The sky islands of southeastern Arizona (AZ) mark a major transition zone between tropical and temperate biota and are considered a neglected biodiversity hotspot. Dispersal ability and host plant specificity are thought to impact the history and diversity of insect populations across the sky islands. We aimed to investigate the population structure and phylogeography of two pine‐feeding pierid butterflies, the pine white (Neophasia menapia) and the Mexican pine white (Neophasia terlooii), restricted to these “islands” at this transition zone. Given their dependence on pines as the larval hosts, we hypothesized that habitat connectivity affects population structure and is at least in part responsible for their allopatry. We sampled DNA from freshly collected butterflies from 17 sites in the sky islands and adjacent high‐elevation habitats and sequenced these samples using ddRADSeq. Up to 15,399 SNPs were discovered and analyzed in population genetic and phylogenetic contexts with Stacks and pyRAD pipelines. Low genetic differentiation in N. menapia suggests that it is panmictic. Conversely, there is strong evidence for population structure within N. terlooii. Each sky island likely contains a population of N. terlooii, and clustering is hierarchical, with populations on proximal mountains being more related to each other. The N. menapia habitat, which is largely contiguous, facilitates panmixia, while the N. terlooii habitat, restricted to the higher elevations on each sky island, creates distinct population structure. Phylogenetic results corroborate those from population genetic analyses. The historical climate‐driven fluxes in forest habitat connectivity have implications for understanding the biodiversity of fragmented habitats.  相似文献

Marine tubificids are abundant and diverse in the carbonate sediments of Bermuda, as well as in many other tropical and subtropical locations. Recently, during microscopic observations of living specimens, crystal-like structures were observed attached to the coelomic peritoneum and in the coelomic cavity of some Bermuda species, including phallodrilines of the genera Aktedrilus and Pectinodrilus,and a rhyacodriline of the genus Heterodrilus. Similar structures were not seen in tubificid species of Thallasodrilides and other limnodriloidines, a second species of Heterodrilus, a tubificine of the genusTubificoides, a phallodriline of the genus Bathydrilus,nor in a number of marine enchytraeid genera and species found in Bermuda. The crystal-like structures have two needle arms, each about 5–10 m long and about 0.5 m in diameter, meeting at an obtuse angle. At the junction of the arms, there is a small membrane-bound `knob', about 1 m in diameter, which may be continuous with the coelomic peritoneum. The numbers of `crystals' per individual worm are estimated at 100–400 per body segment, or well over 2 × 103 in an adult worm. `Crystals' are found: throughout the length of the worms, in all individuals of species in which `crystals' occur, and over the range of environmental conditions where these species are found in Bermuda. Simple digestions with hypochlorite, weak and dilute acids, and staining with nuclear and cytoplasmic stains indicate that the composition of the knob is organic and the arms inorganic. The fluorescent tracer Calcein (Sigma) was not incorporated into any structures during a 24-h bath incubation of living worms, and the `crystals' do not show birefringence when viewed between crossed polarizing filters. These last two results do not support an hypothesis that these are calcium carbonate `crystals'. Geographically, the crystal-like structures are widespread, and have also been observed in a species of immature (unidentified) marine tubificid from Rottnest Island, Western Australia.  相似文献

Virgin females of the subtropical pierid butterfly Eurema daira were observed to actively solicit male courtship. The resulting interactions involved both aggressive contact with male individuals and elaborate female posturing. Cage-based trials showed that there was a direct relationship between the frequency of courtship solicitation attempts and female age. Virgin females under 3 days old showed little or no interest in initiating courtship. By contrast, 23.2 percent of the virgin females over three days old actively solicited available males with peak solicitation behavior exhibited from five day old individuals.  相似文献

Large numbers of cells will be required for successful embryonic stem cell (ESC)-based cellular therapies or drug discovery, thus raising the need to develop scaled-up bioprocesses for production of ESCs and their derived progeny. Traditionally, ESCs have been propagated in adherent cultures in static flasks on fibroblasts layers in serum-containing medium. Direct translation of two-dimensional flatbed cultures to large-scale production of the quantities of cells required for therapy simply by increasing the number of dishes or flasks is not practical or economical. Here, we describe successful scaled-up production of ESCs on microcarriers in a stirred culture system in a serum-free medium. Cells expanded on CultiSpher S, Cytodex 3, and Collagen microcarriers showed superior cell-fold expansions of 439, 193, and 68, respectively, without excessive agglomeration, compared with 27 in static culture. In addition, the ESCs maintained their pluripotency after long-term culture (28 days) in serum-free medium. This is the first time mESCs have been cultured on microcarriers without prior exposure to serum and/or fibroblasts, while also eliminating the excessive agglomeration plaguing earlier studies. These protocols provide an economical, practical, serum-free means for expanding ESCs in a stirred suspension bioprocess.  相似文献

Dark O2 consumption by the green alga Selenastrum minutum was sensitive to inhibition by the cytochrome pathway respiration inhibitor cyanide in the absence of an alternative oxidase inhibitor, consistent with previous work that suggested that this alga lacks alternative oxidase capacity. In contrast, addition of low concentrations of the cytochrome pathway inhibitor azide (50–750 μ M ) resulted in a stimulation of dark O2 consumption, while higher concentrations of azide (1–2 m M ) partially inhibited O2 consumption. Measurements of changes in cellular levels of pyruvate, malate and pyridine nucleotides upon cyanide addition were consistent with the absence of alternative oxidase capacity, and suggested that cyanide inhibition of O2 consumption was not due to nonspecific effects of cyanide. Addition of salicylhydroxamic acid (SHAM) also resulted in an increase in the rate of O2 consumption. Both azide- and SHAM-stimulated O2 consumption were sensitive to inhibition by 50 m M ascorbate or by cyanide. However, the ubiquinone analogs chloroquine and quinacrine specifically inhibited azide-stimulated O2 consumption, with only minor effects on SHAM-stimulated O2 consumption. These results suggest that azide-stimulated O2 consumption was not mediated by the previously characterized SHAM-stimulated oxidase, and are consistent with the possibility that azide-stimulated O2 consumption is mediated by a plasma membrane redox system.  相似文献

Background

Expert opinion recommends performing exercise testing with initiation of Class Ic antiarrhythmic medication.

Objective

To evaluate the rate and reason for discontinuation of Ic agent within the first year of follow up, with particular attention to rate of proarrhythmia and the value of routine treadmill testing.

Methods

This is a single center retrospective cohort study including consecutive patients with atrial arrhythmias who were initiated on a Class Ic agent from 2011 to 2016. Data was collated from chart review and pharmacy database.

Results

The study population included 300 patients (55% male, mean age 61; mean ejection fraction, 56%) started on flecainide (n = 153; 51%) and propafenone (n = 147; 49%). Drug initiation was completed while hospitalized on telemetry and the staff electrophysiologists directed dosing. There was one proarrhythmic event during initiation (0.3%). The primary reason for not being discharged on Ic agent was due to detection of proarrhythmia (n = 15) or ischemia (n = 1) with treadmill testing (5.3%). Exercise testing was the single significant variable to affect the decision to discontinue Ic drug, p < 0.0001 (95% CI: 1.89–6.08%). During follow up, the primary reason for discontinuation of Ic agent was lack of efficacy, 32%.

Conclusions

With proper screening, initiation of Class Ic agent is associated with very low rate of proarrhythmia. Treadmill testing is of incremental value and should be completed in all patients after loading Class Ic antiarrhythmic.  相似文献

The unicellular green alga Chlamydomonas reinhardtii Dang. displays a high capacity for salicylhydroxamic acid (SHAM)—stimulated O2 consumption, mediated by extracellular peroxidaie. Addition of exogenous NADH also resulted in stimulation of O2 consumption. The SHAM-and NADH-stimulated peroxidase activity was partially sensitive to inhibition by exogenous superoxide dismutase, ascorbate, and gentisic acid. These compounds did not inhibit O2 consumption in the absence of effectors. SHAM-and NADH-stimulated peroxidase activity also was sensitive to inhibition by cyanide, and cyanide titration curves indicated that O2 consumption by peroxidase was more cyanide-sensitive than O2 consumption by cytochrome oxidase. The differential sensitivity to cyanide was used to estimate partitioning of O2 consumption between mitochondrial respiration and extracellular peroxidase. We suggest that, despite a large capacity for peroxidase-me-diated O2 consumption, peroxidase did not consume O2 at detectable rates in the absence of effectors. Therefore, in the absence of effectors, measured rates of O2 consumption represented the rate of mitochondrial respiration .  相似文献

The Miami blue butterfly (Cyclargus thomasi bethunebakeri) is a state-endangered taxon in Florida and a candidate for federal listing. Here we report 12 polymorphic microsatellite loci appropriate for use in population and conservation studies. We genotyped 114 individuals sampled from a metapopulation in the lower Florida Keys over a 2-year period (2005-2006). These results show 4-14 alleles per locus, and ranges of observed and expected heterozygosities are 0.02679-0.79630 and 0.06154-0.69565, respectively. Large deviations from Hardy-Weinberg equilibrium (HWE) are observed across the whole sample set. When a single breeding population is analysed alone, seven of the loci are in HWE.  相似文献

The green alga Chlamydomonas reinhardtii Dangeard CW-15 exhibited very low rates of plasma-membrane Fe(III) reductase activity when grown under Fe-sufficient conditions. After switching the medium to an Fe-free formulation, both ferricyanide reductase and ferric chelate reductase activities rapidly increased, reaching a maximum after 3 d under iron-free conditions. Both of the Fe(III) reductase activities increased in parallel over time, they exhibited similar K m values (approximately 10 μM) with respect to Fe(III), displayed the same pH profile of activity, and both exhibited the same degree of light stimulation which could be inhibited by 3-(3′,4′-dichlorophenyl)-1,1-dimethylurea (DCMU). Furthermore, ferricyanide competitively inhibited ferric chelate reduction by iron-limited cells. These results indicate that both Fe(III) reductase activities were mediated by the same iron-limitation-induced plasma-membrane reductase. No evidence was found for the presence of Fe(III)-reducing substances in the culture medium, or for the involvement of active oxygen species in the process of Fe(III) reduction. Pulmonary surfactant has two distinct functions within the lung: reduction of surface tension at the air-liquid interface and participation in innate host defense. Both functions are dependent on surfactant-associated proteins. Pseudomonas aeruginosa is primarily responsible for respiratory dysfunction and death in cystic fibrosis patients and is also a leading pathogen in nosocomial pneumonia. P. aeruginosa secretes a number of proteases that contribute to its virulence. We hypothesized that P. aeruginosa protease IV degrades surfactant proteins and results in a reduction in pulmonary surfactant host defense and biophysical functions. Protease IV was isolated from cultured supernatant of P. aeruginosa by gel chromatography. Incubation of cell-free bronchoalveolar lavage fluid with protease IV resulted in degradation of surfactant proteins (SP)-A, -D, and -B. SPs were degraded in a time- and dose-dependent fashion by protease IV, and degradation was inhibited by the trypsin-like serine protease inhibitor Nalpha-p-tosyl-L-lysine-chloromethyl ketone (TLCK). Degradation by protease IV inhibited SP-A- and SP-D-mediated bacterial aggregation and uptake by macrophages. Surfactant treated with protease IV was unable to reduce surface tension as effectively as untreated surfactant, and this effect was inhibited by TLCK. We speculate that protease IV may be an important contributing factor to the development and propagation of acute lung injury associated with P. aeruginosa via loss of surfactant function within the lung.  相似文献
