The BULT melanoma originated at Brown University as a spontaneous, small black nodule on the tail of an adult female mouse of the LT/Ch strain. Histological examination of a portion of the tumor indicated that it was intradermal and consisted predominantly of heavily melanized, ovoid to fusiform cells with melanin-laden macrophages scattered among them. The BULT melanoma has been maintained in LT/Ch mice for approximately 5 years by periodic transplantation, at first subcutaneously on the flanks and, more recently, intramuscularly in the hind legs. The shift in transplantation site was made following a marked decline in the growth of subcutaneous grafts. The transplants have retained the uniform deep-black melanization and general histology of the primary melanoma. Numerous melanosomes at all stages of development are found within the melanoma cells. DOPA-positive cytoplasmic vesicles are abundant. Occasional autophagic vacuoles containing clusters of melanosomes are also present. A few metastases from the transplanted melanoma have been observed in lymph nodes and on one occasion in the lungs. When grown in vitro, BULT melanoma cells do not require special growth promoting agents (e.g., TPA; cAMP) in order to proliferate. The BULT melanoma differs in one or more respects from each of the other three transplantable spontaneous mouse melanomas widely used in cancer research. In addition, it arose in a strain of mice characterized by the spontaneous death of melanocytes while the latter are engaged in synthesizing eumelanin within hair follicles. The Response of Bean Plants to UV-B Radiation Under Different Irradiances of Background Visible Light 总被引:14,自引:1,他引:13
Plants of Phaseolus vulgaris L. (cv. Stella) were grown in controlledconditions under three different irradiances of visible lightwith or without UV-B (280320nm) radiation. The biologicallyeffective UV-B radiation (UV-BBE) was 6.17 kJ m2 d1,and simulated a c. 5% decrease in stratospheric ozone at 55.7?N,13.4?E. The photon flux densities of the photosyntheticallyactive radiation (PAR, 400700 nm) were either 700 µmolm21 (HL), 500, µmol m2 s1(ML) or 230 µmol m2 s1 PAR (LL). Under highlight (HL) conditions plus UV-B radiation, bean plants appearedmost resistant to the enhanced levels of UV-B radiation, andresponded only by increasing leaf thickness by c. 18%. A smallincrease in UV screening pigments was also observed. Both thelower irradiances (ML and LL) increased the sensitivity of theplants to UV-B radiation. Changes in leaf structure were alsoobserved. Photosystem II was inhibited under ML and LL togetherwith UV-B radiation, as determined by Chi fluorescence inductionand calculation of the fluorescence half-rise times. Leaf reflectivitymeasurements showed that the amount of PAR able to penetrateleaves of UV-B treated plants was reduced, and that a possiblecorrelation may exist between the reduced PAR levels, loss ofChi and lowered photosynthetic activity, especially for LL +UV-Bgrown plants, where surface reflection from leaves was highest.Changes in leaf chlorophyll content were mostly confined toplants grown under LL + UV-B, where a decrease of c. 20% wasfound. With regard to protective pigments (the carotenoids andUV screening pigments) plants subjected to different visiblelight conditions responded differently. Among the growth parametersmeasured, there was a substantial decrease in leaf area, particularlyunder LL + UV-B (c. 47% relative to controls), where leaf dryweight was also reduced by c. 25%. Flow cytometric determination of cell cycle activation duringimbibition and visible germination in five families of jackpine (Pinus banksiana Lamb.) embryos and megagametophytes revealedthat in seeds that had undergone no imbibition the majorityof cells were in the 2C state. As the imbibition period increased,less of the nuclei were blocked in the G0/G1 state and morebecome active in the cell cycle. The augmentation in the nucleiactive in the 2C4C cycle as well as those with DNA levelshigher than the 4C state occured gradually and preceeded radicleemergence. In megagametophyte tissue examined at various stagesof imbibition, cell cycle activity became apparent rapidly followingimbibition. In nuclei of green and white embryos examined separatelythe 2frequency distributions were significantly different forall three families after 144h. As imbibition period increased,fewer nuclei from the green embryos were blocked in the 2C state,and more became active in the 2C4C cell cycle. This wasnot the case for white embryos where no significant linear relationwas noted. Cell cycle activity in the hypocotyl+cotyledons regionand the emerging radicle were examined separately. Functionalrelations found in the hypocotyl+cotyledons region were notevident in the radicle. In a sandy loam under glass, potato cyst-nematode, Globodera pallida, was controlled in soil to 20 cm deep, and tomato yields were greatly increased by sheet steaming for 4 h, by 977 kg methyl bromide ha-1 or by 448 kg Telone (1,3-dichloropropene mixture or Di-Trapex CP (20% methyl isothiocyanate: 15% chloropicrin:65% dichloropropene-dichloropropane mixture) ha-1 followed by 441 kg dazomet or 11.2 kg oxamyl ha-1. The aphid-pathogenic fungus Erynia neoaphidis, as dried fungus-infected aphids, was applied to caged plots of winter wheat infested with cereal aphids at two sites, one in Hertfordshire and the other in Hampshire, in 1983. In each trial, the fungus became established in the aphid populations in the treated plots even though conditions were drier than average and therefore sub-optimal for fungus spread. Treatment applied in the third week of June increased the proportion of infected aphids more than that applied two weeks later at one site, and the early application was the only treatment to have an obvious effect at the other. In spite of the observed effect of treatments on the proportion of infected aphids, the fungus failed to reduce the numbers of aphids relative to those in untreated plots, chiefly because in these plots many aphids were killed by fungi of the same species as that introduced and other related species from natural sources. Artificial introduction of E. neoaphidis acts too slowly and unpredictably to be likely to form a practical alternative to conventional insecticides for cereal aphid control. 相似文献

The symbiotic effectiveness of four cultivars of red clover were compared and breeding programmes undertaken to increase nitrogen fixation and yield of agar- and pot-grown plants. Programme I used the moderately effective Rhizobium trifolii strain 0403 and Programme II the highly effective strain 5. Aggregate scores of plant size (leaf area) were chosen as the criterion of selection. All cultivars, inoculated with strain 0403, differed in time of initial nodulation and in dry matter yield and three differed in leaf area. None differed in nodule number or N-content. Dry matter and leaf area were highly correlated. Diallel crosses among highly effective selections in Programme 1 gave progeny yields that exceeded those of crosses between modal selections by averages of 6% in the first generation, 5% in the second generation and 23% in the third generation. Yields of the high cross category exceeded those of the original cultivar by an average of 9% in the second generation and by 25–101% in the third generation. Effects on leaf area were similar. Highly effective progeny tended to nodulate earlier and have more nodules than the remainder but differences were very small. Crosses between cultivars were generally more effective than those within cultivars, indicating heterotic effects. Similar results for yield were obtained in Programme II. The average increase in yield of crosses of third-generation material between highly effective selections compared with cv. S123 was 63%. The modal crosses were intermediate. Summary Employment of Markowitz's Modern Portfolio Theory, economic models designed to predict the effect of variance and covariance on optimal investment allocation, may explain a wide variety of anomalies in reproductive biology. Natural selection appears to favor genetic diversity among offspring to a greater extent than is predicted by current theory. Consideration of the possible increase in fitness by reducing the covariance among offspring may help explain a variety of phenomena from multiple mating to the evolution of recombination (i.e., overcoming “the cost of meiosis”). Modern Portfolio Theory also make novel predictions as to when hermaphrodites should prefer the male vs. female role, i.e., engage in egg- vs. sperm-trading. It predicts that the sexual role with the lower variance in reproductive success will be preferred in hermaphrodites. This contradicts Bateman's principle that the male role is usually preferred due to energetic considerations but is consistent with Gillespie's principle. The available data suggest that mating hermaphrodites are risk-averse; gamete-trading whether of eggs or sperm is a strategy to reduce risk. In addition to overall variance, the skew of the distribution can be used to predict the mating systems of hermaphrodites and thus clarify the factors that are responsible for observed patterns of sex allocation and sexual conflict. The reduction of covariance among offspring may also help resolve “Williams' paradox”; that the observed distribution of dieoecy vs. simultaneous hermaphroditism in the Animal Kingdom cannot be explained by the prevailing models of the evolution of hermaphroditism. 相似文献
