Pulsed Doppler velocimetry was used to record a grid of velocity waveforms 0.5 cm downstream of the cephalic mesenteric and left renal branches of the aorta in anesthetized dogs. Aortic velocity contours at different phases of the pulse cycle were developed from the grid of waveforms. Transient flow recirculation occurred in the post-systolic phase of the pulse cycle at the aortic wall opposite the branch artery. There was no recirculation at the systolic peak but there was asymmetry of the velocity profile. In contrast, in the in vivo post-stenotic velocity field recirculation persists throughout most of the cycle. These results compare well with similar results from pulsatile model studies. Spectral width of the Doppler signal was not increased in any of the velocity waveforms immediately downstream of the branches, this suggests that flow is relatively undisturbed in these locations.  相似文献

Summary A total of 56 Duchenne muscular dystrophy (DMD) patients and 11 Becker muscular dystrophy (BMD) patients was analyzed by extended multiplex amplification of the DMD/BMD gene; deletions were found in 60% of these patients. The data obtained were used to test the frameshift hypothesis and to compare the distribution of familial versus isolated cases. A significant correlation was found between deletions and isolated cases. Additional experiments were performed in order to determine the deletion breakpoints more precisely. These data are a prerequisite for carrier analysis in the respective families by detection or exclusion of aberrant cDNA fragments derived from ectopic lymphocyte RNA. This diagnostic technique is illustrated by 5 examples.  相似文献

The velocity field around arterial stenoses was investigated using a pulsed doppler velocimeter in vivo. Asymmetric zones of recirculation were identified by systolic flow reversal in the post-stenotic field in carotid and iliac arteries of anesthetised dogs. There was a close correlation between shear intensity and turbulence as estimated by the velocity difference between the jet and the recirculation zone and by maximum spectral width respectively. Under the conditions of these experiments, stenosis grade (% diameter reduction) dominated hemodynamic variables such as Reynolds number, oscillation and pulsatility in determining the intensity of turbulence. The method used does not appear to have sufficient resolution to distinguish between turbulence and discrete oscillating velocities (vorticity) nor to allow determination of wall shear stress though the pattern of change of the latter is similar to that found downstream of axisymmetric stenosis in models using steady flow.  相似文献

Phosphoglucoisomerase (PGI), a soluble enzyme, and AChE, a membrane-bound enzyme were studied in transected peroneal nerves of dog and in isolated segments of these nerves. Although activities of both enzymes increased at the ends of transected nerves, marked differences in their behaviour were observed. The increment in AChE activity was much sharper than that of PGI and continued to grow with time whereas the increase in PGI developed fully within the initial hours after transection and did not change thereafter. In an isolated nerve segment AChE accumulated at both ends with a concomitant decrease in the middle part, whereas changes in PGI activity appeared only in the terminal parts, the rest of the nerve remaining at the normal level. The terminal increase of PGI did not, contrary to that of AChE, depend on the length of the isolated segment. The changes in PGI activity may be features of a local peritraumatic reaction whereas those of AChE indicate involvement of the whole segment along which the enzyme containing organelles are transported.  相似文献

To test the hypothesis that susceptibility to NK cell-mediated cytolysis varies inversely with the levels of target cell class I HLA expression, NK-susceptible K562 and MOLT-4 target cells have been transfected via electroporation with cloned human class I HLA-A2 and HLA-B7 genes. Stably transfected cells expressing varying levels of cell-surface class I HLA have been selected by fluorescent activated cell sorting and tested for susceptibility to NK-mediated cytolysis by freshly isolated peripheral blood NK cells from nine normal volunteers as well as by cloned human NK effectors and tumor cells from a patient with an NK cell lymphoproliferative disorder. Expression of class I HLA did not alter the susceptibility of K562 or MOLT-4 target cells to NK-mediated cytolysis by any of the effectors tested. In addition, the class I HLA-expressing transfectant cells were identical to mock transfected cells in their ability to compete for lysis in cold target inhibition assays. Treatment of both mock-transfected and class I HLA-transfected K562 cells with IFN-gamma resulted in decreased susceptibility to NK-mediated cytolysis which was independent of the total level of class I HLA expression. These results demonstrate that the level of target cell class I HLA expression is not sufficient to determine susceptibility or resistance to NK-mediated cytolysis of the classical NK targets K562 and MOLT-4.  相似文献

A differential analysis of CuZn-superoxide dismutase (SOD. EC 1.15.1.1) isozymes after native-polyacry lamide gel elecrrophoresis (PAGE) and isoelectric focusing (IEF) indicated that germinating seeds of Scots pine (Pinus sylvestris L.) 3 days after the start of imbibition (3 DAI) contain five CuZn-SOD isozymes. Two isozymes co-migrated on native–PAGE but were separated after IEF. CuZn-SODs of Scots pine were purified from germinating seeds (3 DAI) by anion-exchange chromatography, hydrophobic interaction chromatography and chromatofocusing. The final separation of CuZn-SOD isozymes was accomplished by native-PAGE. CuZn-SOD isozymes were electroblotted and their NH2-terminal amino acid sequence was determined. Comparisons of the amino acid sequences with sequences of CuZn-SOD isozymes from other plant sources indicated that one CuZn-SOD isozyme was of the chloroplastic type whereas the other four isozymes belonged to the cytosolic-type CuZn-SODs, The NH2-terminal amino acid sequence of the chloroplastic CuZn-SOD and of one cytosolic-type CuZn-SOD were identical to those of two previously isolated, sequenced and localized CuZn-SOD isozymes from Scots pine needles. Two cytosolic-type CuZn-SOD isozymes showed a homology at 20 out of 21 NH2-terminal amino acids. Mitochondria and glyoxysomes were isolated by differential and Percoll density-gradient centrifugation from germinating seeds (3 DAI). The cell fractionation experiments did not suggest that a major part of the CuZn-SOD activity in germinating seeds was derived from glyoxysomes or mitochondria.  相似文献

Summary The nervous system of the most primitive fresh-water Turbellaria of the order Catenulida was investigated. Neurosecretory cells were found to occur in it. These cells have a structure similar to that of nerve cells, they only differ by the presence of neurosecretion granules and a more developed golgi complex. In the simplest Catenulida,Stenostomun, neurosecretory granules are found both in the brain cells and in the trunks in the nerve fibres where they are rather randomly distributed. Another representative of this family, the more highly organizedCatenula, exhibits specialized brain and nerve trunk regions containing neurosecretion granules. Summary A comparsion of the Feulgen hydrolysis curves and the chromatin compactness of the liver cell nuclei of young and old rats was made. It was found that the rate of DNA depurination and chromatin compactness are higher in the liver cell nuclei of old rats, both in di-and tetraploidal cells. Summary We report a collection of 53 prototypic sequences representing known families of repetitive elements from the human genome. The prototypic sequences are either consensus sequences or selected examples of repetitive sequences. The collection includes: prototypes for high and medium reiteration frequency interspersed repeats, long terminal repeats of endogenous retroviruses, alphoid repeats, telomere-associated repeats, and some miscellaneous repeats.
