The neuropeptides methionine enkephalin and FMRFamide, when injected into intact fiddler crabs, Uca pugilator, produce dark adaptation of the distal retinal pigment. Furthermore, both neuropeptides stimulate release of distal retinal pigment dark-adapting hormone activity from the isolated eyestalk neuroendocrine complex. It is hypothesized that both neuropeptides, when injected into intact fiddler crabs, act only indirectly on the distal retinal pigment, by stimulating release of this dark-adapting hormone.  相似文献

In investigation was conducted annually from 2002 to 2005 between the second week of May and the end of September on the incidence of scarab beetle, Schizonycha ruficollis (Fabricius) in a teak (Tectona grandis L.f.) nursery at Ramdongari, Nagpur, India. The immature stages, that is white grubs of the scarab beetle damaged 14%-52% of teak seedlings in the nursery beds, whereas the adult of the species, that is scarab beetles, were recorded on three new host trees ( Ziziphus xylopyra, Acacia catechu and A. leucophloea) in addition to the reported hosts such as ziziphus (Z. jujuba and Z. mauritiana). Beetles of the S. ruficollis started emerging from the nursery beds just after the pre-monsoon showers and continued for 10-18 days. While beetle of S. ruficollis defoliated tree hosts available in the surroundings, the immature stages of the beetle caused major damage by feeding on root systerfls of teak seedlings with symptoms of wilting and mortality in forest nursery beds. The result of chemical and biological treatments for the management of grubs of S. ruficollis in nursery beds indicated phorate 10% granular at the rate of 20 g/m^2 and chlorpyriphos 20% emulsifiable concentrate at the rate of 5.0 mL/m^2 to be effective. Grubs of S. ruficollis have not been recorded previously as a major pest of teak (T. grandis). This is also the first report on occurrence of S. ruficollis adults on tree hosts such as Z. xylopyra, A. catechu and A. leucophloea. White grubs being pests of economic importance and the toughest insects to manage, information given in this paper on incidence, pest status and management of Schizonycha ruficollis is of importance to researchers and forest nursery managers.  相似文献

Summary

The possibility that biogenic amines affect ovarian development in the red swamp crayfish, Procambarus clarkii, was investigated. Females were administered 15 μg/g body weight (bw) of norepinephrine, dopamine, 5-hydroxytryptamine (5-HT) or octopamine on days 1, 5 and 10 and were sacrificed on day 15. Crayfish given 5-HT showed significant increases in ovarian index (30.5%) and oocyte size (34.0%) over the concurrent controls, while norepinephrine, dopamine and octopamine did not significantly affect either the ovarian index or oocyte size. Significantly more labeling by 14C-leucine of ovarian proteins was found in ovaries of crayfish that were injected with 5-HT in vivo, but when ovarian lobes from crayfish that had not been injected with 5-HT were incubated in vitro with 5-HT added to the incubation medium, no significant change in the level of incorporation of 14C-leucine into ovarian proteins occurred.

The 5-HT receptor blocker LY53857 (25 μg/g bw) retarded ovarian development. The 5-HT releaser fenfluramine and the 5-HT potentiator fluoxetine (both 15 μg/g bw) were also used. Crayfish given fenfluramine, fluoxetine, fenfluramine plus 5-HT or fluoxetine plus 5-HT showed significant increases of ovarian index (24.0–102.8%), oocyte size (20.0–87.4%) and in vitro 14C-leucine labeling of ovarian proteins (30.6–123.6%) over the concurrent controls. The ovaries of crayfish that received the 5-hydroxytryptaminergic neurotoxin 5,6-dihydroxytryptamme (10 μg/g bw) did not show any significant change as compared with the initial control. These findings are consistent with the hypothesis that 5-HT, which is present in the central nervous system of Procambarus clarkii, exerts its stimulatory effect on the ovary of this crayfish indirectly by triggering release of the ovary-stimulating hormone.  相似文献

Dwarf plant height and tillering ability are two of the most important agronomic traits that determine the plant architecture, and have profound influence on grain yield in rice. To understand the molecular mechanism controlling these two traits, an EMS-induced recessive dwarf and increased tillering1 (dit1) mutant was characterized. The mutant showed proportionate reduction in each internode as compared to wild type revealing that it belonged to the category of dn-type of dwarf mutants. Besides, exogenous application of GA3 and 24-epibrassinolide, did not have any effect on the phenotype of the mutant. The gene was mapped on the long arm of chromosome 4, identified through positional candidate approach and verified by cosegregation analysis. It was found to encode carotenoid cleavage dioxygenase7 (CCD7) and identified as an allele of htd1. The mutant carried substitution of two nucleotides CC to AA in the sixth exon of the gene that resulted in substitution of serine by a stop codon in the mutant, and thus formation of a truncated protein, unlike amino acid substitution event in htd1. The new allele will facilitate further functional characterization of this gene, which may lead to unfolding of newer signalling pathways involving plant development and architecture.  相似文献

Comparison and characterization of maize stripe and maize line viruses   总被引:4,自引:0,他引:4  
Two morphologically similar viruses isolated from maize in East Africa induced two distinct symptom types in maize. One, designated maize stripe virus (MSV), showed broad yellow stripes or a yellowing of the entire leaf, acute bending of the shoot apex and severe stunting. The second, maize line virus (MLV), induced continuous, narrow yellow lines along the leaf veins and did not cause apical bending or stunting. MSV and MLV were both transmitted by Peregrinus maidis (Delphacidae), but not by Cicadulina mbila (Jassidae) or by sap inoculation. Both viruses were purified by extracting systemically infected leaves in 0–5 M sodium citrate buffer and clarifying with 7 ml n-butanol/100 ml extract, followed by differential, and finally sucrose density gradient, centrifugation. Partially purified preparations of both viruses contained isometric viruslike particles of two sizes: MSV particles were 35 and 40 nm in diameter with sedimentation coefficients (so2o, w) I09 anI0o respectively; MLV particles were 28 and 34 nm in diameter. Antisera prepared against MSV and MLV had dilution end points of 1/128 and 1/64 respectively in agar-gel diffusion tests. In tests with low-titre antisera, MSV did not react with MLV antiserum and MLV did not react with MSV antisreum; in the presence of antiserum containing antibodies to both MSV and MLV, the two viruses formed precipitin bands which crossed in the pattern of non-identity. Maize streak virus and maize mottle virus showed no serological relationship with MSV or MLV. On the basis of particle size and serology MSV and MLV are shown to be two distinct and possibly unrelated viruses. MSV and MLV apparently are dissimilar from any characterized viruses of the Gramineae.  相似文献

Sterility mosaic disease (SMD), an important biotic constraint on pigeonpea (Cajanus cajan) in the Indian subcontinent, is caused by Pigeonpea sterility mosaic virus (PPSMV) transmitted by the eriophyid mite, Aceria cajani. Distinct PPSMV isolates occur in different geographical regions and broad‐based resistance to all these isolates is scarce in cultivated pigeonpea germplasm. Wild relatives of pigeonpea, which are known to possess resistance to several pests and diseases, were evaluated for broad‐based SMD resistance. One hundred and fifteen wild Cajanus accessions from six species (C. albicans, C. platycarpus, C. cajanifolius, C. lineatus, C. scarabaeoides and C. sericeus) were evaluated against three PPSMV isolates prevailing in peninsular India. Evaluations were done under greenhouse conditions in endemic locations of each isolate through mite‐mediated virus inoculation. Fifteen accessions showed resistance to all three isolates: ICP 15614, 15615, 15626, 15684, 15688, 15700, 15701, 15725, 15734, 15736, 15737, 15740, 15924, 15925 and 15926. Most of the wild accessions did not support mite multiplication. The majority of the accessions resistant to PPSMV following inoculations with viruliferous mites were susceptible by graft inoculation, suggesting that vector resistance is conferring resistance to infection with PPSMV. The 15 accessions identified as being resistant to infection to all three virus isolates tested are cross compatible with pigeonpea by traditional breeding. They are therefore useful for exploitation in breeding programmes to increase both the level of SMD resistance and to diversify its genetic base in the cultivated pigeonpea gene pool.  相似文献

Senescence of attached and detached rooted leaves in Kleiniaarticulata can be regulated by daylength, accelerated by longphotoperiods LD (> 16 h) and retarded by short days SD (8h). Using detached, rooted leaves as stocks in leaf to leafgrafts, senescence was readily transmitted from a LD donor leafto a SD receptor leaf even if the receptor was retained in SD(D8).However, no transfer could be detected where it had to passthrough any stem tissue. Senescence was reversible up to a certain stage, beyond whichgrafts senesced, pointing to an accumulation of the graft transmissiblefactor to a threshold level that causes irreversible death. The long-day-induced senescence in Kleinia articulata leaveswas characterized by a loss in fresh and dry weight, in therate of leaf expansion and progressive loss of chlorophyll inthe detached rooted leaves. Ultrastructural examination of mesophyllcells of leaves from plants grown in continuous light showedthat osmiophilic globules accumulating in the chloroplasts werethe first visible sign of senescence in the organdies. Thesefirst signs of senescence could be detected in very young leavesof plants in continuous light, even before the leaves had expanded.Attempts were made to study the cause of this photoperiodicsenescence which, from previous work, appeared to involve agraft-transmissible substance. Leaves in continuous light showed reduced stomatal opening andextracts from them had very much higher activity in the Commelinastomatal closure assay (ABA-like activity ?) compared with non-senescingleaves grown in short days (8 h). However, even if all the activitywere due to ABA, this on its own does not appear to be the senescencesubstance because a much longer exposure to continuous lightwas required to induce irreversible senescence than to reachmaximum stomatal closure promoting activity in the bioassay.Moreover, severe water stress (high ABA?) did not lead to senescenceunless combined with continuous light or ethylene treatment.It is postulated that while ABA may play an important role inKleinia leaf senescence its lethal effect may not be realizedunless ethylene-induced membrane changes may synergisticallyassist. In a comparison of six cowpea cultivars, we determined the variation in abscisic acid (ABA) production as an ‘early warning signal’ produced in response to drought stress. By imposing drought only to the upper 20 cm rooting zone, we compared the rates of ABA synthesis relative to (i) total root mass and (ii) inherent variation per unit root mass. We were able to relate the intensity of the stress response to these two factors, and determine which is quantitatively more important as the primary signal indicating responsiveness to drought stress. Plants were grown in 1.2 m long columns and a soil drying treatment imposed in such a way that that upper roots were in dry soil and deep roots in soil at field capacity. Relative water contents (RWC) of stressed plants were similar and not significantly different from those of well watered controls. However, roots accumulated ABA in the dehydrated zone, where root water content ranged from 10–12 g g?1 DW. The soil moisture contents and root ‐water contents in the dry zone were similar for each of the different varieties. However, the ABA contents were significantly different in drought‐stressed (upper) roots and ranged from 7.82 nmol g?1 DW in cv. APC 689 to 16.02 nmol g?1 DW in cv. APC 370, such that for varieties with similar overall root weights (e.g. APC 580 and APC 540) the different ABA contents were related to the capacity for ABA synthesis. The relationship between stomatal conductance and total root ABA was assessed, with a negative relation (r= 0.90, n= 24, P= 0.05) suggesting that the intrinsic capacity of cowpea varieties for ABA synthesis could play an important role in regulating stomatal conductance in a drying soil and provide useful selection criteria for tolerance to drought stress.  相似文献

Graft transmission of the floral stimulus was studied in homograftsof Kleinia articulata (SDP) and heterografts between K. articulataand K. repens (LSDP). While receptor shoots of K. repens graftedonto induced donor plants of K. articulata flowered readilyin LD (16 h) as well as SD (8 h), graft-induced flowering failedto take place in LD receptors of K. articulata. Neither theinduced shoots and detached leaves from induced plants of K.articulata nor the induced shoots of K. repens could evoke theflowering response in the K. articulata receptors. Increasingthe donor pool of induced leaves even up to ten per receptoralso had no effect. It is known that the very young leaf primordiaof K. articulata are photoperiodically sensitive, and it seemsthat they may prevent the stimulus from reaching the apex.
