The use of penicillin G acylase (PGA) covalently linked to insoluble carrier is expected to produce major advances in pharmaceutical processing industry and the enzyme stability enhancement is still a significant challenge. The objective of this study was to improve catalytic performance of the covalently immobilized PGA on a potential industrial carrier, macroporous poly(glycidyl methacrylate‐co‐ethylene glycol dimethacrylate) [poly(GMA‐co‐EGDMA)], by optimizing the copolymerization process and the enzyme attachment procedure. This synthetic copolymer could be a very promising alternative for the development of low‐cost, easy‐to‐prepare, and stable biocatalyst compared to expensive commercially available epoxy carriers such as Eupergit or Sepabeads. The PGA immobilized on poly(GMA‐co‐EGDMA) in the shape of microbeads obtained by suspension copolymerization appeared to have higher activity yield compared to copolymerization in a cast. Optimal conditions for the immobilization of PGA on poly(GMA‐co‐EGDMA) microbeads were 1 mg/mL of PGA in 0.75 mol/L phosphate buffer pH 6.0 at 25°C for 24 h, leading to the active biocatalyst with the specific activity of 252.7 U/g dry beads. Chemical amination of the immobilized PGA could contribute to the enhanced stability of the biocatalyst by inducing secondary interactions between the enzyme and the carrier, ensuring multipoint attachment. The best balance between the activity yield (51.5%), enzyme loading (25.6 mg/g), and stability (stabilization factor 22.2) was achieved for the partially modified PGA. © 2015 American Institute of Chemical Engineers Biotechnol. The temperature dependence of delayed millisecond fluorescence (DF) induction curve transients was investigated. The transients were obtained by keeping intact maize leaf segments in the dark for 30 to 300 s before illumination. The temperature dependence of DF induction revealed abrupt changes in activation energy of the recombination process. Those changes highly overcome the lowest energy barriers for certain types of chemical reactions, implying significant possible alterations in the DF mechanism itself. On the other hand, the particular transients responses expressed some specificities. Both the electrochemical gradient controlled changes, and the temperature induced changes contribute to it. 相似文献

Confocal fluorescence microscopy was used to examine the spectral characteristics of lignin autofluorescence in secondary cell walls of normal and compression wood from Pinus radiata. Using UV excitation, fluorescence spectra of normal and compression wood sections showed significant differences, especially in the outer secondary cell wall of tracheids, with a shift in maxima from violet to blue wavelengths between normal and compression wood. A comparison of normal wood, mild and severe compression wood, showed that the wavelength shift was intermediate in the mild compression wood compared to the severe compression wood, thus offering the possibility of quantifying the severity by measuring ratios of fluorescence at violet and blue wavelengths. Fluorescence induced by blue light, rather than UV, was less well differentiated amongst wood types. Spectral deconvolution indicated the presence of a minimum of five discrete lignin fluorophores in the cell walls of both normal and compression wood tracheids. Comparison with lignin model compounds suggest that the wavelength shift may correspond in part to increased levels of p-hydroxy type lignin in the compression wood samples. The combination of confocal fluorescence imaging and related spectral deconvolution therefore offers a novel technique for characterising cell wall lignin in situ. 相似文献

The perinuclear theca (PT) of mammalian sperm is a unique subcellular structure encapsulating the nucleus. Compositionally, the PT is made up of at least six prominent polypeptides (60, 36, 31, 28, 24, and 15 kDa), of which only two have been sequence identified, as well as many less prominent ones. As an ongoing process in unveiling the protein composition of the PT, we have uncovered the sequence identity of the prominent 24-kDa polypeptide (PT24). Initial N-terminal sequence analysis obtained by Edman degradation suggested that PT24 is a RAB2 protein. This was corroborated by mass spectrometric analyses of trypsin-digested fragments of PT24, identifying RAB2A of the RAB2 subfamily as the best sequence match. Quadrapole/time-of-flight analysis identified 72%% sequence coverage between PT24 and bull, human, mouse, or rabbit RAB2A. Since a genome search only identified two RAB2 subfamily members, RAB2A and RAB2B, the 72%% sequence coverage of PT24 provides assurance that this protein is RAB2A and not a new RAB2 subfamily member. Furthermore, commercial RAB2A antibodies, raised against oligopeptide fragments in the unique C-terminal region of RAB2A, specifically labeled PT24 on Western blot analysis of PT extracts. These anti-RAB2A antibodies, along with immune serum that we raised and affinity purified against isolated PT24, demonstrated at both light and electron microscope levels that RAB2 is associated with the periphery of the growing proacrosomic and acrosomic vesicles in the Golgi and cap phases of spermiogenesis and consequently assembled as part of the PT. This pattern of subacrosomal assembly is reminiscent of the pathway used by SubH2Bv (PT15), another prominent and exclusive subacrosomal protein, indicating a common route for subacrosomal-PT assembly. Traditionally somatic RAB2 proteins are involved in vesicular transport between the endoplasmic reticulum and the cis-side of the Golgi apparatus. Our study suggests an unprecedented direction of RAB2A-mediated vesicular transport in spermatids during acrosomal biogenesis, from the trans-side of the Golgi apparatus to the nuclear envelope. 相似文献

Differential gene expression is the major mechanism underlying the development of specific body regions. Here we assessed the role of genes differentially expressed in the Drosophila wing imaginal disc, which gives rise to two distinct adult structures: the body wall and the wing. Reverse genetics was used to test the function of uncharacterized genes first identified in a microarray screen as having high levels of expression in the presumptive wing. Such genes could participate in elaborating the specific morphological characteristics of the wing. The activity of the genes was modulated using misexpression and RNAi-mediated silencing. Misexpression of eight of nine genes tested caused phenotypes. Of 12 genes tested, 10 showed effective silencing with RNAi transgenes, but only 3 of these had resulting phenotypes. The wing phenotypes resulting from RNAi suggest that CG8780 is involved in patterning the veins in the proximal region of the wing blade and that CG17278 and CG30069 are required for adhesion of wing surfaces. Venation and apposition of the wing surfaces are processes specific to wing development providing a correlation between the expression and function of these genes. The results show that a combination of expression profiling and tissue-specific gene silencing has the potential to identify new genes involved in wing development and hence to contribute to our understanding of this process. However, there are both technical and biological limitations to this approach, including the efficacy of RNAi and the role that gene redundancy may play in masking phenotypes. 相似文献

An analysis of theoretical knowledge and experimental results of ultraweak luminescence (UWL) is provided. The role of excited state of molecules and free radicals, formed in various biochemical reactions, in UWL is discussed. UWL of model reactions and in vivo systems are compared. The hypothesis of coherent electromagnetic field as a source of UWL is also discussed. Spectral, kinetic and temporal properties of UWL are summarised, as well as their connection with its origin and role in the organism. Attention is paid to recent progress in experimental methods of low-light detection. The possible use of UWL in environmental studies, selection and other applications is discussed. 相似文献

Lignin, the structural polymer of the plant cell walls, is produced by free radical polymerization of phenolic alcohols, catalyzed by different peroxidases. The mechanism and the structural organization of lignin in the cell have not been completely understood. In this study we applied fractal analysis to images of lignin polymer obtained using scanning tunneling microscope. The analysis showed the regularity of the polymer at different levels of organization. According to the results obtained, at the 95% confidence level, there is no significant difference in the fractal dimension between images representing different organizational levels of lignin. In other words, lignin produced in in vitro conditions has fractal structural organization and, consequently the polymer can be expected to be regular in in vivo conditions. The value of the fractal dimension 1.929 +/- 0.021 is in good agreement with the theoretically predicted value for polyaddition and polycondensation mechanism of polymerization. The mechanism of in vivo lignin synthesis is discussed in terms of various experimental and theoretical evidences. In this paper, we could show that fractal analysis of the lignin polymer is a useful complementary approach to the experimental data collection in structural and phenomenological studies. 相似文献

Fractal analysis was applied to images of photochemical lignin polymer obtained using scanning tunneling microscope. We studied the polymer obtained in vitro by ionic mechanism through UV radiation--induced polymerization. The analysis showed the regularity of the lignin-like polymer at different levels of organization. At the 95% confidence level, there was no significant difference in the fractal dimension between images representing different organizational levels of photochemical lignin. That means that lignin produced in in vitro conditions by photochemical mechanism of synthesis, has a fractal structural organization. The obtained values of the fractal dimension are in good agreement with the theoretically predicted value for the polyaddition and polycondensation mechanism of polymerization, known as the bulk model. 相似文献

Due to the impact of anthropogenic activities on forest extent and integrity across Madagascar, it is increasingly necessary to assess how endangered lemur populations inhabiting human-dominated forest fragments can effectively sustain themselves ecologically. Our research addresses this concern by exploring how the distribution patterns of a small population of crowned lemurs (Eulemur coronatus), occupying a degraded forest fragment at Oronjia Forest New Protected Area in northern Madagascar, are impacted by the availability of key ecological and anthropogenic factors. We hypothesize that the distribution of E. coronatus within the fragment is limited by the availability of critical ecological resources and conditions and the intensity of anthropogenic features and activities. To examine this, we used MaxEnt to develop a species distribution model using presence-only occurrence records and 10 independent background covariates detailing the site's ecological and anthropogenic aspects. The results indicate that the realized distribution patterns of E. coronatus within human-dominated forest fragments are strongly associated with sections of forest that contain sparsely and sporadically distributed resources, such as freshwater and continuous hardwood vegetation. We conclude that the distribution of E. coronatus at Oronjia is shaped by their need to maximize foraging opportunities in a degraded forest landscape where they are subject to both environmental and anthropogenic stressors. 相似文献
