Pea leaf asparaginase is stabilized by asparagine and aspartateduring incubation. In crude extracts this effect was enhancedby products of the light reaction (NADPH, NADH, or reduced ferredoxin),but these compounds were ineffective on the purified enzyme,or in the absence of asparagine. MgATP, MgADP and oxidized ferredoxinreduced asparaginase activity in purified preparation reducedor oxidized glutathione had no effect. Asparaginase activitydoes not appear to be modulated via phosphorylation/dephosphorylation.The presence of calcium during extraction increased asparaginaseactivity more than 2-fold, but addition of calcium to extractsprepared in its absence had no effect; calmodulin had no effecton activity. Co-extraction of light- and dark-treated tissueshowed that soluble factors are not responsible for the diurnalvariation in asparaginase activity. Association of asparaginasewith membranes did not account for changes in extractable activity.Use of the protein synthesis inhibitors cycloheximide, puromycin,emetine, actinomycin D and cordycepin and the thiol proteaseinhibitor leupeptin suggested that mRNA and protein synthesisare required for the increase of asparaginase activity duringthe light period and that proteolytic degradation accounts forthe decrease during the dark. During the growth of leaves of Pisum sativum L., levels of asparaginase(E.C. 3.5.1.1 [EC] ) showed a diurnal variation during a 3 d periodof leaf expansion, increasing in the light and decreasing inthe dark period; the greatest diurnal variation being foundin half-expanded leaves. Asparaginase activity in half-expandedleaves reached a maximum after 4 h exposure to light and thisactivity was maintained over the rest of the light period. Changesin asparaginase activity were not influenced by diurnal temperaturechanges. The increase in asparaginase activity during the lightperiod was directly proportional to the photon flux densityover the range 0–285 µmol m-2 s-1 PAR. The increaseof asparaginase activity during illumination of detached leaveswas inhibited by the photosynthetic electron transport inhibitors3-(3', 4'-dichlorophenyl)-1, 1-dimethylurea (DCMU) and atrazine.These observations indicate that the increase in asparaginaseactivity in half-expanded leaves is dependent upon non-cyclicelectron transport. The growth rate of Tetrahymena setosa cells is stimulated significantly by as little as 0.1 μg and optimally by about 1 μg of ergosterol per ml of medium. Cell yields in the stationary phase are, however, not perceptibly affected by increasing sterol concentrations. Ergosterol, in concentrations that stimulate growth optimally, does not cause a reduction of tetrahymanol synthesis. The latter process is impaired only at much higher ergosterol concentrations. Epicholesterol and coprostanol inhibit ergosterol-stimulated growth competitively. It is concluded that the trace amounts of sterol needed by T. setosa do not serve to replace tetrahymanol but function in some other manner, probably unrelated to the control of membrane fluidity. This conclusion supports the views advanced earlier by Holz, Erwin, Wagner & Rosenbaum.  相似文献

The Oriental butterfly genus Arhopala is by far the most species‐rich genus within the subfamily Theclinae. We investigated whether biotic interactions with larval host plants or ants are phylogenetically constrained in the evolutionary history of Arhopala, by collating available information on the ecology of Arhopala from the literature as well as from personal observations, and analysing and interpreting these data rigorously in a phylogenetic context. Larvae of all species in the sister‐group of Arhopala and Flos feed on Fabaceae. However, the predominant feeding preference of Arhopala s.l., with the exception of a particular monophyletic and species‐rich group, called ‘Group A’, appears to be centred on Fagaceae, with additional records of Dipterocarpaceae feeding. The preference for Fagaceae has strong phylogenetic inertia, as indicated by T‐PTP tests designed to test for phylogenetic signal. Adding all available life‐history data, using the phylogeny presented before as scaffolding, further increased the phylogenetic signal in host plant data. Feeding on Fabaceae (mainly in the outgroup) and Euphorbiaceae also showed significant phylogenetic signal, but feeding on Dipterocarpaceae did not and was found scattered across the phylogeny. Except for the Dipterocarpaceae, phylogenetic signal in feeding preferences was very clear, even despite uneven taxon sampling and apparent lability in host plant use. Ant association also demonstrated historical constraint, but based on the phylogenetic hypothesis presented here it was not clear whether increased ant association intimacy emerged independently various times in Group A, as well as in several basal groups. Our finding of distinct phylogenetic patterns in the host plant and to a lesser degree in ant association data contradicts the hypothesis that life‐history traits are very labile in the Lycaenidae. © 2005 The Linnean Society of London, Biological Journal of the Linnean Society, 2005, 84 , 225–241.  相似文献

THERE is growing evidence to indicate that RNA synthesis in bacteria is regulated through adjustment of the frequency of initiation of new RNA molecules. In this framework an understanding of the process of initiation of RNA synthesis takes on a special importance and for this reason we have investigated in varying conditions the composition of the 5′ terminal or first-inserted nucleotide. It has been previously shown that such initiations do not occur randomly, either in vivo1 or in the proper conditions in vitro2,3, but that RNA chains are exclusively initiated with the purines, adenosine and guanosine. Additionally, there was a recent suggestion based on in vitro studies that the nucleotide guanosine-3′-diphosphate-5′-diphosphate (MS1), proposed to be a regulatory agent in RNA synthesis, functioned by specifically depressing the frequency of initiation of a large fraction of RNA molecules beginning with guanosine4. Here we report, however, that in vivo, in conditions in which regulation of RNA synthesis is manifest, the ratio of molecules initiated with adenosine and guanosine is not changed.  相似文献

Djinga is a monotypic genus restricted to the Cameroon Ridge (‘Dorsale Camerounaise’) of NW Cameroon. Besides the type locality Mount Djinga (Adamawa Province, near Tignère), it also grows in waterfalls near Mount Oku (NW Province). This paper describes the structure and development of Djinga felicis using scanning electron microscopy and microtome sections. Cusset's protologue is enriched considerably. Roots are green, dorsiventrally flattened and adherent to submerged rocks. They are broad ribbons or crusts (up to 1 cm broad) which branch exogenously. Root‐borne shoots (up to 17 cm) have filamentous leaves with sheaths and (occasionally) attached stipules. Flowers are borne as part of reproductive short shoots which arise exogenously along the stems and endogenously along the roots. Djinga shows non‐axillary stem branching. Reproductive shoots along the main stem usually stay short. They arise from the abaxial side of double‐sheathed leaves, a key innovation of Podostemoideae. These short shoots shift into pocket‐like stem positions clearly below their associated leaves. Each flower bud inside a spathella is erect, but with an inclined ovary. The flowers are unistaminate with a broad connective. Unlike many other Podostemoideae, the tricolpate pollen grains are arranged in both dyads and monads. The ovary is globular and unilocular.   • 1 Caterpillars of the facultatively myrmecophilous butterfly Polyommatus icarus were reared on inflorescences, or foliage, of four natural hostplant species and on an artificial diet to study dietary effects on larval growth and secretory capacity.
  • 2 Caterpillars achieved highest weights and relative growth rates when fed flowers of Medicago sativa, Lotus corniculatus or Melilotus officinalis. Larvae reared on Coroniüa varia (flowers and leaves), foliage of M.sativa and on the artificial diet pupated at lower weights and achieved lower growth rates.
  • 3 In standardized experiments with the ant species Lasius flavus, secretion rates from the dorsal nectar organ (DNO) were 2 times higher among flower-fed caterpillars than among foliage-fed siblings or caterpillars on the artificial diet. Larvae reared on C.varia flowers were superior to all other food treatments with respect to secretion rates.
  • 4 High water content of larval diet, as in flowers, appears to be important for lycaenid caterpillars to achieve high secretion rates, whereas the correlation between myrmecophily and nutrient availability, as evidenced by growth rates, was less pronounced.
  • 5 Using experimental data on larval growth and secretion rates, the lifetime volume of secretions from the DNO is estimated to range from 2 to 5 μl in most food treatments. Only on C.varia flowers (5.5–8.7 μl) and on M.sativa leaves (0.9–1.1 μl) did the caterpillars deviate in their absolute investment in myrmecophily.

In experiments with intact tomato plants the effect of potassium on the translocation of photosynthates was studied. Four series of plants were grown in nutrient solutions with two different potassium levels (K1 and K2 treatments). When the fruits of the first truss had developed the plants were treated with labelled CO2 for 2 h in a glove box. The plants were then divided in roots, stems, fruits and leaves, which were prepared for chemical analysis. In all experimental series the plants with the higher K supply grew better and therefore assimilated higher amounts of labelled CO2 (cpm/plant). The assimilation rates per g fresh material, however, were not increased in each experimental set by the K2 treatment. The absolute quantities of labelled photosynthates transported from leaves and stems to the fruits were also greater in the plant with the higher potassium supply. In all experimental series the proportion of labelled material found in the fruits was higher in the plants of the K2 treatment (total label of the plant=100%) than in the plants of the K1 treatment. This beneficial effect of potassium was also observed in cases, where potassium had not affected the CO2 assimilation rate. Expressing the distribution of labelled material in relative terms (percentage proportions) means that the effect of CO2 assimilation on the transport of photosynthates was excluded to a high extent. The higher percentage proportions of labelled material found in fruits and roots of the K2 as compared to the K1 treatment is, therefore, predominantly dependent on a specific effect of potassium on the translocation of assimilates. The labelled material found in the soluble fraction of the stems consisted of about 90% sugars, nearly half of which was sucrose, the rest being mainly glucose and fructose. This finding suggests that sucrose is the most important transport form of carbohydrates in tomatoes.  相似文献

Adult life spans of insects are part of supposedly adaptive life-history syndromes, which involve a variety of reproductive strategies with regard to mating, egg maturation and egg laying. Contingencies of life span with many other morphological, behavioural, and ecological traits are to be expected. We used a data-mining approach, incorporating controls for phylogeny, to uncover broad patterns of trait contingencies in a literature-derived data set of adult life spans for 350 butterfly species. We found that adult feeding habit has strong links with life spans, with pollen-feeders living longer than fruit feeders, which in turn live longer than nectar feeders. Furthermore, traits reducing susceptibility to predators (i.e. aposematism, eye-spots) are associated with longer life. Mud-puddling behaviour as a means of gaining access to minerals, and possibly nitrogen, was generally associated with a short life. We also found strong impacts of the region of occurrence of species (i.e. whether they were tropical or temperate taxa), but received conflicting results depending on whether we analysed field data (longer life in the tropics) or data from caged individuals (shorter life in the tropics). Adult longevity was associated with the growth form of larval host plants (higher on lianas as opposed to herbs, shrubs, or trees), whereas other larval host plant traits did not have strong links with species' life spans. It has been hypothesized that the subalpine Erinus alpinus survived the Pleistocene glaciation in southern French refugia peripheral to the Alps and postglacially immigrated to its northern Alpine distribution (tabula rasa hypothesis). However, E. alpinus is also known from nunataks in the northern Prealps. PCR–RFLP of the cpDNA revealed no variation, whereas AFLPs identified three phylogeographical groups among the 22 populations: (1) a central Swiss group, (2) a single central Swiss population on Mount Rigi, and (3) all other populations located to the west and east of the central Swiss group (west-eastern populations). The population representing the putative French refugium was not distinct from the west-eastern populations but was different from the populations of central Switzerland. We consider this support for locally restricted in situ survival of E. alpinus in the northern Prealps or nearby Jura mountains (dating back to 60–28 ky BP). However, the west-eastern gene-pool probably immigrated postglacially from southern France through the prealpine lowlands (not before 14.6 kyr BP), skirting central Switzerland.
