The importance of sexual selection in population divergence is of much interest, mainly because it is thought to cause reproductive isolation and hence could lead to speciation. Sexually selected traits have been hypothesized to diverge faster between populations than other traits, presumably because of differences in the strength, mechanism or dynamics of selection. We investigated this by quantifying population divergence in eight morphological characters in 12 south Swedish populations of a sexually dimorphic damselfly, the banded demoiselle (Calopteryx splendens). The morphological characters included a secondary sexual character, the male melanized wing spot, which has an important function in both inter- and intrasexual selection. In addition, we investigated molecular population divergence, revealed by amplified fragment length polymorphism (AFLP) analysis. Molecular population divergence was highly significant among these Northern European populations (overall F(st)=0.054; pairwise population F(st)'s ranged from approximately 0 to 0.13). We found evidence for isolation-by-distance (r=0.70) for the molecular markers and a significant correlation between molecular and phenotypic population divergence (r=0.39). One interpretation is that population divergence for the AFLP loci are affected by genetic drift, but is also indirectly influenced by selection, due to linkage with loci for the phenotypic traits. Field estimates of sexual and natural selection from two of the populations revealed fairly strong sexual selection on wing spot length, indicating that this trait has the potential to rapidly diverge, provided that variation is heritable and the observed selection is chronic. 相似文献

Isothermal titration calorimetry and surface plasmon resonance were tested for their ability to study substrate binding to the active site (AS) and to the secondary binding site (SBS) of Bacillus subtilis xylanase A separately. To this end, three enzyme variants were compared. The first was a catalytically incompetent enzyme that allows substrate binding to both the AS and SBS. In the second enzyme, binding to the SBS was impaired by site-directed mutagenesis, whereas in the third enzyme, the AS was blocked using a covalent inhibitor. Hermit beetles of the genus Osmoderma (Coleoptera: Scarabaeidae: Cetoniinae) are known for their fruity odour, which is released in large amounts by males. Two species of the genus occur in Europe, the eastern Osmoderma barnabita (Motschulsky) and the western Osmoderma eremita (Scopoli). Previous studies on Swedish populations of O. eremita showed that the compound responsible for the characteristic scent, γ‐decalactone, functions as a sex pheromone for the attraction of conspecific females. Male O. eremita only release the (R)‐enantiomer of the lactone, and both sexes are anosmic to the opposite enantiomer. As the distribution areas of the two hermit beetle species partly overlap, it may be expected that they use different enantiomeric compositions of γ‐decalactone as pheromones to promote species discrimination. This paper reports on the identification of the sex pheromone of O. barnabita. Surprisingly, males from a Polish population produce only the (R)‐enantiomer of γ‐decalactone, and conspecific females show equal attraction to the (R)‐enantiomer and a racemic mixture of the compound, indicating that O. barnabita is anosmic to the (S)‐enantiomer, similarly to what was observed for O. eremita. A mtDNA sequence analysis of the cytochrome oxidase subunit I gene of Polish and Swedish beetles confirmed their taxonomical status as O. barnabita and O. eremita, respectively, with an average sequence divergence of 10.5% between beetles from the two studied areas. Although genetic data suggest that these species diverged several million years ago, they still rely on the same enantiomer of γ‐decalactone for mate finding. Thus, the male‐produced pheromone in Osmoderma spp. may be regarded as a territorial signal being exploited by females, rather than a cue for determining species identity. Our data show that the same compound can be used to facilitate monitoring of both beetle species, which are considered indicator species of the species‐rich fauna of saproxylic insects in Europe. 相似文献

A number of studies have previously examined the capacity of intracellular Leishmania parasites to modulate the capacity of macrophages to process and present Ags to MHC class II-restricted CD4(+) T cells. However, the bulk culture approaches used for assessing T cell activation make interpretation of some of these studies difficult. To gain a more precise understanding of the interaction between Leishmania-infected macrophages and effector T cells, we have analyzed various parameters of T cell activation in individual macrophage-T cell conjugates. Leishmania-infected macrophages efficiently stimulate Ag-independent as well as Ag-dependent, TCR-mediated capping of cortical F-actin in DO.11 T cells. However, infected macrophages are less efficient at promoting the sustained TCR signaling necessary for reorientation of the T cell microtubule organizing center and for IFN-gamma production. A reduced ability to activate these T cell responses was not due to altered levels of surface-expressed MHC class II-peptide complexes. This study represents the first direct single-cell analysis of the impact of intracellular infection on the interaction of macrophages with T cells and serves to emphasize the subtle influence Leishmania has on APC function. 相似文献

Monte Carlo simulations are used to calculate the binding constant of two Ca2+ ions to the protein bovine calbindin D9k. The change in binding constant with respect to mutation of charged amino acids, presence of various electrolytes, protein concentration, solution pH, and competitive binding of monovalent ions is investigated. Each of these factors may have a large influence on the binding constant. The simulations are performed in a dielectric continuum model, the so-called primitive model of electrolyte theory, with a fixed protein structure and a uniform dielectric permittivity. The calculated binding constants are in excellent agreement with experimental data and describe changes in the binding constant over six orders of magnitude. 相似文献

Within the CAZy database, there are 81 carbohydrate‐binding module (CBM) families. A CBM represents a non‐catalytic domain in a modular arrangement of glycoside hydrolases (GHs). The present in silico study has been focused on starch‐binding domains from the family CBM41 that are usually part of pullulanases from the α‐amylase family GH13. Currently there are more than 1,600 sequences classified in the family CBM41, almost exclusively from Bacteria, and so a study was undertaken in an effort to divide the members into relevant groups (subfamilies) and also to contribute to the evolutionary picture of family CBM41. The CBM41 members adopt a β‐sandwich fold (~100 residues) with one carbohydrate‐binding site formed by the side‐chains of three aromatic residues that interact with carbohydrate. The family CBM41 can be divided into two basic subdivisions, distinguished from each other by a characteristic sequence pattern or motif of the three essential aromatics as follows: (i) “W‐W‐~10aa‐W” (the so‐called Streptococcus/Klebsiella‐type); and (ii) “W‐W‐~30aa‐W” (Thermotoga‐type). Based on our bioinformatics analysis it is clear that the first and second positions of the motif can be occupied by aromatic residues (Phe, Tyr, His) other than tryptophan, resulting in the existence of six different carbohydrate‐binding CBM41 groups, that reflect mostly differences in taxonomy, but which should retain the ability to bind an α‐glucan. In addition, three more groups have been proposed that, although lacking the crucial aromatic motif, could possibly employ other residues from remaining parts of their sequence for binding carbohydrate. Empis borealisfemales form swarms, and males carrying a nuptial gift come to swarms to mate. Males either mated with one of the females (accepted swarms) or left swarms without mating (refused swarms). Males mated with the younger (low wing-wear) and relatively larger females in accepted swarms. They seemed to be able to judge the relative size of the females but to ignore their absolute size. Visiting males stayed shorter in accepted swarms as female size variation increased. This probably reflects their greater ease in choosing a mate among females of relatively different sizes.
