Single- (whole-cell patch) and two-electrode voltage-clamp techniques were used to measure transient (Ifast) and sustained (Islow) calcium currents, linear capacitance, and slow, voltage-dependent charge movements in freshly dissociated fibers of the flexor digitorum brevis (FDB) muscle of rats of various postnatal ages. Peak Ifast was largest in FDB fibers of neonatal (1-5 d) rats, having a magnitude in 10 mM external Ca of 1.4 +/- 0.9 pA/pF (mean +/- SD; current normalized by linear fiber capacitance). Peak Ifast was smaller in FDB fibers of older animals, and by approximately 3 wk postnatal, it was so small as to be unmeasurable. By contrast, the magnitudes of Islow and charge movement increased substantially during postnatal development. Peak Islow was 3.6 +/- 2.5 pA/pF in FDB fibers of 1-5-d rats and increased to 16.4 +/- 6.5 pA/pF in 45-50-d-old rats; for these same two age groups, Qmax, the total mobile charge measurable as charge movement, was 6.0 +/- 1.7 and 23.8 +/- 4.0 nC/microF, respectively. As both Islow and charge movement are thought to arise in the transverse-tubular system, linear capacitance normalized by the area of fiber surface was determined as an indirect measure of the membrane area of the t-system relative to that of the fiber surface. This parameter increased from 1.5 +/- 0.2 microF/cm2 in 2-d fibers to 2.9 +/- 0.4 microF/cm2 in 44-d fibers. The increases in peak Islow, Qmax, and normalized linear capacitance all had similar time courses. Although the function of Islow is unknown, the substantial postnatal increase in its magnitude suggests that it plays an important role in the physiology of skeletal muscle.  相似文献

We have analyzed nucleic acid and amino acid sequence alignments of a variety of voltage-sensitive ion channels, using several methods for phylogenetic tree reconstruction. Ancient duplications within this family gave rise to three distantly related groups, one consisting of the Na+ and Ca++ channels, another the K+ channels, and a third including the cyclic nucleotide-binding channels. A series of gene duplications produced at least seven mammalian homologues of the Drosophila Shaker K+ channel; clones of only three of these genes are available from all three mammalian species examined (mouse, rat, and human), pointing to specific genes that have yet to be recovered in one or another of these species. The Shaw-related K+ channels and the Na+ channel family have also undergone considerable expansion in mammals, relative to flies. These expansions presumably reflect the needs of the high degree of physiological and neuronal complexity of mammals. Analysis of the separate domains of the four-domain channels (Ca++ and Na+) supports their having evolved by two sequential gene duplications and implies the historical existence of a functional two-domain channel.   相似文献

Background

Severe acute malnutrition (SAM) is the most serious form of malnutrition affecting children under-five and is associated with many infectious diseases including Tuberculosis (TB). In India, nutritional rehabilitation centres (NRCs) have been recently established for the management of SAM including TB. The National TB Programme (NTP) in India has introduced a revised algorithm for diagnosing paediatric TB. We aimed to examine whether NRCs adhered to these guidelines in diagnosing TB among SAM children.

Methods

A cross-sectional study involving review of records of all SAM children identified by health workers during 2012 in six tehsils (sub-districts) with NRCs (population: 1.8 million) of Karnataka, India.

Results

Of 1927 identified SAM children, 1632 (85%) reached NRCs. Of them, 1173 (72%) were evaluated for TB and 19(2%) were diagnosed as TB. Of 1173, diagnostic algorithm was followed in 460 (37%). Among remaining 763 not evaluated as per algorithm, tuberculin skin test alone was conducted in 307 (41%), chest radiography alone in 99 (13%) and no investigations in 337 (45%). The yield of TB was higher among children evaluated as per algorithm (4%) as compared to those who were not (0.3%) (OR: 15.3 [95%CI: 3.5-66.3]). Several operational challenges including non-availability of a full-time paediatrician, non-functioning X-ray machine due to frequent power cuts, use of tuberculin with suboptimal strength and difficulties in adhering to a complex diagnostic algorithm were observed.

Conclusion

This study showed that TB screening in NRCs was sub-optimal in Karnataka. Some children did not reach the NRC, while many of those who did were either not or sub-optimally evaluated for TB. This study pointed to a number of operational issues that need to be addressed if this collaborative strategy is to identify more TB cases amongst malnourished children in India.  相似文献

The ratio of fore- to hindlimb size plays an important role in our understanding of human evolution. Although Homo habilis was relatively modern craniodentally, its body proportions are commonly believed to have been more apelike than in the earlier Australopithecus afarensis. The evidence for this, however, rests, on two fragmentary skeletons, OH 62 and KNM-ER 3735. The upper limb of the better-preserved OH 62 from Olduvai Gorge is long and slender, but its hindlimb is represented mainly by the proximal portion of a thin femur of uncertain length. The present analysis shows that upper-to-lower limb shaft proportions of both OH 62 and AL 288-1 (A. afarensis) fall in the modern human range of variation, although OH 62 also falls inside that of chimpanzees due to their overlap in small individuals. Despite being more fragmentary, the larger-bodied KNM-ER 3735 lies outside the chimpanzee range and close to the human mean. Because the differences between any of the three individuals are compatible with the range of variation seen in extant hominoid groups, it is not legitimate to infer more primitive upper-to-lower limb shaft proportions for either H. habilis or A. afarensis. Femur length of OH 62 can only be estimated by comparison. Its closest match in size and morphology is with the gracile OH 34 specimen, which therefore provides a better analogue for the reconstruction of OH 62 than the stocky AL 288-1 femur that is traditionally used. OH 34's slender proportions are hardly due to abrasion, but match those of a modern human of that body-size, suggesting that the relative length of OH 62's leg may have been human-like. Brachial proportions, however, remained primitive. Long legs may imply long distance terrestrial travel. Perhaps this adaptation evolved early in the genus Homo, with H. habilis providing an early representative of this important change.  相似文献

General doctrine holds that early hominids possessed a long lumbar spine with six segments. This is mainly based on Robinson's (1972) interpretation of a single partial Australopithecus africanus skeleton, Sts 14, from Sterkfontein, South Africa. As its sixth last presacral vertebra exhibits both thoracic and lumbar characteristics, current definitions of lumbar vertebrae and lumbar ribs are discussed in the present study. A re-analysis of its entire preserved vertebral column and comparison with Stw 431, another partial A. africanus skeleton from Sterkfontein, and the Homo erectus skeleton KNM-WT 15000 from Nariokotome, Kenya, did not provide strong evidence for the presence of six lumbar vertebrae in either of these early hominids. Thus, in Sts 14 the sixth last presacral vertebra has on one side a movable rib. In Stw 431, the corresponding vertebra shows indications for a rib facet. In KNM-WT, 15000 the same element is very fragmentary, but the neighbouring vertebrae do not support the view that it is L1. Although in all three fossils the transitional vertebra at which the articular facets change orientation seems to be at Th11, this is equal to a large percentage of modern humans. Indeed, a modal number of five lumbar vertebrae, as in modern humans, is more compatible with evolutionary principles. For example, six lumbar vertebrae would require repetitive shortening and lengthening not only of the lumbar, but also of the entire precaudal spine. Furthermore, six lumbar vertebrae are claimed to be biomechanically advantageous for early hominid bipedalism, yet an explanation is lacking as to why the lumbar region should have shortened in later humans. All this raises doubts about previous conclusions for the presence of six lumbar vertebrae in early hominids. The most parsimonious explanation is that they did not differ from modern humans in the segmentation of the vertebral column.  相似文献

Upper-to-lower limb proportions of Homo habilis are often said to be more ape-like than those of its reputed ancestor, Australopithecus afarensis. Such proportions would either imply multiple evolutionary reversals or parallel development of a relatively short upper limb in A. afarensis and later Homo. However, assessments of limb proportions are complicated by the fragmentary nature of the two known H. habilis skeletons, OH 62 and KNM-ER 3735. Initially, KNM-ER 3735 was compared to A.L. 288-1 (A. afarensis) using a single modern human and chimpanzee as reference. Here, based on a larger comparative sample, we find that the relative size of the distal humerus, radial head, and shaft of both KNM-ER 3735 and A.L. 288-1 lie within the range of variation of modern humans, whereas their sacra are small as is the case for all early hominids. In addition, their manual phalanges are similar in having a gracile base but robust midshaft. Contrary to earlier studies, the fossils are not differentiable from each other statistically with respect to all features listed above. On the other hand, they differ in robusticity of the scapular spine and relative length of the radial neck. An exact randomization test suggests only a very low probability of finding a similar degree of difference within a single species of extant hominoids. In contrast to the consensus view, we conclude that A.L. 288-1 had a short, human-like forearm, whereas KNM-ER 3735 possessed a distinctly longer forearm and more powerful shoulder girdle. This interpretation fits with earlier conclusions that suggested human-like humerofemoral proportions but chimpanzee-like brachial proportions for Homo habilis. Thus, the scenario of a unidirectional, progressive change in limb proportions within the hominid lineage is not supported by our work.  相似文献

The association of type 2 diabetes with elevated plasma triglyceride (TG) and very low-density lipoproteins (VLDL), and intrahepatic lipid accumulation represents a pathophysiological enigma and an unmet therapeutic challenge. Here, we uncover a link between insulin action through FoxO1, bile acid (BA) composition, and altered lipid homeostasis that brings new insight to this longstanding conundrum. FoxO1 ablation brings about two signature lipid abnormalities of diabetes and the metabolic syndrome, elevated liver and plasma TG. These changes are associated with deficiency of 12α-hydroxylated BAs and their synthetic enzyme, Cyp8b1, that hinders the TG-lowering effects of the BA receptor, Fxr. Accordingly, pharmacological activation of Fxr with GW4064 overcomes the BA imbalance, restoring hepatic and plasma TG levels of FoxO1-deficient mice to normal levels. We propose that generation of 12α-hydroxylated products of BA metabolism represents a signaling mechanism linking hepatic lipid abnormalities with type 2 diabetes, and a treatment target for this condition.  相似文献
