Longan species (Dimocarpus longan Lour.) exhibit a high agronomic potential in many subtropical regions worldwide; however, little is known about its responses to abiotic stress conditions. Drought and salinity are the most environmental factors inducing negative effects on plant growth and development. In order to elucidate the responses of longan to drought and salinity, seedlings were grown under conditions of drought and salt stresses. Drought was imposed by suspending water supply leading to progressive soil dehydration, and salinity was induced using two concentrations of NaCl, 100 and 150 mM in water solution, for 64 days. Data showed that salt concentrations increased foliar abscisic acid (ABA) and only 150 mM NaCl reduced indole-3-acetic acid (IAA) and increased proline levels. NaCl treatments also increased Na+ and Cl? content in plant organs proportionally to salt concentration. Drought increased leaf ABA but did not change IAA concentrations, and also increased proline synthesis. In addition, drought and salt stresses reduced the photosynthesis performance; however, only drought decreased leaf growth and relative leaf water content. Overall, data indicate that under severe salt stress, high ABA accumulation was accompanied by a reduction of IAA levels; however, drought strongly increased ABA but did not change IAA concentrations. Moreover, drought and high salinity similarly increased (or maintained) ion levels and proline synthesis. Data also suggest that ABA accumulation may mitigate the impact of salt stress through inducing stomatal closure and delaying water loss, but did not mediate the effects of long-term drought conditions probably because leaves reached a strong dehydration and the role of ABA at this stage was not effective to detain leaf injuries. 相似文献

The OmcB protein is one of the most immunogenic proteins in C. trachomatis and C. pneumoniae infections. RANTES plays a pivotal role in attracting and activating various leukocyte populations that control Mycobacterium tuberculosis infection. The present study investigated the relationship between the RANTES polymorphisms (-28C/G; rs2280788, and -403G/A; rs2107538) and susceptibility to active tuberculosis (TB) in Tunisian populations. A total of 168 patients with pulmonary TB (pTB), 55 with extrapulmonary TB (epTB), and 150 control subjects were studied. Genotype analyses were carried out using polymerase chain reaction-restriction fragment length polymorphism method. We found that the -28 GG genotype was significantly associated with susceptibility to pTB (odds ratio [OR]=11.19; 95% confidence intervals [CI], 5.14-25; P corrected for the number of genotypes [Pc]=10(-8)) and epTB (OR=11.67; 95% CI, 4.74-29.33; Pc=10(-8)). However, the -28 CC genotype was found to be significantly associated with resistance to pTB (OR=0.08; 95% CI, 0.04-0.16; Pc=10(-8)) and epTB development (OR=0.11; 95% CI, 0.05-0.27; Pc=10(-8)). -403A allele was associated with increased risk development of epTB (OR=2.21; 95% CI, 1.18-4.14; p=0.007). G-G and A-C haplotypes and the AG/GC diplotype were associated with increase susceptibility to pTB (OR=7.88, 95% CI, 5.38-11.55; Pc=3.10(-8); OR=2.32, 95% CI, 1.32-4.11; Pc=3.10(-3); OR=13.26, 95% CI, 6.06-29.89; Pc=3.10(-8); respectively) and epTB (OR=6.64, 95% CI, 4-11.05; Pc=3.10(-8); OR=2.6, 95% CI, 1.26-5.35; Pc=12.10(-3); OR=11.26, 95% CI, 4.44-29.28; Pc=3.10(-8); respectively). Collectively, our findings suggested an association of the RANTES -28C/G and -403G/A functional polymorphisms with susceptibility to Mycobacterium tuberculosis infection in Tunisian populations. 相似文献

We aimed in the first part of our work to study the effect of cryopreservation on the human sperm DNA integrity and the activation of caspase 3, the main apoptosis indicator. In the second part, we were interested in testing the effect of quercetin, as an antioxidant, in preventing sperm damage during the freeze–thawing process. Seventeen semen samples were obtained from 17 men recruited for infertility investigations. Liquefied sperm was cryopreserved using spermfreeze®. Nine of the used samples were divided into two aliquots; the first one was cryopreserved with spermfreeze only (control) and the second one was cryopreserved with spermfreeze supplemented with quercetin to a final concentration of 50 μM. Sperm motility and viability were assessed according to WHO criteria. We used TUNEL assay and the Oxy DNA assay to assess sperm DNA integrity. Activated caspase 3 levels were measured in spermatozoa using fluorescein-labeled inhibitor of caspase (FLICA). Cryopreservation led to a significant increase in sperm DNA fragmentation, DNA oxidation and caspase 3 activation (p < 0.01). Supplementation of the cryopreservation medium with quercetrin induced a significant improvement in post thaw sperm parameters, compared to those of control, regarding sperm motility (p = 0.007), viability (p = 0.008) and DNA integrity (p = 0.02); however, it had no effect on caspase 3 activation (p = 0.3). We conclude that oxidative stress plays a major role in inducing sperm cryodamage but implication of apoptosis in this impairment requires further investigations. Quercetin could have protective effect during cryopreservation but further research is needed to confirm this effect. 相似文献

The nosocomial transmission of Candida albicans in neonatal intensive care units (NICUs) is an increasing concern and understanding the route of this transmission is critical for adequate infection control measures. The aim of our study was to assess the likeliness of nosocomial acquisition of C. albicans in the NICU of Farhat Hached hospital in Sousse (Tunisia). We genotyped 82 isolates from 40 neonates and 7 isolates from 5 health care workers (HCWs) with onychomycosis, by using CDC3 microsatellite length polymorphism (MLP) and the high-resolution melting (HRM) analysis. Combined MLP and HRM CD3 analysis led to the delineation of 12 genotypes. Five temporal clustering caused by five genotypes occurred during the study period. Three of these genotypes were isolated in both neonates and HCWs. The first clustering included 28 isolates obtained between January 2003 and May 2004 from 16 neonates and 2 HCWs. The second clustering included three isolates collected in 2004 from three neonates and two HCWs. The third clustering included 11 isolates obtained from 6 neonates and 1 HCW in 2006. The two remaining clustering could not be associated with any HCW's contamination. These results argue for the nosocomial transmission of C. albicans in our NICU. The combined MLP and HRM analysis is a rapid first approach for tracking cross-contamination. 相似文献

There is considerable evidence that host genetic factors are important in determining susceptibility to mycobacterial infections.
More recently, functional genetic mutations affecting IL-10 receptor 1 (IL-10R1) were described. In this study, we investigated
the relationship of IL-10R1 S138G loss-of-function polymorphism (A536G: rs3135932) with susceptibility to active tuberculosis
(TB) in Tunisian patients. A total of 168 patients with pulmonary TB, 55 with extrapulmonary TB, and 150 control subjects
were studied. Genomic DNA samples were extracted from leukocytes and used to investigate S138G polymorphism in IL-10R1 gene
by multiplex allele-specific polymerase chain reaction. Associations between G allele [odds ratio OR = 5.01; 95% confidence
intervals CI = 2.58–9.77; P = 10−7], GG genotypes [OR=9.06; 95% CI (1.58–67.33); correcting P-values using the Bonferroni method for multiple tests Pc=0.015] and AG genotype [OR=3.75; 95% CI (1.62–8.7); Pc=0.0012] with
the risk development of active extrapulmonary TB were found. In contrast, the AA genotype was found to be associated with
resistance to extrapulmonary TB [OR=0.19; 95% CI (0.09–0.42); Pc=6.10−6]. No association was found between S138G SNP and pulmonary TB. In conclusion, our study suggested the possible role of IL-10R1
S138G loss-of-function polymorphism in extrapulmonary TB susceptibility-resistance in Tunisia. 相似文献

Current evidence in citrus indicates that gibberellins (GAs) are main determinants of early fruit set while subsequent growth
of developing fruits is mostly dependent upon carbohydrate availability. In this work, branch girdling performed at anthesis
in Satsuma mandarin (Citrus unshiu (Mak.) Marc.) cv. Okitsu transitorily reduced early abscission rates (12–32 days after anthesis, DAA) delaying initially
the process of natural fruitlet drop. The effects of girdling on growth, gibberellin (GA) and carbohydrate concentrations
in developing ovaries and fruitlets were assessed during this initial growth stage (0–69 DAA). In girdled branches, abscission
rate reduction was preceded by elevated concentrations of carbohydrate and GA in developing ovaries and fruitlets. Girdling
at anthesis stimulated higher hexose (21 DAA) and starch (6–20 DAA) concentrations and also higher GA1 (6 DAA), GA19 (13–20 DAA) and GA20 (6–20 DAA). The results established a relationship between the reduction of early abscission rates and higher concentrations
of carbohydrates and GAs induced by girdling in developing fruitlets. These findings revealed that girdling certainly increased
GA concentration and strongly suggested that its effect on early fruitlet abscission delay is likely mediated by both GA and

AimsIn this study, the effects of four single nucleotide polymorphisms (SNPs), ? 3860G > A, ? 2467delT, ? 739T > G and ? 163C > A, of CYP1A2 gene on lung cancer were evaluated in Tunisian population.Main methodsFour polymorphisms of CYP1A2 gene were analysed in 109 healthy smokers and in 101 lung cancer cases, including 63 with squamous cell carcinoma (SCC) and 41 with adenocarcinoma (AD). The genotyping for the SNPs ? 3860 G > A, ? 2467delT, ? 739T > G and ? 163C > A was performed by polymerase chain reaction (PCR)-restriction fragment length polymorphism analysis.Key findingsThe results showed that smokers with CYP1A2 gene polymorphisms were associated with an increased risk for the development of lung AD. There was however no significant increased risk of developing lung SCC in smokers having CYP1A2 gene polymorphisms. An increased risk of developing AD was observed in smokers who are carriers of at least one copy of ? 3680A or ? 739G giving a significant odds ratio (OR) of 6.02 (CI = 2.91–12.9) and 3.01 (CI = 1.54–5.98), respectively.SignificanceThese genotyping data are consistent with the hypothesis that tobacco-specific-N-nitrosamines (TSN) such as 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK) are major contributors to the development of lung AD and that CYP1A2 gene product plays an important role in the metabolic activation of NNK. The effects of exogenous foliar glycine betaine (GB) and abscisic acid (ABA) on papaya responses to water stress were investigated
under distinct water regimes. Papaya seedlings (Carica papaya L. cultivar “BH-65”) were pretreated with GB or ABA and subsequently subjected to consecutive periods of drought, rehydration,
and a second period of drought conditions. Results indicated that water stress induced ABA, jasmonic acid (JA), and proline
accumulation but did not modify malondialdehyde (MDA) concentration. In addition, water deprivation reduced photosynthetic
rate, stomatal conductance, relative water content (RWC), leaf fresh weight, and increased leaf abscission. GB applied prior
to drought imposition decreased the impact of water stress on ABA, JA, proline accumulation, leaf water status, growth, and
photosynthetic performance. However, ABA-pretreated plants did not show alteration of most of these parameters under water
stress conditions when compared with non-pretreated plants except a clear induction of JA accumulation. Taken together, the
data suggest that GB may modulate ABA, JA, and proline accumulation through the control of stomatal movement and the high
availability of compatible solutes, leading to improvement of leaf water status, growth, and photosynthetic machinery function.
In contrast, exogenous ABA did not stimulate papaya physiological responses under drought, but interestingly ABA in combination
with drought could induce progressive JA synthesis, unlike drought alone, which induces a transitory JA increase and may trigger
endogenous ABA accumulation. The data also suggest that irrespective of the pretreatments, papaya did not suffer oxidative
