In Australia, in the past, pasture legumes were rotated mainly with cereals, but increasingly these rotations now involve pasture legumes with a wider range of crops, including legumes. This increasing frequency of the leguminous host in the rotation system may be associated with increased root rots in legumes in the current pasture-crop rotations. The primary aim of this study was to see whether the pathogenicity on pasture legumes of strains of Rhizoctonia solani sourced from lupins and cereals (common crops in rotation with pastures) is associated with increased incidence of root rots in pasture legumes in the disease conducive sandy soils of the Mediterranean regions of southern Australia. The second aim was to determine sources of resistance among newly introduced pasture legumes to R. solani strains originating from rotational crops as this would reduce the impact of disease in the pasture phase. Fifteen pasture legume genotypes were assessed for their resistance/susceptibility to five different zymogram groups (ZG) of the root rot pathogen R. solani under glasshouse conditions. Of the R. solani groups tested, ZG1–5 and ZG1–4 (both known to be pathogenic on cereals and legumes) overall, caused the most severe root disease across the genotypes tested, significantly more than ZG6 (known to be pathogenic on legumes), in turn significantly >ZG4 (known to be pathogenic on legumes) which in turn was >ZG11 (known to be pathogenic on legumes including tropical species). Overall, Ornithopus sativus Brot. cvs Cadiz and Margurita, Trifolium michelianum Savi. cvs Paradana and Frontier and T. purpureum Loisel. cv. Electro showed a significant level of resistance to root rot caused by R. solani ZG11 (root disease scores ≤1.2 on a 1–3 scale where 3 = maximum disease severity) while O. sativus cvs Cadiz and Erica showed a significant level of resistance to root rot caused by R. solani ZG4 (scores ≤1.2). O. compressus L. cvs Charano and Frontier, O. sativus cv. Erica, and T. purpureum cv. Electro showed some useful resistance to root rot caused by R. solani ZG6 (scores ≤1.8). This is the first time that cvs Cadiz, Electro, Frontier, Margurita and Paradana have been recognised for their levels of resistance to root rot caused by R. solani ZG11; and similarly for cvs Cadiz and Erica against ZG4; and for cvs Charano, Erica, and Electro against ZG6. These genotypes with resistance may also serve as useful sources of resistance in pasture legume breeding programs and also could potentially be exploited directly into areas where other rotation crops are affected by these R. solani strains. None of the tested genotypes showed useful resistance to R. solani ZG1–4 (scores ≥2.0) or ZG1–5 (scores ≥2.5). This study demonstrates the relative potential of the various R. solani ZG strains, and particularly ZG1–4, ZG1–5, ZG4 and ZG6 to attack legume pastures and pose a significant threat to non-pasture crop species susceptible to these strains grown in rotation with these pasture legumes. Significantly, the cross-pathogenicity of these strains could result in the continuous build-up of inoculum of these strains that may seriously affect the productivity eventually of legumes in all rotations. In particular, when choosing pasture legumes as rotation crops, caution needs to be exercised so that the cultivars deployed are those with the best resistance to the R. solani ZGs most likely to be prevalent at the location.  相似文献

During the past 40 years, more than 400 Sudden Unexplained Deaths (SUDs) have occurred in Yunnan, southwestern China. Epidemiological and toxicological analyses suggested that a newly discovered mushroom called Trogia venenata was the leading culprit for SUDs. At present, relatively little is known about the genetics and natural history of this mushroom. In this study, we analyzed the sequence variation at four DNA fragments among 232 fruiting bodies of T. venenata collected from seven locations. Our ITS sequence analyses confirmed that all the isolates belonged to the same species. The widespread presence of sequence heterozygosity within many strains at each of three protein-coding genes suggested that the fruiting bodies were diploid, dikaryotic or heterokaryotic. Within individual geographic populations, we found significant deviations of genotype frequencies from Hardy-Weinberg expectations, with the overall observed heterozygosity lower than that expected under random mating, consistent with prevalent inbreeding within local populations. The geographic populations were overall genetically differentiated. Interestingly, while a positive correlation was found between population genetic distance and geographic distance, there was little correlation between genetic distance and barium concentration difference for the geographic populations. Our results suggest frequent inbreeding, geographic structuring, and limited gene flow among geographic populations of T. venenata from southwestern China.  相似文献

Heterotrimeric G protein is involved in plant growth and development, while the role of rice (Oryza sativa) G protein γ subunit qPE9-1 in response to low-phosphorus (LP) conditions remains unclear. The gene expression of qPE9-1 was significantly induced in rice roots under LP conditions. Rice varieties carrying the qPE9-1 allele showed a stronger primary root response to LP than the varieties carrying the qpe9-1 allele (mutant of the qPE9-1 allele). Transgenic rice plants with the qPE9-1 allele had longer primary roots and higher P concentrations than those with the qpe9-1 allele under LP conditions. The plasma membrane (PM) H+-ATPase was important for the qPE9-1-mediated response to LP. Furthermore, OsGF14b, a 14-3-3 protein that acts as a key component in activating PM H+-ATPase for root elongation, is also involved in the qPE9-1 mediation. Moreover, the overexpression of OsGF14b in WYJ8 (carrying the qpe9-1 allele) partially increased primary root length under LP conditions. Experiments using R18 peptide (a 14-3-3 protein inhibitor) showed that qPE9-1 is important for primary root elongation and H+ efflux under LP conditions by involving the 14-3-3 protein. In addition, rhizosheath weight, total P content, and the rhizosheath soil Olsen-P concentration of qPE9-1 lines were higher than those of qpe9-1 lines under soil drying and LP conditions. These results suggest that the G protein γ subunit qPE9-1 in rice plants modulates root elongation for phosphorus uptake by involving the 14-3-3 protein OsGF14b and PM H+-ATPase, which is required for rice P use.  相似文献

应用控制授粉、软 X-射线法、常规石蜡制片法和荧光检测等手段,研究了鹅掌楸(Lirio-dendron chinense(Hemsl.)Sarg.胚胎发育和控制授粉与结籽率的相关性。控制授粉后2小时花粉萌发,6小时萌发率最高,柱头可授期持续30小时左右。花粉管借助于柱头毛之间的分泌物进入柱头沟,经花柱沟、珠孔塞和珠心冠原进入胚囊,行珠孔受精。授粉后2周,胚乳为2至3细胞厚的狭组织;第6周,胚乳充满胚囊腔,珠心随之解体殆尽;第7到8周,球形胚、心形胚发生;第14到16周,子叶形成;第22周种子或熟,胚乳丰富。单株自然授粉结籽率不足1%。控制授粉后,单个聚合果的最高结籽率可达39%,9个聚合果的平均结籽率为17.7%。  相似文献

高原鼢鼠挖掘取食活动的能量代价及其最佳挖掘取食行为   总被引:3,自引:1,他引:2  
高原鼢鼠(Myospalax baileyi)是青藏高原典型的地下鼠,它主要挖掘取食杂类草之地下根茎,在挖掘过程中形成取食洞道。本文根据此特点,建立了挖掘取食过程中能量代价(E_(?))与取食道结构参数(土丘间距S、侧道长L、取食道半径b及洞道深度D)之间以及取食获能E_(fg)与S和b之间的两个函数关系。分别以挖掘效率极大和取食效率极大为优化目标,导出同样的结果,即相同的最佳土丘间距S_(opt)。通过实验观测,计算出在矮嵩草(Kobresiahumilis)草甸生境条件下,最佳土丘间距S_(opt)=127.4厘米,与野外实测土丘间距(126.30±47.85厘米)无显著差异(t=0.312,P>0.50),表明高原鼢鼠的挖掘及取食行为符合最佳化原则。  相似文献

The pyridine nucleotide (NAD and NADP)-linked enzymes are a large class of enzymes constituting approximately 17% of all classified enzymes. When these enzymes catalyze their reactions, the hydride transfer between the substrate and the reaction site (i.e., C-4 of the nicotinamide/dihydronicotinamide ring) of the coenzyme takes place in a stereospecific manner. Thus, in the reaction of oxidation of the reduced coenzyme, one group of enzymes catalyzes the extraction of only the hydrogen having the R configuration at the No. 4 carbon, while the other group catalyzes the removal of only that with the S configuration. Because this aspect of enzyme stereospecificity provides essential information for a given enzyme's reaction mechanism, active site structure, and evolutionary relationship with other enzymes, intensive effort has been made to establish the stereospecificities of as many enzymes as possible. This review presents the compilation of the stereospecificities of these enzymes. Some empirical rules, which are useful but not definitive, in predicting a given enzyme's stereospecificity are also described. In addition, the stereospecificity in enzymatic reactions is compared to the stereo-preference in chemical oxidoreduction of the coenzyme. In order to elucidate the mechanism for the enzyme stereospecificity, the conformations of the coenzyme in free-state and enzyme-bound state are extensively discussed here.  相似文献

The rabbit genome encodes an opal suppressor tRNA gene. The coding region is strictly conserved between the rabbit gene and the corresponding gene in the human genome. The rabbit opal suppressor gene contains the consensus sequence in the 3' internal control region but like the human and chicken genes, the rabbit 5' internal control region contains two additional nucleotides. The 5' flanking sequences of the rabbit and the human opal suppressor genes contain extensive regions of homology. A subset of these homologies is also present 5' to the chicken opal suppressor gene. Both the rabbit and the human genomes also encode a pseudogene. That of the rabbit lacks the 3' half of the coding region. Neither pseudogene has homologous regions to the 5' flanking regions of the genes. The presence of 5' homologies flanking only the transcribed genes and not the pseudogenes suggests that these regions may be regulatory control elements specifically involved in the expression of the eukaryotic opal suppressor gene. Moreover the strict conservation of coding sequences indicates functional importance for the opal suppressor tRNA genes.  相似文献

棕色固氮菌中电子载体Fld直接向固氮酶铁蛋白传递电子。Fld_(ox)至Fld_R是双电子二步还原反应,极谱半波电位分别为-210、-550 mV。Fld_(ox)至Fld_(SR)的中点电位为-280 mV,Fld_(SR)至Fld_R为-500mV。铁蛋白中点电位为-256mV,加MgATP后为-390 mV。Fld_R与铁蛋白ox组成的电池电动势为244mV,电子传递可自发进行,反应的J△G~o为-23KJ/摩尔,铁蛋白被Fld_R还原的K_a=1.3×120~4,加入MgATP后△G~o为-10.6KJ/摩尔,K_a=72。因此,未加入MgATP时电子传递反应更易进行。  相似文献
