Aims Our aim is to reconstruct decadal scale development of historical landscapes during the last 1000 years by means of fossil pollen analysis of annually laminated lake sediments, and detailed historical maps and documents. Location Lake Rõuge Tõugjärv (Estonia), a small lake with annually laminated lake sediments situated in a dense prehistoric setting. Methods The chronology of the palaeodata is based on the annual laminations supported by AMS 14C and 210Pb dating and 137Cs, 241Am, and spheroidal carbonaceous particle marker horizons. The time‐scale and resolution allows fine sampling (the pollen samples generally comprise 3.5 years) and vegetation change reconstruction. Relevant source area of pollen (RSAP) of the lake was estimated, and the statistical zonation, rate of change, palynological richness, and DCA and PCA ordinations were generated on the basis of the pollen data. The historical calibration data set (maps, numerical information on population, domestic stock, farmland division, etc.) is based on archival material preserved in the Estonian Historical Archives. Results The topmost part (0–180 cm) of the sediment column of Lake Rõuge Tõugjärv, covering the last 1000 years, is visibly laminated carbonaceous gyttja. The varve chronology extends from ad 2000 to ad 1339, with a cumulative ± 9‐year error estimate. Beyond this the chronology is extrapolated using the 14C date and varve age–depth estimations. The simulation of the RSAP of Lake Tõugjärv shows that the major portion of the pollen loading originating from local vegetation is derived from plants growing within 2000 m of the sampling site. The pollen record divides into five statistically significant subgroups, which fall on the PCA plot into three clusters reflecting the general openness–closedness of the landscape. During the period between ad 1000 and 1200 (RT 1) the Rõuge area was generally wooded with birch, spruce and pine forests. The advancement of extensive farming gradually opened up the landscape between ad 1200 and 1650 (RT 2 and RT 3). The maximum openness of the landscape was reached between ad 1650 and 1875 (RT 4), with the most open period in the late eighteenth century. Historical maps from 1684 and 1870–99 and available quantitative data on population, domestic stock, farmland division, etc. show the same trend. The pollen data covering the last 125 years, and maps from 1935 and 1995, show the reduction of arable land in RSAP of the lake under investigation and the reduction of open land to an extent comparable with the end of the seventeenth century. Main conclusions The formation and development of the cultural landscape at Rõuge over the last 1000 years is characterized by rapid changes in floristic richness and rates of vegetation change attributed to certain historic processes in the RSAP. Five phases of landscape and social development are clearly distinguished during the last 1000 years. The decadal scale vegetation response to human‐induced forcing agrees with historical maps and documents and could be used for past landscapes prior to the period with solid historical data.  相似文献

Protective properties of immunoglobulin A (IgA) monoclonal antibodies (MAbs) directed against O and H antigens of Salmonella enterica serotype Enteritidis (S. enteritidis) were evaluated in a model of generalized infection after intranasal (i.n.) inoculation of BALB/c mice. Passive i.n. instillation of antibodies 1 h before i.n. challenge did not prevent infection, and mice developed rapid inflammatory response in the lower respiratory tract. The passive systemic immunization was partially protective and a single intravenous (i.v.) injection of both O and H antigen specific IgA antibodies prolonged survival period of the infected animals. Permanent secretion of O:9 specific IgA MAb 177E6 into the respiratory tract in a "backpack" tumor model protected 50% of animals infected i.n. with a high dose of virulent S. enteritidis strain. Thus, secretory IgA (S-IgA) directed against O:9 antigen alone can prevent bacterial invasion in the respiratory epithelium.  相似文献

Over 40 different mutations in the cardiac myosin heavy chain gene (MYH7) have been associated with familial hypertrophic cardiomyopathy (FHC), but no study has analyzed variation at this locus within the normal human population. Here we determine the extent and distribution of nucleotide variation in the 5808-bp MYH7 coding sequence in 25 normal individuals without FHC. We identified six single-nucleotide polymorphisms, none of which changes the encoded amino acid. At one of these sites, the frequencies of both alleles are equal; at the other five sites, the frequency of the rarer allele varies from 0.02 to 0.08. The nucleotide diversity (π) calculated from these data is 1.73×10−4±0.49×10−4, which is lower than the nucleotide diversity found in most other human autosomal genes. Substitution analysis of homologous genes between human and rodent also indicates that the MYH7 sequence has evolved at a very slow rate. The rate of both synonymous and nonsynonymous substitutions, especially in the portion of the sequence that encodes the α-helical myosin rod, is extremely low. The low level of even silent sequence variation in MYH7 in comparisons between human sequences and between human and rodent sequences may be a consequence of strong selective pressure against mutations that cause cardiomyopathy.  相似文献

Lecanosticta acicola is a pine needle pathogen causing brown spot needle blight that results in premature needle shedding with considerable damage described in North America, Europe, and Asia. Microsatellite and mating type markers were used to study the population genetics, migration history, and reproduction mode of the pathogen, based on a collection of 650 isolates from 27 countries and 26 hosts across the range of L. acicola. The presence of L. acicola in Georgia was confirmed in this study. Migration analyses indicate there have been several introduction events from North America into Europe. However, some of the source populations still appear to remain unknown. The populations in Croatia and western Asia appear to originate from genetically similar populations in North America. Intercontinental movement of the pathogen was reflected in an identical haplotype occurring on two continents, in North America (Canada) and Europe (Germany). Several shared haplotypes between European populations further suggests more local pathogen movement between countries. Moreover, migration analyses indicate that the populations in northern Europe originate from more established populations in central Europe. Overall, the highest genetic diversity was observed in south‐eastern USA. In Europe, the highest diversity was observed in France, where the presence of both known pathogen lineages was recorded. Less than half of the observed populations contained mating types in equal proportions. Although there is evidence of some sexual reproduction taking place, the pathogen spreads predominantly asexually and through anthropogenic activity.  相似文献

The red list has become a ubiquitous tool in the conservation of species. We analyzed contemporary trends in the threat levels of European orchids, in total 166 species characterized in 27 national red lists, in relation to their reproductive biology and growth form, distribution area, and land cover where they occur. We found that species in central Europe are more threatened than those in the northern, southern, or Atlantic parts of Europe, while species were least threatened in southern Europe. Nectarless and tuberous species are significantly more threatened than nectariferous and rhizomatous taxa. Land cover (ratios of artificial land cover, area of pastures and grasslands, forests and inland wetlands) also significantly impacted the threat level. A bigger share of artificial land cover increases threat, and a bigger share of pasture and grassland lowers it. Unexpectedly, a bigger share of inland wetland area in a country increased threat level, which we believe may be due to the threatened nature of wetlands themselves relative to other natural land cover types. Finally, species occurring in multiple countries are on average less threatened. We believe that large‐scale analysis of current IUCN national red lists as based on their specific categories and criteria may particularly inform the development of coordinated regional or larger‐scale management strategies. In this case, we advocate for a coordinated EU protection and restoration strategy particularly aimed at central European orchids and those occurring in wetland area.  相似文献

It has been shown that the key homologous recombination protein Rad51accumulates in DNA damage‐induced nuclear foci that are attached to the nuclear matrix. In the present communication we attempted to find whether Rad51 contains a functional domain responsible for nuclear matrix binding. By alignments of the sequences encoding nuclear matrix targeting signals of human nuclear matrix binding proteins with the whole length human Rad51sequence a putative nuclear matrix targeting signal was identified. To prove that it is responsible for the nuclear matrix association of Rad51 18 base pairs encoding a cluster of hydrophobic amino acids in the human Rad51 Flag‐tagged gene were deleted. The formation of damage‐induced Rad51 foci and their association with the nuclear matrix were monitored in HeLa cells transfected with the wild‐type and the mutated Rad51gene after treatment with mitomycin C. The results showed that while the wild‐type protein formed Rad51 foci attached to the nuclear matrix, the mutated Rad51 failed to form DNA damage‐induced nuclear foci. The loss of foci formation activity of the mutated protein was not due to impaired ability to bind double‐stranded DNA in an ATP‐dependant way in vitro and to bind chromatin in vivo. These data suggest that the assembly of Rad51 into nuclear foci is assisted by association with the nuclear matrix, which may support the spatial organization of the process of repair by homologous recombination. Clothing evaporative resistance is an important input in thermal comfort models. Thermal manikin tests give the most accurate and reliable evaporative resistance values for clothing. The calculation methods of clothing evaporative resistance require the sweating skin surface temperature (i.e., options 1 and 2). However, prevailing calculation methods of clothing evaporative resistance (i.e., options 3 and 4) are based on the controlled nude manikin surface temperature due to the sensory measurement difficulty. In order to overcome the difficulty of attaching temperature sensors to the wet skin surface and to enhance the calculation accuracy on evaporative resistance, we conducted an intensive skin study on a thermal manikin ‘Tore’. The relationship among the nude manikin surface temperature, the total heat loss and the wet skin surface temperature in three ambient conditions was investigated. A universal empirical equation to predict the wet skin surface temperature of a sweating thermal manikin was developed and validated on the manikin dressed in six different clothing ensembles. The skin surface temperature prediction equation in an ambient temperature range between 25.0 and 34.0 °C is Tsk=34.0–0.0132HL. It is demonstrated that the universal empirical equation is a good alternative to predicting the wet skin surface temperature and facilitates calculating the evaporative resistance of permeable clothing ensembles. Further studies on the validation of the empirical equation on different thermal manikins are needed however.  相似文献

The aim of this study was to explore the extent of varroa mite resistance to fluvalinate in Israel and to determine the underlying biochemical mechanism. Assays at different apiaries indicated varroa mite resistance at three of the five sites tested. Dose response assays conducted with tau-fluvalinate on mites obtained from different sites indicated uneven resistance. A monooxygenase assay revealed an increased rate (approximately 20-fold) of activity in mites that were not controlled by the pesticide, as compared to activity in mites from untreated colonies. A minor, 1.5–2.5 fold, increase of esterase activity was also noted in the resistant mites. This first demonstration of a fluvalinate-resistance mechanism in varroa mites points to the need for more vigorous resistance management practices to control the pest.  相似文献

The goal of this study was to characterize the genetic contribution to both forced and voluntary exercise performance and to determine whether performance in these two paradigms is controlled by similar genetic influences. There were marked strain differences in treadmill exercise performance, with Swiss Webster (SW) and FVB/NJ mice showing elevated performance and C57BL/6J animals showing decreased performance compared with all other strains. There was no apparent relationship between treadmill performance and voluntary wheel performance, with the exception of SW mice, which demonstrated high performances on both the treadmill and the voluntary wheel. Numerous properties were measured to attempt to understand the basis for these differences in exercise performance. DBA/1J and SW mice exhibited significantly greater cardiac contractility than all other analyzed strains. Conversely, BALB/cByJ mice exhibited significantly reduced cardiac contractility compared with all other strains. Expression of molecular indicators of hypertrophy (atrial natriuretic factor and beta-myosin heavy chain) was significantly elevated in DBA/2J myocardium compared with all other analyzed strains.  相似文献
