Collagen fibrils from the mesenteric connective sheath of the adult cockroach Periplaneta americana were extracted by enzymatic digestion with pepsin and were purified. Chromatographic studies and sodium dodecylsulfate electrophoresis revealed the presence of a single chain. It was demonstrated that the structure of this collagen could be represented by the formula (alpha)3. The amino acid composition is typical of collagens (one-third glycine, and a high imino acid content) and similar to that of type II. The carbohydrate content was high (8.8%), and the cyanogen bromide pattern was different from that of known collagens. The chains were linked by the stable intermolecular bond dihydroxylysinonorleucine. The banding patterns of the segment-long-spacing crystallites and of the reconstituted fibrils were similar to type I collagen. The molecular weight (Mr 280,000) and length (285 nm) were typical, but the denaturation temperature was high (38.5 degrees C). It was concluded that cockroach mesenteric collagen showed the characteristic features of invertebrate mesodermal collagens, except that of the thermal stability of the triple-helical structure.  相似文献

Purpose

Since the implementation of the European directive (EC/2001/42) on strategic environmental assessment, an ex ante evaluation has become mandatory for plans and programs. This requirement could have significant consequences for the environment. Local authorities, who are in charge of land planning issues, must therefore conduct such assessments. However, they are faced with lack of uniform methodology. The aim of this paper is thus to propose a methodological framework for the required environmental assessment stages in land planning.

Methods

Life cycle assessment (LCA) has been identified as a promising tool to perform environmental assessment at a meso-level (i.e., territories). Yet, the standardized LCA framework has never been used for assessing the environmental impacts of a territory as such, which can be explained by the complexity that its application would involve. Four major methodological bottlenecks have been identified in this paper, i.e., (1) functional unit definition, (2) boundary selection, (3) data collecting, and (4) the refinement of the life cycle impact assessment phase in order to provide useful indicators for land planning. For each of these challenges, recommendations have been made to adapt the analytical framework of LCA.

Results and discussion

A revised framework is proposed to perform LCA of a territory. One of the major adaptations needed concerns the goal and scope definition phase. Henceforth, the association of a territory and the studied land planning scenario, defined by its geographical boundaries and its interactions with other territories, will be designated as the reference flow in LCA. Consequently, two kinds of indicators will be determined using this approach, i.e., (1) a vector of environmental impacts generated (conventional LCA) and (2) a vector of land use functions provided by the territory for different stakeholders (e.g., provision of work, recreation, culture, etc.). This revised framework has been applied to a theoretical case study in order to highlight its utility in land planning.

Conclusions

This work is a first step in the adaptation of the LCA framework to environmental assessment in land planning. We believe that this revised framework has the potential to provide relevant information in decision-making processes. Nonetheless, further work is still needed to broaden and deepen this approach (i.e., normalization of impacts and functions, coupled application with GIS, uncertainties, etc.).  相似文献

Methane can be produced by anaerobic digestion. The Biochemical Methane Potential (BMP) test is widely applied to determine the anaerobic biodegradability of wastes. It is based on a fermentation process, which is time consuming, about 30 days. This study investigates the use of near infrared spectroscopy to predict the Biochemical Methane Potential value of municipal solid waste. Near infrared spectroscopy has the advantage to be very fast and applicable to solid waste with a light sample preparation. Satisfying results were obtained: R(2)=0.76; Standard Error of Prediction=28 ml CH(4) g(-1) VS, that compare very favourably with reported results for other more expensive and more time-consuming methods. To our knowledge, it is the first time near infrared spectroscopy is used to predict the Biochemical Methane Potential value. Using near infrared spectroscopy for waste management would thus lead to a real benefit from an industrial point of view.  相似文献

Lewis lung carcinoma cells from tumors, metastasis nodules, or from culture bind fluorescent derivatives of neoglycoproteins containing α-D -glucose residues: This binding is competitively inhibited by neoglycoproteins containing α-D -glucose, by mannan, and by several other neoglycoproteins. Cell binding and uptake of the fluorescent derivatives of the neoglycoproteins was quantified by lysing the cells with an alkylpolyol (MAC 19 or MAC 18) and measuring the fluorescence intensity of the supernatant. The amount of cell-associated neoglycoprotein was higher at 37°C than at 4°C with LLC from tumor. The binding and uptake were inhibited by glycoconjugates containing α-D -glucose. These results suggest the presence of sugar specific receptors in Lewis lung carcinoma cells which are involved in a sugar-specific binding and endocytosis phenomenon. The implication of the existence of a carbohydrate-binding protein on the surface of Lewis lung carcinoma cells are discussed with regard to the in vivo behaviour of these cells, especially in relation to their metastatic properties and to the possibility of using neoglycoproteins as specific carriers of cytotoxic drugs. Hybrid molecules of gelonin and a neoglycoprotein containing α-D -glucose were used as targetted toxin: The targetted toxin was found to bind to and to enter the intact cells and was 100 times more toxic than free drug.  相似文献

In prokaryotes, transglutaminase (TGase) has been found only in actinomycetes from the genus Streptoverticillium. The role of this TGase, as well as the mechanism regulating the enzyme expression, are still unknown. In order to improve TGase production by Streptoverticillium cinnamoneum CBS 683.68 and simultaneously elucidate the relationship between growth and TGase activity, we decided to study these two responses using different designs of statistical analysis. Among the five factors tested, casein, glycerol, peptones, yeast extract and oligoelements, only oligoelements were found to have no effect either on growth or on TGase production in a complete factorial design. The two factors casein and glycerol were found to have a highly significant effect on both dry weights and TGase activity in a Box-Behnken design used to improve the model. Finally, the TGase activity was increased three times to reach 0.331±0.038 U/ml with optimum concentrations of casein (38.4 g/l) and glycerol (31.2 g/l) calculated with the help of a composite design. In the course of these experiments, the two responses varied in the same way, demonstrating that growth and TGase production were tightly correlated under the conditions described. However, TGase was produced during the stationary phase of growth in optimized medium, indicating that the enzyme production could be induced.
