Spinosyns A and D are the active ingredients in an insect control agent produced by fermentation of Saccharopolyspora spinosa. Spinosyns are macrolides with a 21-carbon, tetracyclic lactone backbone to which the deoxysugars forosamine and tri-O-methylrhamnose are attached. The spinosyn biosynthesis genes, except for the rhamnose genes, are located in a cluster that spans 74 kb of the S. spinosa genome. DNA sequence analysis, targeted gene disruptions and bioconversion studies identified five large genes encoding type I polyketide synthase subunits, and 14 genes involved in sugar biosynthesis, sugar attachment to the polyketide or cross-bridging of the polyketide. Four rhamnose biosynthetic genes, two of which are also necessary for forosamine biosynthesis, are located outside the spinosyn gene cluster. Duplication of the spinosyn genes linked to the polyketide synthase genes stimulated the final step in the biosynthesis — the conversion of the forosamine-less pseudoaglycones to endproducts. Duplication of genes involved in the early steps of deoxysugar biosynthesis increased spinosyn yield significantly. Journal of Industrial Microbiology & Biotechnology (2001) 27, 399–402. Bidens cordylocarpa is a high polyploid species restricted in distribution to stream sides in the mountains of Jalisco, Mexico. The morphologically enigmatic species was originally described as a member of the genus Coreopsis, but later transferred to Bidens, largely because the involucral bracts appear most similar to Bidens. Characters of the cypselae, often useful in generic placement, are of no value for this species because the fruits have features not detected in either Bidens or Coreopsis. Sequences from the internal transcribed spacer region of nuclear ribosomal DNA (ITS) were used to assess the relationships of Bidens cordylocarpa. The molecular phylogeny places B. cordylocarpa in a strongly supported clade of Mexican and South American Bidens, and provides more definitive evidence of relationships than morphology, chromosome number, or secondary chemistry. Molecular, morphological, and chromosomal data suggest that B. cordylocarpa is an ancient polyploid, perhaps the remnant of a polyploid complex. The purpose of this review is to provide a global perspective on Oncorhynchus salmonine introductions and put-and-take fisheries based on modern stocking programs, with special emphasis on freshwater ecosystems. We survey the global introductions of nine selected salmonines of the genus Oncorhynchus: golden trout, cutthroat trout, pink salmon, chum salmon, coho salmon, masu/cherry salmon, rainbow trout/steelhead, sockeye salmon/kokanee, and chinook salmon. The information is organized on a geographical basis by continent, and then by species and chronology. Two different objectives and associated definitions of ‘success’ for introductions are distinguished: (a) seed introduction: release of individuals with the purpose of creating a wild-reproducing, self-sustaining population; and (b) put-and-take introduction: release of individuals with the purpose of maintaining some level of wild population abundance, regardless of wild reproduction. We identify four major phenomena regarding global salmonine introductions: (1) general inadequacy of documentation regarding introductions; (2) a fundamental disconnect between management actions and ecological consequences of introductions; (3) the importance of global climate change on success of previous and future introductions; and (4) the significance of aquaculture as a key uncertainty in accidental introductions. We conclude this review with a recognition of the need to terminate ongoing stocking programs for introduced salmonines worldwide.  相似文献

Contrary to our expectations, soil salinity and moisture explained little of the spatial variation in plant establishment in the upper intertidal marsh of three southern California wetlands, but did explain the timing of germination. Seedlings of 27 species were identified in 1996 and 1997. The seedlings were abundant (maximum densities of 2143/m2 in 1996 and 1819/m2 in 1997) and predominantly annual species. CCAs quantified the spatial variation in seedling density that could be explained by three groups of predictor variables: (1) perennial plant cover, elevation and soil texture (16% of variation), (2) wetland identity (14% of variation) and (3) surface soil salinity and moisture (2% of variation). Increasing the spatial scale of analysis changed the variables that best predicted patterns of species densities. Timing of germination depended on surface soil salinity and, to a lesser extent, soil moisture. Germination occurred after salinity had dropped below a threshold or, in some cases, after moisture had increased above a critical level. Between 32% and 92% of the seedlings were exotic and most of these occurred at lower soil salinity than native species. However, Parapholis incurva and Mesembryanthemum nodiflorum were found in the same environments as the native species. In 1997, the year of a strong El Niño/Southern Oscillation event with high rainfall and sea levels, the elevation distribution of species narrowed and densities of P. incurva and other exotic species decreased but densities of native and rare species did not change. The ‘regeneration niche’ of wetland plant communities includes the effects of multiple abiotic and biotic factors on both the spatial and temporal variations in plant establishment.  相似文献

The mechanism of IP3-induced activation of saponin-permeabilised platelets has been examined. Saponin permeabilization resulted in the leakage of low-Mr substances into and from the cells without loss of cytoplasmic proteins. Addition of IP3 rapidly induced a dose-related formation of thromboxane B2 and release into the medium, leading to the responses of shape change, aggregation and [14C]5HT release. These responses were inhibited by the thromboxane A2 receptor antagonist AH23848. The IP3-induced release of 45Ca from intracellular stores was not affected by indomethacin. Synthesis of thromboxane was inhibited if Ca2+ elevation was prevented by using Ca-EGTA buffers during permeabilization. These studies indicate that IP3-induced activation was due to Ca2+ mobilisation leading to phospholipase activation and thromboxane synthesis.  相似文献

This study documents odontometric variation in four populations of related genetic background. It is found that the transplanted populations of Cuanalan and Saltillo have undergone significant microdifferentiation in tooth size relative to the two home valley populations. The extent and direction of this microdifferentiation is seen as reflecting differential amounts of admixture from African and European sources in demographic subunits of the two transplanted populations. The dental variables found to discriminate significantly between populations are the same variables predicted to be evolutionarily labile by Butler's Field Theory. Principal component analysis confirms the presence of morphological fields in these data and supports the position that each class has a stable tooth, with stability decreasing with increasing distance from the key tooth. The importance of hybridization in determining which variables significantly discriminate between populations is confounded by the effects of development of the dentition in morphogenetic fields. These data suggest the odontometric data provide an adequate, if somewhat conservative, base from which to evaluate microdifferentiation of human populations.  相似文献
