Storage in dimethyl sulphoxide (DMSO) of Escherichia coli K12 hybrids carrying nif+ genes from Klebsiella pneumoniae can result in selection of a defective nitrogen-fixing phenotype. Similar results are obtained with E. coli K12 hybrids containing the nitrogen-fixing capacity from Rhizobium trifolii. DMSO appears to affect particular inner membrane proteins associated with energy metabolism in E. coli K12 and four chromosomal regions (chlD, chlG, his and unc) are associated with resistance to DMSO.  相似文献

Conjugal transfer of three IncP1 plasmids and one IncFII plasmid into strains of the ethanol-producing bacterium Zymomonas mobilis was obtained. These plasmids were transferred at high frequencies from Escherichia coli and Pseudomonas aeruginosa into Z. mobilis and also between different Z. mobilis strains, using the membrane filter mating technique. Most of the plasmids were stably maintained in Z. mobilis, although there was some evidence of delayed marker expression. A low level of chromosomal gene transfer, mediated by plasmid R68.45, was detected between Z. mobilis strains. Genetic evidence suggesting that Z. mobilis may be more closely related to E. coli than to Pseudomonas or Rhizobium is discussed.  相似文献

Several strains of Rhizobium resistant to spectinomycin also had nitrogenase activity (C2H2 reduction and H2 production) in static culture under 95% Ar/1%O2/4%C2H2. This relationship between nitrogenase activity and spectinomycin resistance was observed in both fast-growing (R. trifolii and R. leguminosarum) and slow-growing (R. japonicum) rhizobia. The effect of different media and various carbon sources on nitrogenase activity was investigated in more detail in R. trifolii strain TlSp. This communication demonstrates that fast-growing rhizobia can have nitrogenase activity in the absence of any plant component.  相似文献

Strain T1K, reported to be Rhizobium trifolii strain T1 carrying the drug resistance plasmid RU-1drd, was able to transfer a cluster of nif+ genes to Escherichia coli K-12. Additional genetic material, resembling the gal-chlA region of E. coli, was also transferred from strain T1K. The segregation pattern of these transferred genes suggested that they were on a plasmid. Although strain TIK was able to nodulate red and white clover, it also formed very slow-growing galls on tomato stems and shared many physiological properties with Agrobacterium tumefaciens, to which it seemed more closely related than to R. trifolii. The R. trifolii hybrid T1 (R1-19drd), constructed by conjugation, did not share any of these properties of both A. tumefaciens. Thus, strain T1K appears to be a bacterium with properties of both A. tumefaciens and R. trifolii and with the capacity to transfer nif+ genes and other functions which it may have "cloned" from another bacterium such as Klebsiella.  相似文献

OBJECTIVE: To analyze cytologic and histologic parameters in bone marrow trephine biopsy in an attempt to define heterogeneity of hairy cell leukemia cells. STUDY DESIGN: The study group consisted of 28 trephine biopsies. Immunohistochemistry for CD20 antigen was used. Image processing and measurements were performed with AnalySIS 3.0 image analysis system (Soft Imaging System GmbH, Germany) and custom built programs. For planimetric measurements of nuclei, automatic segmentation was implemented. The measured parameters were: surface area, perimeter, minimum, mean and maximum diameter, and a set of form factors. Relative volumes of bone trabeculae, adipose tissue, hematopoietic tissue and neoplastic infiltrate were assessed by the point counting method. Nuclear volume was measured by the point sampled intercept method. Bone marrow fibrosis was assessed using a curvilinear line test system. RESULTS: Significant variability of cell nuclei was found, and their classification into 3 types was possible. The relative frequency of those types was different in various cases and allowed subdivision of cases into 3 groups that differed in some clinical and histologic manifestations. CONCLUSION: The present study demonstrated the heterogeneity of cell populations of hairy cell leukemia.  相似文献

Using steel pins inserted into growing moss colonies near Casey Station, Wilkes Land, continental Antarctica, we have measured the growth rate of three moss species: Bryum pseudotriquetrum and Schistidium antarctici over 20 years and Ceratodon purpureus over 10 years. This has provided the first long-term growth measurements for plants in Antarctica, confirming that moss shoots grow extremely slowly in Antarctica, elongating between 1 and 5 mm per year. Moss growth rates are dependent on availability of water. Antheridia were observed on some stems of B. pseudotriquetrum; no archegonia or sporophytes were observed. Stems bearing antheridia elongated much more slowly than vegetative stems in the same habitat. Two other methods of growth rate measurement were tested, and gave similar rates of elongation over shorter periods of time. However, for long-term measurements, the steel pin measurements proved remarkably reproducible and reliable.  相似文献

Sixty-six isolates of the moss Sarconeurum glaciale were collected from sites in continental Antarctica at Ross Island, southern Victoria Land and the Vestfold Hills. Genetic variation within and among the populations was estimated using isozymes and random amplified polymorphic DNA (RAPD) technology. Isozyme results only reproducibly showed variation between the populations with one enzyme; RAPDs indicated significantly higher levels of genetic variability within and among the Vestfold Hills samples than in the Ross Sea region samples. A dendrogram produced from the RAPD bands suggested that the Ross Island and southern Victoria Land samples form one population, and those from the Vestfold Hills form a separate and more variable population, possibly resulting from separate colonisation events on the continent. The extent of genetic variation and dispersal mechanisms were investigated over short distances of 1–100 m, and up to 3 km, by the random amplified polymorphic DNA (RAPD) technique, for the moss Sarconeurum glaciale , at three locations on Ross Island, Antarctica. At Arrival Heights, genetic variation occurred within single colonies, and the relationships between clumps indicated that they were dispersed down small, meltwater drainage channels by water. The genetic similarities between the colonies from Arrival Heights and others from Scott Base and Crater Hill, a few km away, together with the prevailing wind direction and absence of this moss in the intervening snow-covered area, suggested longer-distance dispersal by wind. Overall, the Ross Island samples appeared to form a single, polymorphic population that was distinct from another population at Canada Glacier, 110 km distant. Somatic mutation, rather than immigration of genetically different propagules from elsewhere, appeared to be the most probable cause of genetic variability in these haploid, vegetatively reproducing Antarctic moss populations. Initiation of recolonization of S. glaciale across a dirt track at Arrival Heights was also investigated by RAPDs, to investigate how regrowth of mosses in disturbed areas occurred in the extreme environment of Antarctica.  相似文献

The Random Amplified Polymorphic DNA technique was used to assess the level of genetic diversity in Bryum argenteum from Ross Island and southern Victoria Land, Antarctica. Samples were collected from three separate transects, and from other geographically distinct populations within 150 km of Ross Island. Moss growth in two transects, sampled down small exposed meltstream channels at Cape Royds and Cape Chocolate, was very sparse with no other moss colonies found within 0.4 or 4 km, respectively. However, samples from these channels showed similar levels of genetic variation to those from a transect at Granite Harbour, where moss colonies were large, luxuriant and turf-like between boulders. In all transects, high levels of genetic diversity were apparent both within and between colonies, and some spatial relationships were observed down the length of the channels, with more extensive variation at the top than the bottom of two transects. Samples from other sites in the region showed varying but high levels of genetic diversity; overall, the majority showed some clustering according to site of collection, with short-distance dispersal of propagules by water and transmission between sites presumably by wind. The extensive genetic diversity observed appears mainly due to somatic mutation within colonies, with some contribution by immigration of propagules from elsewhere into established colonies.
