Synopsis The incorporation of dissolved oceanic constituents in the otoliths of fish has potential as a chemical tracer for reconstructing the early life history of marine fish. Wavelength dispersive spectrometers on an electron microprobe were used to measure Na, Mg, P, S, Cl, K, Ca, and Sr concentrations on the outer margins of 57 juvenile walleye pollock, Theragra chalcogramma, otoliths from five locations in the Gulf of Alaska and Bering Sea. Discriminant analyses that used various combinations of Na, P, K, Sr, and fish standard length and/or age showed that 60–80% of the samples could be assigned to the correct capture locality. While the concentrations of some of the measured elements correlated with standard length or age of the fish, there are measurable differences among localities when concentrations are length or age corrected, mainly due to differences in Na and K concentrations. Elemental composition of otoliths potentially could be used to assign fish from a mixed stock fishery to original stocks, information that is greatly needed for the effective management of fish stocks. 相似文献

Experimental work was performed on drill cuttings sampled from off-shore drilling operations. The cuttings contained remnants of two different types of drilling muds: a water-ilmenite based mud and an olefin-barite based mud. In a pilot experiment, a gradient of up to 65 mm thick layers of water-ilmenite based cuttings were added to 78 cm2 core samples. In another set-up, 3 mm layers of water-ilmenite and olefin-barite based cuttings were added to replicate 1000 cm2 box-core samples with natural benthic communities transferred from the Oslofjord, S.E. Norway. Boxes with no addition and addition of 3 mm “clean” sediment were used for control. Increased consumption of oxygen and nitrate indicated the presence of degradable organic phases in both mud systems. The release of silicate showed a general decrease with increasing thickness of the cuttings layer, but maximum rates in the 3.1 mm treatment indicated increased bioturbation at low and moderate doses. The initial (field) composition of the macrobenthic communities was maintained throughout the 3 months experimental period, and at community level, no significant difference was observed between the four treatments at the end of the exposure period. However, after grouping the treatments into “clean” (untreated control and 3 mm clean sediment) and “cuttings” (3 mm water-ilmenite and 3 mm olefin-barite based cuttings), multivariate statistical analyses revealed a significant difference in community composition between clean and cuttings treatments, and three taxa showed significantly reduced abundances in the cuttings treatments. Chemical toxicity of mud components is assumed to be small, and the observed effect was more likely a result of physical properties such as the size or shape of cuttings particles. 相似文献

Noroviruses are major pathogens associated with acute gastroenteritis. They are diverse viruses, with at least six genogroups (GI-GVI) and multiple genotypes defined by differences in the major capsid protein, VP1. This diversity has challenged the development of broadly cross-reactive vaccines as well as efficient detection methods. Here, we report the characterization of a broadly cross-reactive monoclonal antibody (MAb) raised against the capsid protein of a GII.3 norovirus strain. The MAb reacted with VLPs and denatured VP1 protein from GI, GII, GIV and GV noroviruses, and mapped to a linear epitope located in the inner shell domain. An alignment of all available VP1 sequences showed that the putative epitope (residues 52–56) is highly conserved across the genus Norovirus. This broadly cross-reactive MAb thus constitutes a valuable reagent for the diagnosis and study of these diverse viruses. 相似文献

Several quinazoline derivatives were made as mitochondrial complex 1 inhibitors. Compound 4 showed an IC(50) of 11.3 nM and was the most potent compound of this series. The (18)F analog of 4, [(18)F] 4, was injected in the rat and showed high and rapid heart uptake, fast liver clearance, and low blood uptake. Images obtained using a microPET showed clear delineation of the myocardium in normal rats and perfusion deficit in ischemic rats. In the non-human primate, [(18)F] 4 showed rapid uptake and clearance from the myocardium and high liver uptake. 相似文献

A crucial and distinctive feature of tuberculosis infection is that Mycobacterium tuberculosis (Mtb) resides in granulomatous lesion at various stages of disease development and necrosis, an aspect that is little understood. We used a novel approach, applying high resolution magic angle spinning nuclear magnetic resonance spectroscopy (HRMAS NMR) directly to infected tissues, allowing us to study the development of tuberculosis granulomas in guinea pigs in an untargeted manner. Significant up-regulation of lactate, alanine, acetate, glutamate, oxidized and the reduced form of glutathione, aspartate, creatine, phosphocholine, glycerophosphocholine, betaine, trimethylamine N-oxide, myo-inositol, scyllo-inositol, and dihydroxyacetone was clearly visualized and was identified as the infection progressed. Concomitantly, phosphatidylcholine was down-regulated. Principal component analysis of NMR data revealed clear group separation between infected and uninfected tissues. These metabolites are suggestive of utilization of alternate energy sources by the infiltrating cells that generate much of the metabolites in the increasingly necrotic and hypoxic developing granuloma through the glycolytic, pentose phosphate, and tricarboxylic acid pathways. The most relevant changes seen are, surprisingly, very similar to metabolic changes seen in cancer during tumor development. 相似文献

Background
Haemophilus influenzae type b (Hib) vaccine and pneumococcal conjugate vaccine (PCV) are relatively expensive, newly introduced vaccines in China. This study evaluates the impact of residency and urbanicity on Hib vaccine and PCV coverage for children aged 2 to 7 years living in Shanghai, China, in August 2012.Methods
In this exploratory cohort study, a sample of children aged 2 to 7 years, all of whom were eligible to have received the complete series of Hib vaccine and PCV, was obtained from the Shanghai Immunization Program Information System. Three measures of vaccination coverage for Hib vaccine and PCV were examined: dose 1 coverage, series completion, and timeliness of dose 1 vaccination. Multivariable binomial regression was used to estimate the difference in vaccination coverage between locals and the floating population.Results
Dose 1 coverage was 50.9% for Hib vaccine and 11.4% for PCV for the 28,141 abstracted pediatric records. For both vaccines, dose 1 coverage was higher in locals than in the floating population. The disparity in coverage between locals and the floating population was greater in suburban areas than urban areas. Of all children who received dose 1, 79.7% completed the Hib vaccine series, and 91.3% completed the PCV series. Timely dose 1 coverage was 8.2% for Hib vaccine and 0.5% for PCV.Conclusion
Low vaccination coverage and extremely low levels of timely dose 1 vaccination indicate that current vaccination efforts are inadequate to reduce the burden of Hib and pneumococcal disease among Chinese children, especially infants. Government funding of the Hib vaccine and PCV through the Expanded Program on Immunization would increase uptake and could also ensure that improvement in the timeliness of administration and series completion is targeted for all demographic groups. 相似文献10.

Accelerating rhizome growth is crucial to enhancing propagule production in rhubarb (Rheum rhabarbarum L.) because the crop is propagated through rhizome divisions. This can be achieved through manipulating source-sink activity.
This study tested the hypothesis that synthetic plant growth retardants Prohexadione-Ca and CCC enhance rhizome growth in
rhubarb. Two different concentrations of these plant growth retardants and GA3 (positive control) were foliarly applied on the cultivar German Wine at three stages of shoot growth under greenhouse conditions.
Both Prohexadione-Ca and CCC favorably enhanced rhizome growth through suppressing shoot growth. CCC at 3000 mg L−1 produced the best results and the effect was apparent when applied at 12 weeks after shoot emergence. The rhizome diameter,
fresh weight, and the number of viable buds were enhanced significantly in plants sprayed with CCC 3000 mg L−1. Both Prohexadione-Ca and CCC were equally effective in enhancing dry mass and starch allocation preferentially toward the
rhizome. Prohexadione-Ca- and CCC-induced rhizome growth enhancement could possibly be due to their known role as GA biosynthesis
inhibitors or through increasing photosynthetic efficiency and preferentially reallocating carbohydrates to the rhizome. 相似文献
