Shallow, near-shore water habitats on the continental shelf of the Northeast Atlantic have been productive fishing areas in the past. Here, we review the present knowledge about (i) recent trends in the abundance of plaice and cod in these habitats and (ii) hypotheses regarding the factors responsible for any trends. At present, only a few studies exist on the trends of abundance of plaice or cod, namely from the Bay of Biscay, the North Sea and the Skagerrak/Kattegat. They suggest a declining abundance in coastal, shallow areas and – at least for plaice – a latitudinal gradient with an erosion of the southern distribution boundary in the Bay of Biscay and deepening of stocks in the North Sea. In contrast, no trend in shallow water abundance of plaice similar to a decline in deep-water stocks during the 1970s and their slow recovery during the 2000s is apparent in the Skagerrak/Kattegat. Although shallow habitats fundamentally differ from deeper areas by the prevalence of juvenile stages, the declining trends coincide with decreasing abundance/landings and spatial stock relocations in the deeper areas. Whether this indicates a common trend pointing at connectivity between shallow and deep water remains open. Fundamental differences exist in the suggested causes of the trends in different geographical areas. High fishing pressure together with low local recruitment apparently prevents the recovery of overexploited plaice and cod stocks in the Skagerrak/Kattegat. In contrast, the responses of juveniles and adult fish to increasing seawater temperature are the main hypotheses for changes in distribution and abundance of both fish species in the North Sea/Bay of Biscay. However, temperature alone cannot explain the observed decline of fish in coastal areas, and the causes may be more complex, involving nutrient loading, primary productivity or food availability, although at present, knowledge of these factors is insufficient.  相似文献

Experimental cerebral malaria (ECM) resulting from Plasmodium berghei ANKA infection involves T lymphocytes. However, the mechanisms of T cell-mediated pathogenesis remain unknown. We found that, in contrast to ECM-susceptible C57BL6 mice, perforin-deficient (PFP-KO) mice were resistant to ECM in the absence of brain lesions, whereas cytoadherence of parasitized erythrocytes and massive accumulation of activated/effector CD8 lymphocytes were observed in both groups of mice. ECM is induced in PFP-KO mice after adoptive transfer of cytotoxic CD8+ cells from infected C57BL6 mice, which were directed to the brain of PFP-KO mice. This specific recruitment might involve chemokine/chemokine receptors, since their expression was up-regulated on activated CD8 cells, and susceptibility to ECM was delayed in CCR5-KO mice. Thus, lymphocyte cytotoxicity and cell trafficking are key players in ECM pathogenesis.  相似文献

In nature adult insects, such as parasitic wasps or 'parasitoids' often depend on supplemental nutritional sources, such as sugars and other carbohydrates, to maximize their life-expectancy and reproductive potential. These food resources are commonly obtained from animal secretions or plant exudates, including honeydew, fruit juices and both floral and extra-floral nectar. In addition to exogenous sources of nutrition, adult parasitoids obtain endogenous sources from their hosts through 'host-feeding' behavior, whereby blood is imbibed from the host. Resources obtained from the host contain lipids, proteins and sugars that are assumed to enhance longevity and/or fecundity. Here we conducted an experiment exploring the effects of naturally occurring sugars on longevity and fecundity in the solitary hyperparasitoids, Lysibia nana and Gelis agilis. Although both species are closely related, L. nana does not host-feed whereas G. agilis does. In a separate experiment, we compared reproduction and longevity in G. agilis reared on either honey, a honey-sugar 'mimic', and glucose. Reproductive success and longevity in both hyperparasitoids varied significantly when fed on different sugars. However, only mannose- and water-fed wasps performed significantly more poorly than wasps fed on four other sugar types. G. agilis females fed honey produced twice as many progeny as those reared on the honey-sugar mimic or on glucose, whereas female longevity was only reduced on the mimic mixture. This result shows not only that host feeding influences reproductive success in G. agilis, but also that non-sugar constituents in honey do. The importance of non-sugar nutrients in honey on parasitoid reproduction is discussed.  相似文献

This work describes the synthesis and characterization of four new ligands derived from 1,3-propanediamine in addition to the preparation and characterization of their respective platinum(II) complexes by reaction with K2PtCl4. These ligands were obtained by the reaction of the corresponding alkyl mesylate with 1,3-propanediamine. We have prepared compounds having different carbon chains lengths in an attempt to correlate this factor, which influences the lipophilicity of the compounds, with cytotoxic activity. Octanol/water partition coefficients, the effect of the four complexes on the growth of two tumoral cell lines, and their cellular uptake were investigated. Increasing lipophilicity enhances the rate of cellular uptake and, consequently, the cytotoxic activity.  相似文献

Large-scale sequence analyses of influenza viruses revealed that nonstructural 1 (NS1) proteins from avian influenza viruses have a conserved C-terminal ESEV amino acid motif, while NS1 proteins from typical human influenza viruses have a C-terminal RSKV motif. To test the influence of the C-terminal domains of NS1 on the virulence of an avian influenza virus, we generated a wild-type H7N1 virus with an ESEV motif and a mutant virus with an NS1 protein containing a C-terminal RSKV motif by reverse genetics. We compared the phenotypes of these viruses in vitro in human, mouse, and duck cells as well as in vivo in mice and ducks. In human cells, the human C-terminal RSKV domain increased virus replication. In contrast, the avian C-terminal ESEV motif of NS1 increased virulence in mice. We linked this increase in pathogenicity in mice to an increase in virus replication and to a more severe lung inflammation associated with a higher level of production of type I interferons. Interestingly, the human C-terminal RSKV motif of NS1 increased viral replication in ducks. H7N1 virus with a C-terminal RSKV motif replicated to higher levels in ducks and induced higher levels of Mx, a type I interferon-stimulated gene. Thus, we identify the C-terminal domain of NS1 as a species-specific virulence domain.Interspecies transmission of influenza viruses can lead to the introduction of new subtypes of influenza virus into the human population (31). The emergence of a new influenza virus that is able to spread efficiently between humans can cause a pandemic, as evidenced by the recent introduction of the swine-origin 2009 A/H1N1 virus to humans (10). The spread of avian influenza A viruses from birds to humans could also lead to the introduction of a new viral subtype with pandemic potential (22). Fortunately, the efficient replication of avian influenza A viruses in humans and interhuman transmission are generally limited and require further adaptations of the virus to humans. One determinant of host adaptation lies in the receptor binding specificity of hemagglutinin (HA) (52). In addition, several reports have underlined the role of amino acid 627 of the PB2 polymerase subunit in determining viral host range and virulence (15, 36, 44, 45). Large-scale sequence analyses of viruses isolated from different bird and mammalian species have been performed in order to identify previously unrecognized determinants of host adaptation and virulence (2, 32). Those studies have identified a 4-amino-acid motif in the C-terminal domain of NS1 that could represent a previously unnoticed host adaptation motif. Indeed, the vast majority of avian influenza viruses have an NS1 protein with a C-terminal ESEV domain, while typical human viruses have a conserved RSKV domain. The conservation of these species-specific motifs in the NS1 protein despite important sequence variability in the rest of the protein suggests that these four C-terminal amino acids are under strong selection pressure in their respective natural hosts (3, 5, 25).NS1 is a multifunctional protein implicated in the regulation of viral gene expression and in the inhibition of the host antiviral response (12). In order to test the role of these newly identified NS1 domains, Jackson et al. previously introduced various C-terminal motifs into NS1 of the mouse-adapted human influenza virus A/WSN/33 strain by use of reverse genetics (24). Mice inoculated with a virus containing an avian C-terminal ESEV NS1 domain had high viral loads in the lungs and decreased survival compared to mice inoculated with a virus containing a C-terminal RSKV domain. These results showed that the C-terminal ESEV motif found in avian NS1 proteins increases virulence in mice when introduced into a human strain of influenza virus. Whether this finding also applies to avian influenza viruses remains unknown. Moreover, whether the C-terminal ESEV domain of NS1 increases replication in human cells remains unknown. Finally, how the C-terminal domains of NS1 modulate virulence in nonmammalian hosts, such as birds, is also unknown.Here, we assessed the contribution of the C-terminal domains of NS1 to the pathogenicity of an avian influenza virus. By using reverse genetics, we generated H7N1 viruses containing an NS1 protein with a C-terminal avian ESEV domain or a C-terminal human RSKV domain. The replications of these viruses in human, mouse, and duck cell were compared. In addition, we assessed their pathogenicity in mice and ducks. Our results show that the C-terminal RSKV domain increases the replication of an avian influenza virus in human cells. To our surprise, we observed that the C-terminal RSKV domain increases replication in ducks. In contrast, the C-terminal ESEV domain increases virulence in mice. Thus, we identify the C-terminal domain of NS1 as a species-specific virulence domain.  相似文献

Mining companies are expected to return land to a stable, productive, and self‐sustaining condition by rehabilitating degraded areas to also deliver social benefits, an essential dimension of sustainable land management. This research aimed to develop a framework for mine rehabilitation planning based on an integrated analysis of the social‐ecological system provided by the ecosystem services concept to facilitate community engagement and the delivery of social benefits. An Ecosystem Services Assessment for Rehabilitation framework was tested at two bauxite mines undergoing ecological restoration. The mines are operated by the same company in two countries. Key results showed that the framework can help companies, regulators, and community members alike identify whether biophysical restoration efforts translate into key human benefits. Overall the framework provides a means for enhancing community engagement to explicitly address social benefits that, with a business as usual focus on ecological goals, may not be delivered. The ecosystem services concept provides a practical approach to link ecological and social outcomes of mine restoration.  相似文献

We studied the cytotoxic effect and the uptake of Pd(II) complexes of doxycycline (Dox), [Pd(Dox)Cl2], and tetracycline (Tc), [Pd(Tc)Cl2], in chronic myelogenous leukemia cells. The effect of the compounds on macrophage viability was also investigated. Compound 1 is more effective than compound 2 in inhibiting the growth of K562 cells with the IC(50) values of 14.44 and 34.54 microM, respectively. There is a good correlation between cell-growth inhibition and intracellular metal concentrations, determined by inductively coupled plasma atomic emission spectroscopy (ICP-AES). Incubation of the cells with equitoxic concentrations of both compounds yields approximately the same intracellular Pd concentration. At the IC(50) doses, intracellular concentration is ca. 33 x 10(-16) mol/cell for both compounds 1 and 2. This suggests that more [Pd(Tc)Cl2] is needed to produce a cytotoxic effect, because it enters cells more slowly. Both compounds up to 16 microM did not affect the viability of mouse peritoneal macrophages after a 48-h incubation. After 72 h of incubation, the IC(50) values are 22 for [Pd(Dox)Cl2] and 40 microM for [Pd(Tc)Cl(2)]. Therefore, the cytotoxic effect in cancer cells exhibited by both compounds is higher than their effect in macrophages.  相似文献

The degree to which evolutionary trajectories and outcomes are repeatable across independent populations depends on the relative contribution of selection, chance and history. Population size has been shown theoretically and empirically to affect the amount of variation that arises among independent populations adapting to the same environment. Here, we measure the contribution of selection, chance and history in different-sized experimental populations of the unicellular alga Chlamydomonas reinhardtii adapting to a high salt environment to determine which component of evolution is affected by population size. We find that adaptation to salt is repeatable at the fitness level in medium (Ne = 5 × 104) and large (Ne = 4 × 105) populations because of the large contribution of selection. Adaptation is not repeatable in small (Ne = 5 × 103) populations because of large constraints from history. The threshold between stochastic and deterministic evolution in this case is therefore between effective population sizes of 103 and 104. Our results indicate that diversity across populations is more likely to be maintained if they are small. Experimental outcomes in large populations are likely to be robust and can inform our predictions about outcomes in similar situations.  相似文献
