Melanoma depends on, interacts with and reacts to the stroma in which it is embedded, including fibroblasts, extracellular
matrix, endothelial cells and immune cells. However, the impact of melanoma on the epidermal tumor microenvironment—the multilayered
epithelium of the skin—is poorly understood. Gap junctions are essential for intercellular communication and involved in proliferation,
differentiation and homeostasis of keratinocytes. We have shown previously that the gap junction proteins connexin 26 and
30 (Cx26 and Cx30) are induced in the epidermal tumor microenvironment of skin cancers including melanoma. This study compares
the extent of Cx26, Cx30 and Cx43 expression in the epidermal microenvironment of melanocytic nevi and melanomas and its association
with melanoma thickness, proliferative index of the tumor and its microenvironment, and with 5-year metastasis and survival.
We found that induction of Cx26 and Cx30 cell–cell border expression in the epidermal tumor microenvironment correlates to
malignancy. Importantly, there was a significant correlation of tumor thickness with the vertical epidermal Cx26 and Cx30
expression pattern and the horizontal Cx26 dissemination. Furthermore, horizontal Cx26 expression correlated with metastasis.
Vertical epidermal expression patterns of Cx26 and Cx30 significantly correlated with the proliferative index in the epidermal
tumor microenvironment but not with the proliferative index in the tumor. In contrast, Cx43 did not correlate with malignancy,
thickness or proliferative index. In summary, here we show for the first time a significant association between the progression
of melanoma and alterations in its epithelial tumor microenvironment. 相似文献

A new species of land planarian is described from Kenya. Microplana rermirophaga sp. nov. preys on termites by capturing them at the openings to their nest. The pharynx is unusually far forwards for a land planarian and this is probably related to the active nature of the prey. There is an anterior depression caused by the termination of parenchymal longitudinal muscle bundles which might act as a sucker. Summary The partitioning of energy in three species of nematode, Paroigolaimella bernensis, Diplogasteritus nudicapitatus and Rhabditis curvicaudata, from a polysaprobic environment is considered. Temperature was shown to have a profound impact on the rate at which these organisms obtained food resources and on the partitioning of energy into growth, reproduction and maintenance. Declining temperature resulted in a reduction in energy consumption and in production and maintenance costs. Absorption efficiencies were relatively low, rarely exceeding 20% and lying on average between 5–15%. Net production efficiencies varied throughout the life-cycle, but attained maximum levels of 70–80%. Females achieved higher net production efficiency than males. 1. Cellulose decomposition in forest and orchard soils was investigated by studying the breakdown of boiled and washed cellophane in the soils and in vitro. Decomposition occurred from quick to slow in the order: orchard on clay soil, forest on clay soil, forest on sandy loam, and in the latter in the order: calcareous mull, acid mull and mor. 2. In the different forest soils which were investigated the rate of decomposition was parallel to their water capacity. It slowed down considerably when the water content of the soil decreased, especially after the wilting point was reached. 3. Of the fungi isolated from these soils, those from orchard soil — 5% to 50%Fusarium spp. — were among the fastest decomposers of cellulose. This agrees with, and may explain the high rate of decomposition in orchard soil. 4. Decomposition in pure culture is quicker than in soil. As filtersterilized soil extract checked the decomposition in pure culture, but heat-sterilized soil extract did not, an extractable but heat-sensitive substance may be one retarding factor. 相似文献

Direct enantiomeric separations of 17 chiral amidotetralins by means of high performance liquid chromatography were performed on stationary phases composed of tris(3,5-dimethylphenylcarbamate) derivatives of cellulose and amylose, coated on silica gel. The enantiomers of 15 out of 17 amidotetralins were resolved with a resolution of more than 1.5 by at least one of the chiral stationary phases. The stationary phases showed complementary results with regard to the separation of the amidotetralins, that is, pairs that did not separate on the cellulose-type column were well separated on the amylose-type column, and vice versa. CD40 is a receptor with numerous functions in the activation of antigen presenting cells (APCs), particularly dendritic cells (DC). Using phage display technology, we identified linear peptides containing a novel FPGN/S consensus sequence that enhances the binding of phage to a purified murine CD40-immunoglobulin (Ig) fusion protein (CD40-Ig), but not to Ig alone. To examine the ability the FPGN/S peptides to enhance adenoviral infection of CD40-positive cells, we used bifunctional peptides consisting of an FPGN-containing peptide covalently linked to an adenoviral knob-binding peptide (KBP). One of these, FPGN2-KBP, was able to enhance adenoviral infection of both murine and human DCs in a dose-dependent manner. FPGN2-KBP also improved infection of murine B cell blasts, a murine B lymphoma cell line (L10A), and immortalized human B cells. To demonstrate that enhancement of adenoviral infection depended on the presence of CD40, we analyzed infection of the breast cancer line, SKBR3, that does not express CD40 or the adenovirus cellular receptor, CAR. Infection of SKBR3 cells was enhanced by FPGN2-KBP following transient transfection with a plasmid vector that expresses murine CD40, but not when the cells were mock-transfected. In conclusion, we have isolated a peptide that binds to murine CD40, and promotes the uptake of adenoviruses into CD40-expressing cells of both murine and human origin, suggesting that it may have potential applications for antigen delivery to CD40-positive antigen-presenting cells. 相似文献
