Two closely related pseudoisocyanins, N,N'-diethyl-6,6'-dichlorpseudoisocyanin chloride and N, N'-diethylpseudoisocyanin chloride, were tested for their metachromatic staining behavior with oxidized insulin. N,N'-diethyl-6,6-dichlorpseudoisocyanin chloride gave nonspecific metachromasia with collagen, mucus, and mast cells of adult tissues; almost all tissues of rat embryos exhibited nonspecific staining. Nonspecific reactions were rarely observed in adult or fetal tissues with the extremely labile metachromasia of N, N'-diethylpseudoiso-cyanin chloride. When oxidation time and temperatures are carefully controlled, this reagent apears to be highly specific for insulin-containing cells and can be used as a selective stain for beta cells. Paraffin sections of formalin fixed material were oxidized 45 sec at 28-29 C in freshly prepared acidified permanganic (2.5% KMnO4, 1; 5% H2SO4, 1; distilled water, 7—parts by volume), decolorized 30 sec in 5% oxalic acid, and washed 5 min in running tap water. After rinsing in 2 changes of distilled water, sections were stained 20 min in a 36 mg/100 ml aqueous solution of N, N'-diethylpseudoisocyanin chloride. Sections were then washed in running tap water until the albumen adhesive was decolorized, and mounted in Karo syrup diluted with an equal amount of distilled water. The insulin-containing cells are stained light to dark purple; all other tissue components, various shades of red. N, N'-diethylpseudoisocyanin chloride was used as a reference for evaluating the specificity of 5 commonly used empirical methods for demonstrating alpha and beta cells in pancreatic islets. Cells exhibiting pseudo isocyanin metachromasia were stained selectively by aldehyde-fuchsin, Heidenhain's azan, and chrome-hematoxylin. Aldehyde-Iuchsin was the only empirical stain tested which gave results comparable to pseudoisocyanin for clarity and definition of beta cells. After oxidation in acidified permanganate, azocarmine and phosphotungstic acid-hematoxylin differentially stained alpha cells; cells demonstrated by these two methods did not exhibit pseudoisocyanin metachromasia. This histochemical procedure can precede empirical methods which require preliminary oxidation in acidified permanganate or it can follow empirical methods which do not extract the insulin nor alter its intramolecular disulfide bonds. 相似文献
Folate polyglutamate and monoglutamate accumulation was measured in normal diploid and SV40-transformed human fibroblasts by Sephadex G-10 gel filtration chromatography. The cells were first depleted of folates and then provided with limiting amounts of [3H]-folic acid in order that the cells would accumulate only forms of folate necessary for proliferation. Both the normal and the transformed cells accumulated monoglutamate and polyglutamate forms, but by 72 hours of labeling the transformed cells contained 3-10 times more polyglutamate than the normal cells. The growth rates for the normal and transformed cells were similar at this limiting folic acid concentration. Thus, if folate polyglutamates are more important for the proliferation of SV40-transformed cells than the normal cells, then inhibition of polyglutamate formation may be an important potential target for chemotherapy. 相似文献
Typically, laboratory studies on the physiological effects of temperature are conducted using stable acclimation temperatures. Nonetheless, information extrapolated from these studies may not accurately represent wild populations living in thermally variable environments. The aim of this study was to compare the growth rate, metabolism and swimming performance of wild Atlantic salmon exposed to cycling temperatures, 16–21°C, and stable acclimation temperatures, 16, 18.5, 21°C. Growth rate, metabolic rate, swimming performance and anaerobic metabolites did not change among acclimation groups, suggesting that within Atlantic salmon's thermal optimum range, temperature variation has no effect on these physiological properties. 相似文献
The mammalian target of rapamycin complex 1 (mTORC1) integrates nutrients, growth factors, stress, and energy status to regulate cell growth and metabolism. Amino acids promote mTORC1 lysosomal localization and subsequent activation. However, the subcellular location or interacting proteins of mTORC1 under amino acid‐deficient conditions is not completely understood. Here, we identify ADP‐ribosylation factor GTPase‐activating protein 1 (ArfGAP1) as a crucial regulator of mTORC1. ArfGAP1 interacts with mTORC1 in the absence of amino acids and inhibits mTORC1 lysosomal localization and activation. Mechanistically, the membrane curvature‐sensing amphipathic lipid packing sensor (ALPS) motifs that bind to vesicle membranes are crucial for ArfGAP1 to interact with and regulate mTORC1 activity. Importantly, ArfGAP1 represses cell growth through mTORC1 and is an independent prognostic factor for the overall survival of pancreatic cancer patients. Our study identifies ArfGAP1 as a critical regulator of mTORC1 that functions by preventing the lysosomal transport and activation of mTORC1, with potential for cancer therapeutics. 相似文献
Seed predation may influence community assembly and invasion dynamics when seed predators preferentially select some seed species over others. However, the role of different seed predators and their preferences for seeds in multiple ecological contexts, including the endangered California sage scrub ecosystem, remain unresolved making predictions about which processes limit or promote invasions difficult. In addition, selection criteria, specifically how seed size and biogeographic origin (native versus invasive) influences selection, requires further research across multiple granivore guilds. We examined seed predator preference for common native and invasive seeds across small and large seed classes using seed dish experiments with motion-sensor video observation. We also quantified the relative importance of ants, birds, and mammals as seed predators in sage scrub. Community-wide, granivores preferred the small-seeded invasive mustard Brassica nigra, avoided the large-seeded invasive grass Bromus rubens and the native large-seeded shrub Encelia californica, and did not show significant selection or avoidance for the small-seeded native shrub Salvia apiana. Birds, notably California towhees (Melozone crissalis), were the most frequent granivore while rodents and ants rarely removed seeds. This study reveals that birds are important seed predators in California sage scrub and have the potential to contribute to biotic resistance to mustard invasions (Brassica nigra and potentially others).
