Molecular crowding has been shown to be important in many cellular processes. The crowded environment in the cell results in a significant proportion of the cellular water being in contact with macromolecules such as proteins and DNA. These interfacial water molecules show a reduced dynamic motion that has been observed with isolated macromolecules using several biophysical techniques. Previously we investigated the inelastic neutron scattering properties of water closely associated with isolated biomolecules, and showed that interfacial water is strongly perturbed, as judged by its energy transfer spectrum. Here we have probed living cells using inelastic and quasielastic neutron scattering. We have found that mild heat stress ('heat shock'), which causes some proteins to become unfolded in the cell, results in changes in the inelastic neutron scattering in the librational region (45-130 meV). Heat shock also causes a narrowing of the quasielastic scattering peak. These changes can be understood in terms of an increase in the proportion of interfacial water molecules, and a net reduction in proton dynamics.  相似文献

Abnormalities in sonic hedgehog (SHH) signaling pathway components are major contributing factors in the development of nevoid basal cell carcinoma syndromes (NBCCS) that include SHH, PTCH, SMO and GLI. The novel patched homologue (PTCH) mutation and clinical manifestations with NBCCS links PTCH haplosufficiency and aberrant activation of the sonic hedgehog/Patched/smoothened pathway. To investigate further the molecular genetics of NBCCS, we performed mutation analysis of PTCH gene in a family case with five affected members. These clinical manifestations might be associated with a novel constitutional mutation of the PTCH gene, 3146A → T (1049N → I), in exon 17. The analyzed results of tumor tissue show a high expression of GLI. Our findings suggested that the mutation of 3146A → T may be the cause of high expression of GLI and permit SMO to transmit signal to the nucleus through SHH/PTCH/SMO pathway.  相似文献

In grass, the evolutionary relationship between lemma and palea, and their relationship to the flower organs in dicots have been variously interpreted and wildely debated. In the present study, we carried out morphological and genetic analysis of a palealess mutant (pal) from rice (Oryza sativa L.), and fine mapping the gene responsible for the mutated trait. Together, our findings indicate that the palea is replaced by two leaf-like structures in the pal flowers, and this trait is controlled by one recessive gene, termed palealess1 (pal1). With a large F2 segregating population, the pal1 gene was finally mapped into a physical region of 35 kb. Our results also suggest that the lemma and palea of rice are not homologous organs, palea is likely evolutionarily equivalent to the eudicot sepal, and the pal1 should be an A function gene for rice floral organ identity.  相似文献

Modern soil phytoliths can potentially provide analogues for phytolith assemblages from archaeological and palaeoecological contexts. To assess the reliability of soil phytoliths for representing different plant communities, we analysed phytoliths in surface soils and parent plants at 65 sites representing five types of regional vegetation in Northeast China. The results demonstrated that surface soil phytolith assemblages could clearly differentiate samples from herbaceous and woody communities, and samples from Poaceae and non-Poaceae communities could be separated statistically. In addition, woody communities could be differentiated into a broadleaf-Poaceae community, a broadleaf-non-Poaceae community and a conifer and broadleaf-non-Poaceae community, except for some overlapping samples. Soil phytolith assemblages are thus able to differentiate regional vegetation types into different plant community types. In the present study, soil phytoliths represented about 30% of the phytoliths present in the aboveground vegetation. In addition, soil phytoliths from different communities reflected the aboveground vegetation with slightly different degrees of accuracy, and in addition different morphotypes exhibited different degrees of representational bias. Some morphotypes (e.g. rondel, elongate psilate, lanceolate) overrepresented the abundance of the associated plant taxa; morphotypes such as tracheid, conical epidermal, stomata and others under-represented the original plant richness; and other morphotypes, e.g. saddle, trapeziform sinuate, scutiform, were in good agreement with the numbers of plant taxa in the plot inventory. Thus, any quantitative palaeovegetation reconstruction using phytoliths should begin with the calibration of soil phytolith assemblages. We conclude that our findings provide improved phytolith analogues for different plant communities, with applications in palaeoenvironmental reconstruction, and they also provide additional insights into the mechanisms of phytolith production and deposition.  相似文献

Agrostis stolonifera L. ‘Penn A-4’ is a common creeping bentgrass species that is widely used in urban landscaping and golf courses. To prolong the green stage of this grass, a dehydrin gene PicW isolated from Wilson’s spruce (Picea wilsonii) was transformed into plants of ‘Penn A-4’ cultivar via a straightforward stolon node infection system. A putative transgenic plant was obtained and its tolerance to low-temperature stress was evaluated. When the transgenic line was subjected to a freezing (??5 °C) treatment, it showed better viability and more robust physiology than wild type, as evidenced by higher soluble sugar and proline contents, and lower relative electrical conductivity and malondialdehyde content. The transgenic line also showed tolerance to a chilling treatment (5 °C), although its performance was not significantly different from that of wild-type plants. Overall, the research here clearly revealed the explicit role of PicW in increasing freezing tolerance of grass at the whole-plant level, and demonstrated that the straightforward stolon node transformation method could be well used to genetically modify turfgrass. The obtained transgenic line might be as genetic resource for breeding program and practiced to grow in cold temperate zones.  相似文献

DNA methylation is a general epigenetic mechanism for plants, animals, and fungi to adapt to environmental variation. Two biotypes of the Russian wheat aphid (Diuraphis noxia), Biotype 1 and Biotype 2, have different virulence to host plants. In this study, in addition to a high polymorphism, DNA methylation at cytosines were observed in genomic fragments of four genes in Biotype 1 and Biotype 2, after the genomic DNA was treated with sodium bisulfite. These genes presumably encode proteins and enzymes in salivary glands of aphids. The two Biotype 1 showed different methylation levels, that is, Biotype 1 showed a higher methylation on the four genes. Two thirds of methyl cytosines were in a sequence context of CHH (H = A, C, or T). Some polymorphism and methylation sites were located at important positions in terms of enzyme function, such as close to catalytic residues or inhibitor binding residues. These findings may provide clues to explore the evolutionary mode between Russian wheat aphid virulence and resistance genes of host plants.  相似文献

本研究以窄叶青8号(籼稻)×京系17(粳稻)的F1花培株系──DH群体为基础建立了1个水稻的RFLP连锁框架图,该图含RFLP标记、同功酶标记等共108个位点,标记间的平均间距为8.6cM。该图谱与已发表的用其他群体构建的图谱有很高的可比性。利用该框架图定位了2个未知位点的同功酶标记基因和1个籼稻亲本的抗稻瘟病基因。研究表明,目前的群体可进一步扩大成为一个永久性的作图群体,并应用于水稻基因定位和基因组研究  相似文献
