Stimulation of the desaturation of 1-alkyl-2-acyl-sn-glycero-3-phosphoethanolamine (GPE), which forms ethanolamine plasmalogens, by a component of the 105,000g supernatant has been previously reported. We have isolated the stimulatory protein and identified it as catalase. Purified rat liver catalase or commercial bovine liver catalase is as effective in stimulating microsomal 1-alkyl-2-acyl-GPE desaturation as the soluble proteins. The stimulatory effect of these proteins is eliminated by catalase inhibitors. It appears that catalase stimulates the desaturation of 1-alkyl-2-acyl-GPE by preventing inactivation of the enzyme system by H2O2 or a decomposition product of H2O2. The cytochrome b5 content and NADH oxidation are depressed in Fischer R-3259 sarcoma microsomes by H2O2; this effect is eliminated by catalase. However, since measurable inhibition of 1-alkyl-2-acyl-GPE desaturase by H2O2 still occurred in the presence of catalase, the inhibition by H2O2 cannot be explained solely on the basis of cytochrome b5 inactivation. The desaturation of stearoyl-coenzyme A, a reaction analogous in many respects to 1-alkyl-2-acyl-GPE desaturation, was also found to be stimulated by catalase.  相似文献

Aflatoxins are produced by Aspergillus flavus and A. parasiticus in oil-rich seed and grain crops and are a serious problem in agriculture, with aflatoxin B1 being the most carcinogenic natural compound known. Sexual reproduction in these species occurs between individuals belonging to different vegetative compatibility groups (VCGs). We examined natural genetic variation in 758 isolates of A. flavus, A. parasiticus and A. minisclerotigenes sampled from single peanut fields in the United States (Georgia), Africa (Benin), Argentina (Córdoba), Australia (Queensland) and India (Karnataka). Analysis of DNA sequence variation across multiple intergenic regions in the aflatoxin gene clusters of A. flavus, A. parasiticus and A. minisclerotigenes revealed significant linkage disequilibrium (LD) organized into distinct blocks that are conserved across different localities, suggesting that genetic recombination is nonrandom and a global occurrence. To assess the contributions of asexual and sexual reproduction to fixation and maintenance of toxin chemotype diversity in populations from each locality/species, we tested the null hypothesis of an equal number of MAT1-1 and MAT1-2 mating-type individuals, which is indicative of a sexually recombining population. All samples were clone-corrected using multi-locus sequence typing which associates closely with VCG. For both A. flavus and A. parasiticus, when the proportions of MAT1-1 and MAT1-2 were significantly different, there was more extensive LD in the aflatoxin cluster and populations were fixed for specific toxin chemotype classes, either the non-aflatoxigenic class in A. flavus or the B1-dominant and G1-dominant classes in A. parasiticus. A mating type ratio close to 1∶1 in A. flavus, A. parasiticus and A. minisclerotigenes was associated with higher recombination rates in the aflatoxin cluster and less pronounced chemotype differences in populations. This work shows that the reproductive nature of the population (more sexual versus more asexual) is predictive of aflatoxin chemotype diversity in these agriculturally important fungi.  相似文献

Knowledge of roost selection by northern yellow bats (Lasiurus intermedius) is limited to a small number of known roost locations. Yet knowledge of basic life history is fundamental to understanding past response to anthropogenic change and to predict how species will respond to future environmental change. Therefore, we examined male northern yellow bat roost selection on 2 Georgia, USA, barrier islands with different disturbance histories. Sapelo Island has a history of extensive disturbance and is dominated by pine (Pinus spp.) forests; Little Saint Simons Island has a limited disturbance history with maritime oak (Quercus spp.) forest as the dominant cover type. From March–July 2012 and 2013, we radio-tracked 35 adult male northern yellow bats to diurnal roosts and modeled roost characteristics at the plot and landscape scales. We located 387 roosts, of which 95% were in Spanish moss (Tillandsia usneoides) hanging in hardwood trees. On both islands, bats selected roost trees with larger diameters than surrounding trees and selected roost locations with greater open flight space (i.e., low midstory clutter) underneath. Roosts were located farther from open areas on Sapelo and closer to fresh water on Little Saint Simons compared to random locations. Lower availability of hardwood forest on Sapelo may have resulted in small-scale roost site selection (i.e., plot level) despite potential increased costs of commuting to water and open areas for foraging. In contrast, greater availability of hardwood forest on Little Saint Simons likely allowed selection of roosts closer to fresh water, which provides foraging and drinking opportunities. Our results indicate that mature hardwood trees in areas with low midstory clutter are important in male northern yellow bat roost selection, but landscape-level features have varying influences on roost selection, likely as a result of differences in disturbance history. Therefore, management will differ depending on the landscape context. Further research is needed to examine roost selection by females, which may have different habitat requirements. © 2020 The Wildlife Society.  相似文献

A novel series of 3-O-carbamoyl erythromycin A derived analogs, labeled carbamolides, with activity versus resistant bacterial isolates of staphylococci (including macrolide and oxazolidinone resistant strains) and streptococci are reported. An (R)-2-aryl substituent on a pyrrolidine carbamate appeared to be critical for achieving potency against resistant strains. Crystal structures showed a distinct aromatic interaction between the (R)-2-aryl (3-pyridyl for 4d) substituent on the pyrrolidine and G2484 (G2505, Escherichia coli) of the Deinococcus radiodurans 50S ribosome (3.2 Å resolution).  相似文献

Ahnak is a gigantic (700 kD) phosphoprotein with a unique structure whose expression and cellular localization are dynamically regulated during cell cycle progression. Here, we report that Ahnak is localized to sites of major morphogenesis during mouse placentation and neurulation. Ahnak was found in: (i) derivatives of trophectoderm, including chorionic ectoderm prior to and during union with the ectoplacental cone, presumptive syncytiotrophoblast cells in the chorionic labyrinth, and giant cells at the trophoblast-uterine interface; (ii) the allantois prior to, during, and after union with the chorion; and (iii) the tips of the neural plate during formation of the neural tube. On the basis of these observations, we suggest that Ahnak may play heretofore unrecognized roles in tissue union during normal mouse development.  相似文献

Previous studies have shown that insulin receptor substrate (IRS)1 and IRS2 mediate proliferative and antiapoptotic signaling through the IL-4R in 32D cells; however their role in regulating normal B cell responses is not clear. To investigate the role of IRS2 in normal B cell function, we developed IRS2 transgenic (Tg) mice on the C57BL/6 background. Western blot analysis revealed a 2-fold elevation in IRS2 protein levels in Tg(+) mice compared with littermate controls and a 3-fold increase in basal tyrosine phosphorylated IRS2 in the absence of IL-4 stimulation. IL-4-induced tyrosine phosphorylation of IRS2 was elevated in Tg(+) B cells, whereas IL-4-induced phosphorylation of STAT6 was similar between Tg(+) and Tg(-) B cells. Tg expression of IRS2 had little effect on IL-4-mediated proliferation and no effect on protection from apoptosis. However, production of IgE and IgG1 by Tg(+) B cells using standard in vitro conditions was diminished 50-60%. Because Ig production in vitro is known to be highly cell concentration-dependent, we performed experiments at different cell concentrations. Interestingly, at very low B cell concentrations (1000-5000 B cells/well), IgE and IgG1 production by Tg(+) B cells was greater than that of controls, whereas at higher cell concentrations (10,000-20,000 cells/well) Ig production by Tg(+) B cells was less than controls. Furthermore, in vivo immunization with OVA-alum or goat anti-IgD resulted in elevated serum IgE levels in the Tg(+) mice. These results indicate that overexpression of IRS2 alters the B cell intrinsic density-dependence of IgE and IgG1 production in vitro and enhances IgE responses in vivo.  相似文献
