 after azaC treatment with an intriguing regularity in the number of cytosine nucleotides within the 5′- and 3′- ends of the protein-coding sequences. Thus, cytosine methylation is crucial for the regulation of the resveratrol biosynthetic pathway.  相似文献

The morphological patterns of the cultivated cells of primary mesenchyme and the spicules of the larval skeleton of the sea urchin Strongylocentrotus nudus were quantified, and the value of their fractal dimensions (D) was determined with ImageJ 1.20s software. It was shown that during cytodifferentiation, the values of D in the fractal (fractional) dimension, which reflects the complex spatial organization of the spiculogenous mesenchyme elements in two-dimensional space, increase to values close to 1.7. The invertible treatment with cytochalasin, which destroys the system of the actin filaments, suppresses the normal control of biomineralization and causes a complex form of spicules, the fractal dimension of which varies within 1.5–1.6. Thus, the determination of the fractal dimension value serves as evidence of the fractional essence of the patterns studied, quantifies the spatially complex organization of cells and their assemblies during morphogenesis, and allows us to estimate the variation in the spicule morphology after cytochalasin treatment.  相似文献

The box-counting method for calculating the fractal dimension (D) with the ImageJ 1.20s software is used as a tool for quantitative analysis of the neuronal morphology in the fish brain. The fractal dimension was determined for several types of neurons in the brain of two teleost species, Pholidapus dybowskii and Oncorhynchus keta. These results were compared with those obtained for some neurons of the human brain. The fractal (fractional) dimension (D), as a quantitative index of filling of two-dimensional space by the black and white image of a cell, is shown to vary from 1.22 to 1.72 depending on the type of neuron. The fractal dimension reaches its maximum in less specialized neurons that carry out a number of different functions. On the other hand, highly specialized neurons display a relatively low fractal dimension. Thus, the fractal dimension serves as a numerical measure of the spatial complexity of the neuron and correlates with the morphofunctional organization of the cell.  相似文献

For the quantitative characteristics of the morphology of two groups of spinal cord neurons in the cherry salmon at the early stages of ontogenesis, the fractal dimension and some more traditional morphometric indices, such as the total length of neuronal branches, the number of terminal branches and branchpoints, and the cell area, are determined. The values of the morphometric indices and the fractal dimension of the bivariate neuronal pattern are shown to increase in the course of ontogenesis from the first to the second year of fish life. The correlation between the main morphometric value and the fractal dimension is found, and their correspondence to the morphometric modifications of the dendrite tree in the neurons investigated during ontogenesis is detected.  相似文献

This paper deals with the dendritic field structure of three large ganglion cell types in the retina of a marine teleost, Pholidapus dybowskii. Cells were retrograde labeled with horseradish peroxidase applied to lesioned fibers of the optic nerve. Their morphology was studied in wholemounted retinae. Dendritic fields of αab cells were more complex. Their structural complexity measured using Kolmogorov and information fractal dimensions exceeded significantly those of αa and biplexiform cells. The latter two types exhibited no significant differences in complexity and spatial heterogeneity of dendritic field. The cell types studied differed dramatically in the relationships between fractal and nonfractal parameters of their dendritic arbors. The functional and evolutionary implications of the dendritic field structure of retinal ganglion cells are discussed.  相似文献
