By using newly hatched (approximately 2 weeks old) brown trout(Salmo trutta) from six families of wild and six families ofsea-ranched origin (seventh generation), we tested the hypothesesthat (1) the hatchery environment selects for increased boldness,and (2) boldness predicts dominance status. Sea-ranched troutspend their first 2 years in the hatchery before being releasedinto the wild at the onset of seaward migration. Trout werepresented with a novel object (tack) and with food (brine shrimp),and their responses were measured and scored in terms of boldness.Siblings with increasing difference in boldness were then pairedin dyadic contests. Fish of sea-ranged origin were on averagebolder than were fish of wild origin, and bolder individualswere more likely to become dominant regardless of origin. Boldnesswas not related to RNA levels, indicating that bold behaviorwas not a consequence of higher metabolism or growth rate. Neitherwas size a predictor of bold behavior or the outcome of dyadiccontests. These results are consistent with studies on olderlife stages showing increased boldness toward predators in hatchery-selectedfish, which suggests that behavioral consequences of hatcheryselection are manifested very early in life. The concordancebetween boldness and dominance may suggest that these behaviorsare linked in a risk prone-aggressive phenotype, which may bepromoted by hatchery selection. However, we also found significantvariation in behavioral and growth-related traits among families,suggesting that heritable variation has not been exhausted bysea-ranching procedures. 相似文献

The septins are a conserved family of GTP-binding proteins that, in the baker''s yeast, assemble into a highly ordered array of filaments at the mother bud neck. These filaments undergo significant structural rearrangements during the cell cycle. We aimed at identifying key components that are involved in or regulate the transitions of the septins. By combining cell synchronization and quantitative affinity-purification mass-spectrometry, we performed a screen for specific interaction partners of the septins at three distinct stages of the cell cycle. A total of 83 interaction partners of the septins were assigned. Surprisingly, we detected DNA-interacting/nuclear proteins and proteins involved in ribosome biogenesis and protein synthesis predominantly present in alpha-factor arrested that do not display an assembled septin structure. Furthermore, two distinct sets of regulatory proteins that are specific for cells at S-phase with a stable septin collar or at mitosis with split septin rings were identified.Complementary methods like SPLIFF and immunoprecipitation allowed us to more exactly define the spatial and temporal characteristics of selected hits of the AP-MS screen. 相似文献

Summary Cellular fatty acids inLactobacillus büchneri were transmethylated with H2SO4 catalyst in methanol at elevated temperature. By optimising the reaction time and the amounts of catalyst and methanol used at a fixed temperature it was possible to maximise the lactobacillic acid yield. A soil nitrogen model was used for a 4-year simulation of nitrogen dynamics and nitrate leaching, both during grass ley growth
and after ploughing a grass ley. Model results were compared with field measurements of soil mineral-N status and leaching.
A soil water and heat model provided daily values for abiotic conditions, which were used as driving variables in the nitrogen
simulation.
Simulated values for mineral-N levels in the soil agreed well with field data for the first 3 years of the simulation. During
the final year the model predicted considerably higher levels of soil mineral-N content compared with measurements. To reach
the mineral-N level measured at the time of ploughing the ley, the simulated N-uptake by plants had to be increased by 8 g
N m−2.
Simulations of nitrate leaching suggested that estimates of leaching based on measurements in tile-drained plots can be considerably
underestimated. Accurate quantification of leaching in tile-drained plots often requires additional information on water-flow
paths.
A substantial increase in simulated and measured values for the mineral-N content of the soil occurred after ploughing the
ley. In the simulation, most of the increase was due to a high crop residue input and the absence of a growing crop after
ploughing. Litter accumulations in the soil during the 4-year period contributed little to the increase in soil mineral-N. 相似文献

Simulation of field scale denitrification losses from soils under grass ley and barley 总被引:2,自引:0,他引:2
Denitrification losses from soils under barley and grass ley crops were simulated. The model, which includes the major processes determining inputs, transformations and outputs of nitrogen in arable soils, represents a scale compatible with information generally available in agricultural field research. The denitrification part of the model includes a field potential denitrification rate and functions for the effect of soil aeration status, soil temperature and soil nitrate content. Easily metabolizable organic matter is assumed not to limit denitrification. Simulated values were compared with denitrification measurements made during two growing seasons in the barley and grass ley treatments of a field experiment in central Sweden.Calibration revealed that the optimal parameter values describing the effect of soil aeration on denitrification rates were similar for both treatments. The response function derived agreed well with two data sets found in the literature. The potential denitrification rate constant, derived in the simulations, was higher for grass ley than for barley, which was consistent with the differences in overall rates of carbon and nitrogen turnover found between treatments.The simulated mean denitrification rates for the two seasons were within 20% of the mean of the measured values. However, simulated denitrification showed less temporal variability and a less skewed frequency distribution than measured denitrification. Some of the measured denitrification events not explained by the model could have been due to the stimulating effects of soil drying/wetting and freezing/thawing on microbial activity. 相似文献

A loose structure of some 30 residues at the amino-terminus contains the phosphorylatable tyrosine and the binding site for the p11 regulatory chain. The following p33 domain retains the lipid-binding site as well as the Ca2+ site which influences the spectral properties of the single tryptophan and one tyrosine. From a previously published theory (Israelsson and Johnsson 1967) for circumnutations in Helianthus annuus it is possible to predict the geotropical curvatures of the hypocotyls. This extension of the theory is given in the present paper and some geotropical experiments are performed and discussed. Nejvy??í intensitu fotosynthesy i nejvy??í obsah chlorofylu (vzhledem k plo?e listové) mají mladé, ale ji? dob?e rozvinuté listy, tj. t?etí a? ?tvrté od vrcholu (prvním listem se rozumí list o plo?e asi 20 cm2). Tyto listy nazýváme ?fotosyntheticky dospělými“. Listy nejmlad?í a zejména pak listy star?í mají intensitu fotosynthesy i obsah chlorofylu ni??í; u nejstar?ích list? je intensita fotosynthesy prakticky nulová. Intensita fotosynthesy i obsah chlorofylu se během vývoje mění: jejich momentální rozdíly u list? v genetické spirále jsou z?ejmě shodné s jejich změnami v ontogenesi listu. Pokles intensity fotosynthesy p?i stárnutí list? je rychlej?í ne? pokles obsahu chlorofylu. P?i ur?itém obsahu chlorofylu (tj. asi 2,25 a? 2,45 mg/dm2) klesá intensita ?isté fotosynthesy k nule. Intensita fotosynthesy je v lineárním vztahu k mno?ství chlorofylu (p?i p?epo?tu na plo?nou jednotku), a to nezávisle na poloze listu v genetické spirále. Pneumocystis carinii is a pathogen which, causes fatal pneumonia in patients with the acquired immune deficiency syndrome (AIDS). To facilitate the basic study of P. carinii , we have analyzed its major surface proteins by both immunochemical and biochemical methods. The major protein components of both cysts and trophozoites are a group of proteins called "P115" with apparent masses of 105–120 kd. It includes 6 isoelcclric variants. A monoclonal antibody raised against cysts recognizes all 6 variants and reacts with epitopes located in the cell wall indicating that P115 is an immunorcactive surface component. The isoelectric variants contain identical or closely related protein components and they are mannose-rich glycoproteins. The isoelectric variation may be due primarily to differences in glycosylation. The majority of sera from humans with diagnosed pneumocystosis that were tested reacted strongly with the P115 proteins. To develop probes for DNA diagnosis and to facilitate molecular studies, a genomic DNA library of P. carinii has been constructed. Some of these clones were used for DNA hybridization analysis of rat and human lungs. 相似文献
