Abstract Electrofusion of protoplasts of two mutant strains of Hansenula polymorpha resulted in high fusion and hybrid yields when the calcium ions present in the conventional fusion medium replaced by zinc ions. The optimal fusion conditions were an alignment field of 0.4 kV cm−1 strength and 2 MHz frequency for 30 s, followed by two consecutive pulses of 12 kV cm−1 strength and 15 μs duration. With 0.05–0.1 mM zinc ions in the fusion medium an average clone number of 104 –105 clones per 108 input cells was reached. The presence of about 0.6 mM magnesium ions in the zinc fusion medium was essential. 相似文献

both spent a short time in culture. We have previously demonstrated that although the T1 values of paired normal and neoplastic Syrian hamster (SH) fibroblastic cells in culture are not significantly different
when measured at 300 MHz, the 300 MHz T2 values for the neoplastic cells are smaller than those of the normal cells. (Xin et al. (1986),Cell Biophysics
8, 213.) Since treatment of normal diploid cells with polypeptide growth factors or tumor promoters frequently results in reversible
expression of neoplasia-associated phenotypes, T1 and T2 were obtained at 300 MHz for treated and untreated SH cells to see if these compounds could also produce smaller 300 MHz
T2 values. Secondary culture SH fetal fibroblast cells were treated with epidermal growth factor (EGF), fibroblast growth factor
(FGF), phorbol-12,13-didecanoate (PDD) and 4-α-phorbol-12,13-didecanoate (4αPDD). Treatment with either growth factor resulted
in smaller T2 values, but a statistically significant decrease was not observed for PDD or 4αPDD. The observed reductions in T2 values were correlated with the morphological and growth-stimulatory effects of these compounds on the cells. 相似文献

The laser flow microfluorometer (FMF) can determine the amounts of certain components in single cells at sample rates of several thousand cells per second. This technique has been employed to characterize Bacillus subtilis populations in batch fermentations with different inocula. Protein and nucleic acid distributions obtained by FMF analyses at different times during the batch have been decomposed using an optimized fit of summed subpopulation distributions. The results of these decomposition calculations, some of which have been approximately confirmed by independent microscopic observations, indicate that the relative numbers of single rods, cell chains, spores, and swollen rounded cells change dramatically during the entire fermentation including the stationary phase. The dynamics of these subpopulations may be related to secondary metabolite production. 相似文献

Objective
To examine the extent of implementation for patient safety (PS) and patient-centeredness (PC) strategies and their association with hospital characteristics (type, ownership, teaching status, annual evaluation grade) in Iran.Methods
A cross-sectional study through an adapted version of the MARQuIS questionnaire, eliciting information from hospital and nursing managers in 84 Iranian hospitals on the implementation of PS and PC strategies in 2009–2010.Results
The majority of hospitals reported to have implemented 84% of the PS and 72% of the PC strategies. In general, implementation of PS strategies was unrelated to the type of hospital, with the exception of health promotion reports, which were more common in the Social Security Organization (SSO), and MRSA testing, which was reported more often in nonprofit hospitals. MRSA testing was also more common among teaching hospitals compared to non-teaching hospitals. The higher grade hospitals reported PS strategies significantly more frequently than lower grade hospitals. Overall, there was no significant difference in the reported implementation of PC strategies across general and specialized hospitals; except for the provision of information in different languages and recording of patient’s diet which were reported significantly more often by general than specialized hospitals. Moreover, patient hotel services were more common in private compared to public hospitals.Conclusions
Despite substantial reporting of PS and PC strategies, there is still room for strengthening standard setting on safety, patient services and patient-centered information strategies in Iranian hospitals. To assure effective implementation of PS and PC strategies, enforcing standards, creating a PS and PC culture, increasing organizational responsiveness, and partnering with patients and their families need more attention. 相似文献7.

Microglia are the primary resident immune cells in the retina. They regulate neuronal survival and synaptic pruning making them essential for normal development. Following injury, they mediate adaptive responses and under pathological conditions they can trigger neurodegeneration exacerbating the effect of a disease. Retinal organoids derived from human induced pluripotent stem cells (hiPSCs) are increasingly being used for a range of applications, including disease modelling, development of new therapies and in the study of retinogenesis. Despite many similarities to the retinas developed in vivo, they lack some key physiological features, including immune cells. We engineered an hiPSC co-culture system containing retinal organoids and microglia-like (iMG) cells and tested their retinal invasion capacity and function. We incorporated iMG into retinal organoids at 13 weeks and tested their effect on function and development at 15 and 22 weeks of differentiation. Our key findings showed that iMG cells were able to respond to endotoxin challenge in monocultures and when co-cultured with the organoids. We show that retinal organoids developed normally and retained their ability to generate spiking activity in response to light. Thus, this new co-culture immunocompetent in vitro retinal model provides a platform with greater relevance to the in vivo human retina. 相似文献

This study, for the first time, investigated the total protein changes and protease activity pattern of three cultivars of
kiwifruit including Abbott, Allison and Hayward, cultivated in the north of Iran, during 6 months of fruit development from
July to December 2008. During the last month of sampling, fruits were stored in ambient temperature. According to the result
of fruit extracts, pattern of changes in total protein concentration showed three significant steps: low concentration of
protein during the early stages of fruit development until middle of July for Allison and Hayward and early August for Abbott,
increasing protein concentration until middle of October for Allison and Hayward and late October for Abbott, and steady state
level of protein concentration until harvesting time. During storage time of fruits, all three cultivars showed increasing
level of total protein concentration. Among the cultivars, Hayward and Allison showed the lowest and highest total protein
concentration, respectively. Protease activity showed increasing pattern in all of the investigated cultivars significantly
until harvesting time. During postharvest time, Hayward did not show any significant change in protease activity but, the
other two cultivars had increasing pattern from 10 to 20th of storage and then decreased significantly. Among the cultivars,
Abbott showed the lowest and Allison the highest level of total protease activity at harvesting and postharvesting time. SDS-PAGE,
ion-exchange chromatography and zymogram native-PAGE of fruits extraction showed that dominant protein and protease activity
in these cultivars during fruit development and ripening process is related to actinidin. 相似文献
