Abstract The production of fimbrial antigen F165 by Escherichia coli strains was found to be dependent on the composition of the culture medium and was repressed in the presence of alanine or high levels of glucose, in anaerobic conditions or at growth temperatures of lower than 37°C. Optimal F165 production was found on a minimal medium containing 1% (w/v) casamino acids (MD-1). F165 antigen was isolated from bacteria by mechanical shearing, precipitated with ammonium sulfate, and purified by deoxycholate treatment and gel filtration on Superose 12. The purified fimbriae retained their native morphology as observed by electron microscopy and consisted of two separate protein subunits with apparent molecular weights of 17 500 and 19 000 on sodium dodecyl sulfate-polyacrylamide gels. 相似文献

Chloroplasts are bounded by a pair of outer membranes, the envelope, that is the only permanent membrane structure of the
different types of plastids. Chloroplasts have had a long and complex evolutionary past and integration of the envelope membranes
in cellular functions is the result of this evolution. Plastid envelope membranes contain a wide diversity of lipids and terpenoid
compounds serving numerous biochemical functions and the flexibility of their biosynthetic pathways allow plants to adapt
to fluctuating environmental conditions (for instance phosphate deprivation). A large body of knowledge has been generated
by proteomic studies targeted to envelope membranes, thus revealing an unexpected complexity of this membrane system. For
instance, new transport systems for metabolites and ions have been identified in envelope membranes and new routes for the
import of chloroplast-specific proteins have been identified. The picture emerging from our present understanding of plastid
envelope membranes is that of a key player in plastid biogenesis and the co-ordinated gene expression of plastid-specific
protein (owing to chlorophyll precursors), of a major hub for integration of metabolic and ionic networks in cell metabolism,
of a flexible system that can divide, produce dynamic extensions and interact with other cell constituents. Envelope membranes
are indeed one of the most complex and dynamic system within a plant cell. In this review, we present an overview of envelope
constituents together with recent insights into the major functions fulfilled by envelope membranes and their dynamics within
plant cells.

Hairy root cultures of Atropa belladonna L. were established by infection either with Agrobacterium rhizogenes ATCC 15834 or MAFF 03-01724, and transgenic plants were obtained from both hairy root cultures. Doubly transformed roots were induced by re-infection of the leaf segments of transgenic Atropa belladonna plants (A. rhizogenes 15834) with MAFF 03-01724. Shoots and viviparous leaves were regenerated from the doubly transformed roots. The genetic transformation was determined by the opine assay (agropine, mannopine and/or mikimopine) and polymerase chain reaction. Physiological changes and tropane alkaloid biosynthesis in the hairy roots (singly and doubly transformed) were investigated. The alkaloid content in the doubly transformed root strain was intermediate as compared to the root strains which were singly transformed. On the other hand endogenous IAA levels in doubly transformed roots were significantly decreased compared to both singly transformed roots.Abbreviations BA

Binding of arachidonic acid to myeloid-related proteins (S100A8/A9) enhances phagocytic NADPH oxidase activation 总被引:1,自引:0,他引:1
Activation of the O(2)(-) generating NADPH oxidase of phagocytes results from the assembly of the membrane-bound flavocytochrome b(558) with cytosolic proteins, p67(phox), p47(phox), and Rac. However, it has been recently reported that the arachidonic acid- and calcium-binding heterodimer S100A8/A9, abundant in neutrophil cytosol, influences the activation process. In a semi-recombinant system comprising neutrophil membranes, recombinant proteins, p67(phox), p47(phox), GTPgamma S-loaded Rac2, and arachidonic acid (AA), both the rate and the extent of the oxidase activation were increased by S100A8/A9, provided it was preloaded with AA. Binding of [(14)C]AA to S100A8/A9 was potentiated by recombinant cytosolic phox proteins and GTPgammaS, suggesting the formation of a complex, comprising oxidase activating proteins and S100A8/A9, with a greater affinity for AA. The rate constant of oxidase activation was not increased by AA-loaded S100A8/A9, whereas the maximal oxidase activity elicited was twice as high. AA-loaded S100A8/A9 increases oxidase activation probably by decreasing the deactivation rate. 相似文献

Most studies on the importance of the neighbourhood on a plant's risk ofherbivory have focused on palatable plants and how they are protected byunpalatable neighbours. This study examined the grazing intensity of arelatively unpalatable shrub, Buxus sempervirens, indifferent neighbours. Exactly 2683 plants of Buxussempervirens (including 172 controls) were sampled in 12 enclosedpastures belonging to 4 sheep farms. The enclosures were grazed at 3 differentseasons (spring, summer and autumn). Plants were divided in 4 age/hight classes(first year, < 4 cm, 4–10 cm, 10–40cm) and into 8 neighbourhoods. The first of these wascharacterisedby the absence of any plants within a radius of 5 cm around theBuxus individual and the 7 others by the identity of thedominant species in contact with the Buxus plant. Theintensity of grazing on the neighbouring plants were also recorded. At the endof one year's monitoring, 26.2% of Buxus sempervirensplants had been grazed. The proportion of plants grazed was significantlyhigherin spring than in the other two seasons. It decreased with increasing plantage.It was higher in neighbourhoods that were intensively grazed than in those withlight grazing. The proportion grazed in the absence of a neighbour plant wasintermediate between the previous two. The probability of a plant of aninvadingspecies being grazed is influenced by factors other than its life-historytraits. Some neighbourhoods consisting of unpalatable plants facilitate theestablishment of Buxus sempervirens by protecting theyoungplants from grazing, whereas other highly palatable neighbourhoods are readilygrazed by sheep, thus indirectly increasing the proportion of Buxussempervirens that are grazed. In Cupriavidus metallidurans CH34, the proteins CnrX, CnrY, and CnrH regulate the expression of the cnrCBA operon that codes for a cation-efflux pump involved in cobalt and nickel resistance. The periplasmic part of CnrX can be defined as the metal sensor in the signal transduction complex composed of the membrane-bound anti-sigma factor CnrY and the extra-cytoplasmic function sigma factor CnrH. A soluble form of CnrX was overproduced and purified. This protein behaves as a dimer in solution as judged from gel filtration, sedimentation velocity experiments, and NMR. Native crystals diffracting to 2.3 A using synchrotron radiation were obtained using the hanging-drop vapor-diffusion method. They belong to the primitive monoclinic space group P2(1), with unit cell parameters a = 31.87, b = 74.80, c = 93.67 A, beta = 90.107 degrees. NMR data and secondary structure prediction suggest that this protein is essentially formed by helices. 相似文献

Abstract. The native flora of tropical oceanic islands is known to be particularly susceptible both to displacement and extinction, following the invasion of alien organisms. Miconia calvescens DC. (Melastomataceae), first introduced to Tahiti (French Polynesia, South Pacific Ocean) in 1937 as an ornamental plant, now covers over two-thirds of the Island. As it forms dense monotypic stands which have progressively overwhelmed the native forests, this plant pest is a direct threat to the rich Tahitian indigenous flora. Between 40 and 50 species of the 107 species endemic to Tahiti are thought to be on the verge of extinction. M. calvescens was finally declared a'noxious species in French Polynesia'in 1990. Without efficient control efforts and effective endangered plant conservation and protection legislation, M. calvescens could cause Tahiti and all the high islands of French Polynesia to become ecological deserts. 相似文献

We examined the impact of ploughing on soil seed banks of plant communities living in temporary marshes located in agricultural
fields. The quantity, quality and vertical distribution of seeds were quantified under ploughed or unploughed treatment at
community level. We also focussed on a typical semi-aquatic species, Damasonium alisma, to investigate the impact of ploughing at population level. We used two complementary techniques to study seed banks: hand
sorting and seedling emergence. We found that species richness of seeds, seed abundance and germination ability were strongly
affected by ploughing at community level. Concerning D. alisma, most of the seeds (56%) were stored in the two deepest soil layers among the four considered in ploughed pools. Moreover,
the germination rate was higher for buried seeds (84%) than for seeds collected at the surface (33.6%). These patterns were
almost inverted in unploughed pools. Our results agree with the temporal storage effect generally suggested to describe the
seed bank property of plant communities. But in addition, we showed that ploughing induces a spatial storage effect in accumulating
species and individuals in the seed banks that favourably influence community dynamics. We conclude that, in contrast to what
is usually thought, ploughing disturbance can be of benefit for such ephemeral wetland vegetation. 相似文献

Résumé Du cordo-mésoderme dorsal greffé sur la région ventrale d'un massif endodermique de même âge provoque sa différenciation locale (st. 17/neurula). Des néo-formations nombreuses et variées sont obtenues en faisant varier les caractéristiques du greffon (cordo-mésoderme) et du porte-greffe (endoderme). L'existence, la nature et le volume de ces néo-formations dépendent: de l'étroitesse et de la durée du contact greffon/porte-greffe, de l'âge du cordo-mésoderme et de l'âge de l'endoderme, des niveaux de contact, plus ou moins antérieurs, du greffon et du porte-greffe.Les résultats des greffes (st. 17) sont interprétés grâce à la construction de modèles théoriques. Ces modèles sont bâtis sur les quatre paramètres suivants: 1) l'existence de 2 facteurs morphogénétiques, l'un de nature endodermique et l'autre de nature cordo-mésodermique. Ces 2 facteurs sont responsables de l'apparition de néo-formations; 2) la localisation de ces 2 facteurs actifs dans les deux tiers (antérieur et moyen) de l'endoderme et du cordo-mésoderme; 3) la décroissance de l'activité des 2 facteurs selon l'axe antéro-postérieur de l'embryon; 4) le rôle différent de chacun de ces 2 facteurs. Le facteur endodermique déterminerait la nature et la taille des néo-formations; le facteur cordo-mésodermique jouerait un rôle stimulant.Une discussion est engagée sur la méthode, la démarche et l'intérêt de ce type d'interprétation des résultats.
Effect of dorsal chordo-mesoderm on regionalisation and differentiation of the endodermal mass inRana dalmatina bon (Amphibia Anura)Elaboration of a theoretical model
Summary Chordo-mesoderm grafted onto the ventral area of yolk endoderm of the same age causes its local differentiation (Rana dalmatina: st. 17/neurula). Numerous and various neo-formations are achieved by variation of the grafted tissue (chordo-mesoderm) and host (endoderm) characteristics. The existence, the constitution and the volume of the neo-formations are dependant on: the tightness and the duration of the graft/host contact, the age of the chordo-mesoderm and the age of the endoderm, and the antero-posterior level of contact with grafted tissue and host.The results of the grafts (st. 17) are explained by the elaboration of a theoretical model. These models are elaborated according to four parameters: (1) the existence of two morphogenetic factors, one endodermal and the other mesodermal. These two factors are responsible for the constitution of neo-formations; (2) the localization of these two factors, active in the anterior and median thirds of the endoderm and the chorda-mesoderm; (3) decreasing activity of these two factors along to the antero-posterior axis of the embryo; (4) the different notes of each of these two factors. The endodermal factor might determine the constitution and the size of the neo-formations; the chordo-mesodermal factor might play a stimulatory role.The method, the procedure and the interpretation of this kind of results are discussed.相似文献

Stochastic simulation of daily air temperature and precipitation from monthly normals in North America north of Mexico 总被引:2,自引:0,他引:2
A simple, stochastic daily temperature and precipitation generator (TEMPGEN) was developed to generate inputs for the study
of the effects of climate change on models driven by daily weather information when climate data are available as monthly
summaries. The model uses as input only 11 sets of monthly normal statistics from individual weather stations. It needs no
calibration, and was parameterized and validated for use in Canada and the continental United States. Monthly normals needed
are: mean and standard deviation of daily minimum and maximum temperature, first and second order autoregressive terms for
daily deviations of minimum and maximum temperatures from their daily means, correlation of deviations of daily minimum and
maximum temperatures, total precipitation, and the interannual variance of total precipitation. The statistical properties
and distributions of daily temperature and precipitation data produced by this generator compared quite favorably with observations
from 708 stations throughout North America (north of Mexico). The algorithm generates realistic seasonal patterns, variability
and extremes of temperature, precipitation, frost-free periods and hot spells. However, it predicts less accurately the daily
probability of precipitation, extreme precipitation events and the duration of extreme droughts. 相似文献
