Two new species of rhacophorid tree frog were identified in Taiwan. In both new taxa, derived reproductive characteristics of laying eggs in tree holes and oophagous tadpoles are shared with Kurixalus
eiffingeri, but they are divergent from each other in molecular genetics, mating calls, and tadpole and adult morphology. The morphological characteristics and the molecular phylogenetic evidence support the hypothesis that the two new species, Kurixalus
berylliniris
sp. n. and Kurixalus
wangi

Matrix metalloproteinase-9 plays an important role in the invasion and metastasis of various types of cancer cells. We have previously reported that excretory–secretory products from Clonorchis sinensis increases matrix metalloproteinase-9 expression. However, the regulatory mechanisms through which matrix metalloproteinase-9 expression affects cholangiocarcinoma development remain unclear. In the current study, we examined the potential role of excretory–secretory products in regulating the migration and invasion of various cholangiocarcinoma cell lines. We demonstrated that excretory–secretory products significantly induced matrix metalloproteinase-9 expression and activity in a concentration-dependent manner. Reporter gene and chromatin immunoprecipitation assays showed that excretory–secretory products induced matrix metalloproteinase-9 expression by enhancing the activity of nuclear factor-kappa B. Moreover, excretory–secretory products induced the degradation and phosphorylation of IκBα and stimulated nuclear factor-kappa B p65 nuclear translocation, which was regulated by extracellular signal-regulated kinase 1/2. Taken together, our findings indicated that the excretory–secretory product-dependent enhancement of matrix metalloproteinase-9 activity and subsequent induction of IκBα and nuclear factor-kappa B activities may contribute to the progression of cholangiocarcinoma. 相似文献

Severe Clonorchis sinensis infection is a significant risk factor for malignant changes in bile ducts and surrounding liver tissues occurring as a result of direct contact with C. sinensis worms and their excretory–secretory products (ESP). However, the intrinsic molecular mechanisms involved in these processes remain obscure. To determine the effects of C. sinensis infection on protein expression in host bile duct epithelium, we examined proteomic profile changes in the human cholangiocarcinoma cell line (HuCCT1) treated with ESP at 24 h. Using a combination of 2‐DE, quantitative image and MALDI‐TOF MS analysis, we identified 83 proteins that were translationally modulated in response to ESP, among which 49 were up‐regulated and 34 down‐regulated. These proteins were classified under various biological categories, including metabolism, cell structure and architecture, proteolysis, protein modification, transport, signal transduction, and reactive oxygen species (ROS) detoxification. In particular, ESP induced the expression of redox‐regulating proteins, including peroxiredoxins (Prdx 2, 3, and 6) and thioredoxin 1 (Trx 1), possibly via intracellular ROS generation. Application of the proteomic approach to identify ESP response proteins should be a prerequisite before further investigation to clarify the molecular pathways and mechanisms involved in C. sinensis infection of host cells. A large-scale enclosure experiment for lake restoration was carried out in Lake Wuli, a northern bay of shallow and eutrophic Lake Taihu in China. The large enclosure with an area of 10 ha was set up in the littoral zone and was bordered by waterproof fabric which did not cover the sediments. Multiple approaches were used and included fish removal, piscivorous fish stocking, shoreline reconstruction, aquatic macrophyte planting, benthic macro-animal stocking, and silver carp cultivation in pens for reduction of cyanobacteria. The results showed that the coverage of aquatic macrophytes increased from 0% to 45.7%. Mean concentrations of TN and TP inside the enclosure from May 2004 to May 2008 were 22.2% and 26.0% of those outside, respectively. Secchi depth was 0.40 m outside the enclosures and 0.75 m inside. However, responses of phytoplankton to the restoration project lagged behind improvement of water quality and reestablishment of aquatic plants. The phytoplankton biomass gradually decreased after the third year of the restoration. Stocking piscivorous fish and planting submerged macrophytes could not increase zooplankton biomass and enhance graze pressure on phytoplankton, most likely due to high omnivorous fish density and lower nutrition inside the enclosure. Higher grazing pressure of zooplankton on phytoplankton was observed in May and October every year. Zooplankton to phytoplankton biomass ratios were significantly negatively correlated with phytoplankton biomass outside (r = −0.440, p < 0.01) and inside the enclosure (r = −0.336, p < 0.05) from February 2004 to March 2007. Therefore, phytoplankton biomass inside and outside the enclosure was lower in May and October. Higher grazing pressure of zooplankton on phytoplankton in spring may result in occurrence of the clear-water phase that facilitated growth of submerged macrophytes in the littoral in Lake Wuli, and a clear-water state and improved water quality would likely be sustained throughout the year after reestablishment of submerged macrophytes. 相似文献

Numerous experimental and epidemiological studies have demonstrated a correlation between Clonorchis sinensis (C. sinensis) infestation and cholangiocarcinoma (CCA). However, the role of C. sinensis in the increased invasiveness and proliferation involved in the malignancy of CCA has not been addressed yet. Here, we investigated the possibility that C. sinensis infestation promotes expression of focal and cell-cell adhesion proteins in CCA cells and secretion of matrix metalloproteinases (MMPs). Adhesion proteins help maintain cell aggregates, and MMPs promote the three-dimensional invasion of cells into the neighboring extracellular matrix (ECM). Using a novel microfluidic assay, we quantitatively addressed the role of excretory-secretory products (ESPs) gradients from C. sinensis in promoting the invasion of cells into the neighboring ECM. 相似文献

Selegiline, an anti-Parkinson drug, has antioxidant and anti-apoptotic effects. To explore the effect of selegiline on sepsis, we used a clinically relevant animal model of polymicrobial sepsis. Cecal ligation and puncture (CLP) or sham operation was performed in male rats under anesthesia. Three hours after surgery, animals were randomized to receive intravenously selegiline (3 mg/kg) or an equivalent volume of saline. The administration of CLP rats with selegiline (i) increased arterial blood pressure and vascular responsiveness to norepinephrine, (ii) reduced plasma liver and kidney dysfunction, (iii) attenuated metabolic acidosis, (iv) decreased neutrophil infiltration in liver and lung, and (v) improved survival rate (from 44% to 65%), compared to those in the CLP alone rats. The CLP-induced increases of plasma interleukin-6, organ superoxide levels, and liver inducible nitric oxide synthase and caspase-3 expressions were ameliorated by selegiline treatment. In addition, the histological changes in liver and lung were significantly attenuated in the selegiline -treated CLP group compared to those in the CLP group. The improvement of organ dysfunction and survival through reducing inflammation, oxidative stress and apoptosis in peritonitis-induced sepsis by selegiline has potential as an adjuvant agent for critical ill. 相似文献

通过受控试验,研究了4个光照处理(100%、50%、20%、10%的自然光)对菹草(Potamogeton crispus)外部形态与内部结构的影响.结果显示:(1)100%和50%自然光处理对菹草的总生物量影响不显著,20%和10%的自然光显著降低了菹草的总生物量.(2)在生物量分配方面,50%自然光处理的根冠比值显著低于其他3个处理组,表现出菹草对地上部分茎叶生物量的投资较多.(3)在50%自然光条件下,菹草的株高、植株叶片数、顶枝叶片数明显高于其他3个处理组的对应参数,说明适当的遮荫会促进菹草植株的生长;10%自然光下的各参数值均最小(除根/冠比值),表明已较严重地限制了菹草的正常生长.(4)20%和10%自然光的遮光处理增加了菹草的叶片厚度,100%、50%、20%和10%的自然光下分别为0.260、0.297、0.440、 0.306 mm.(5)100%自然光叶片的叶肉细胞没有形成发达通气组织,50%自然光的菹草叶肉细胞开始形成较发达的通气组织,通气组织中间隔分布有单层细胞组成的隔膜,20%和10%自然光的叶片中隔膜细胞破裂,形成了发达的通气组织;20%和10%自然光的叶片通气组织细胞面积显著大于100%和50%自然光处理组,均值分别为0.558 mm2、 0.284 mm2和0.108 mm2、 0.087 mm2.(6)20%自然光处理组根的表皮细胞中可以发现四端尖锐晶体存在,表明植物生长受到一定程度胁迫;50%自然光处理的根通气组织彼此相连,形成大的通气结构,而其它处理组皮层没有形成通气结构.综合上述研究结果表明,50%自然光处理较有利于菹草的生长发育. 相似文献
