Background  

The need for discovery of alternative, renewable, environmentally friendly energy sources and the development of cost-efficient, "clean" methods for their conversion into higher fuels becomes imperative. Ethanol, whose significance as fuel has dramatically increased in the last decade, can be produced from hexoses and pentoses through microbial fermentation. Importantly, plant biomass, if appropriately and effectively decomposed, is a potential inexpensive and highly renewable source of the hexose and pentose mixture. Recently, the engineered (to also catabolize pentoses) anaerobic bacterium Zymomonas mobilis has been widely discussed among the most promising microorganisms for the microbial production of ethanol fuel. However, Z. mobilis genome having been fully sequenced in 2005, there is still a small number of published studies of its in vivo physiology and limited use of the metabolic engineering experimental and computational toolboxes to understand its metabolic pathway interconnectivity and regulation towards the optimization of its hexose and pentose fermentation into ethanol.  相似文献

Caloric restriction (CR) has been associated with health benefits and these effects have been attributed, in part, to modulation of oxidative status by CR; however, data are still controversial. Here, we investigate the effects of seventeen weeks of chronic CR on parameters of oxidative damage/modification of proteins and on antioxidant enzyme activities in cardiac and kidney tissues. Our results demonstrate that CR induced an increase in protein carbonylation in the heart without changing the content of sulfhydryl groups or the activities of superoxide dismutase and catalase (CAT). Moreover, CR caused an increase in CAT activity in kidney, without changing other parameters. Protein carbonylation has been associated with oxidative damage and functional impairment; however, we cannot exclude the possibility that, under our conditions, this alteration indicates a different functional meaning in the heart tissue. Streptococcus agalactiae isolates are more common among pregnant women, neonates and nonpregnant adults with underlying diseases compared to other demographic groups. In this study, we evaluate the genetic and phenotypic diversity in S. agalactiae strains from Rio de Janeiro (RJ) that were isolated from asymptomatic carriers. We analysed these S. agalactiae strains using pulsed-field gel electrophoresis (PFGE), serotyping and antimicrobial susceptibility testing, as well as by determining the macrolide resistance phenotype, and detecting the presence of the ermA/B, mefA/E and lnuB genes. The serotypes Ia, II, III and V were the most prevalent serotypes observed. The 60 strains analysed were susceptible to penicillin, vancomycin and levofloxacin. Resistance to clindamycin, chloramphenicol, erythromycin, rifampin and tetracycline was observed. Among the erythromycin and/or clindamycin resistant strains, the ermA, ermB and mefA/E genes were detected and the constitutive macrolides, lincosamides and streptogramin B-type resistance was the most prevalent phenotype observed. The lnuB gene was not detected in any of the strains studied. We found 56 PFGE electrophoretic profiles and only 22 of them were allocated in polymorphism patterns. This work presents data on the genetic diversity and prevalent capsular serotypes among RJ isolates. Approximately 85% of these strains came from pregnant women; therefore, these data may be helpful in developing future prophylaxis and treatment strategies for neonatal syndromes in RJ.  相似文献

Though, it is quite well-known how retinoic acid (RA) is able to induce neuritogenesis in different in vitro models, the putative role exerted by reactive oxygen species (ROS) during this process still need to be further studied. For such purpose, we used a neuronal-like cell line (SH-SY5Y cells) in order to investigate whether the antioxidant Trolox (a hydrophilic analog of alpha-tocopherol) could have any effect on the number of RA-induced neurites, and how significant changes in cellular redox homeostasis may affect the cellular endogenous expression of tyrosine hydroxylase (TH). Our results show a significant enhancement of RA (10 μM)-induced neuritogenesis and TH endogenous expression, when cells were co-treated with Trolox (100 μM) for 7 days. Moreover, this effect was associated with an improvement in cellular viability. The mechanism seems to mainly involve PI3 K/Akt rather than MEK signaling pathway. Therefore, our data demonstrate that concomitant decreases in basal reactive oxygen species (ROS) production could exert a positive effect on the neuritogenic process of RA-treated SH-SY5Y cells.  相似文献

Rheumatoid arthritis (RA) is a destructive arthropathy with systemic manifestations, characterized by chronic synovial inflammation. Under the influence of the pro-inflammatory milieu synovial fibroblasts (SFs), the main effector cells in disease pathogenesis become activated and hyperplastic while releasing a number of signals that include pro-inflammatory factors and tissue remodeling enzymes. Activated RA SFs in mouse or human arthritic joints express significant quantities of autotaxin (ATX), a lysophospholipase D responsible for the majority of lysophosphatidic acid (LPA) production in the serum and inflamed sites. Conditional genetic ablation of ATX from SFs resulted in attenuation of disease symptoms in animal models, an effect attributed to diminished LPA signaling in the synovium, shown to activate SF effector functions. Here we show that administration of 1-bromo-3(S)-hydroxy-4-(palmitoyloxy)butyl-phosphonate (BrP-LPA), a metabolically stabilized analog of LPA and a dual function inhibitor of ATX and pan-antagonist of LPA receptors, attenuates collagen induced arthritis (CIA) development, thus validating the ATX/LPA axis as a novel therapeutic target in RA.  相似文献

Vole–vegetation interactions in a predation‐free taiga environment of northern Fennoscandia were studied by transferring vegetation from natural Microtus habitats into a greenhouse, where three habitat islands of about 30 m2 were created. The ‘islands’ were subjected to simulated summer conditions and a paired female field vole, Microtusagrestis, was introduced to each ‘island’. The development of the female and her young was followed by recurrent live trapping. The development of the vegetation was followed by recurrent marking and censusing of plant shoots at intervals of five days. In the next growing season, two ‘islands’ were subjected to a new grazing treatment to study the impacts of repeated grazing on the vegetation and on the growth and reproduction of voles. Plant biomasses were harvested at the end of each trial. In all trials, the biomasses of graminoids and non‐toxic herbs other than ferns, fireweeds and rosaceous plants were profoundly decimated. Even the biomass of a toxic herb Aconitum lycoctonum decreased largely at pace with the palatable herbs. The least preferred plant categories maintained their biomasses at control levels. Their neutral collective response was created by opposite species‐level trends. Species typical for moist and nutrient‐rich forests suffered from vole grazing, whereas the biomass of species adapted to disturbed habitats increased. In spite of the dramatic changes in the vegetation, the introduced female voles survived throughout the trials and reproduced normally. The young of their first litters survived well and reached the final weights typical for individuals starting to winter as immatures. We conclude that most of the plant biomass found on productive boreal forest floors is potential food for field voles and remains palatable for them even when subjected to recurrent, severe grazing. If nothing else than summer resources were limiting the growth of the field vole populations, the plants currently dominating moist and nutrient‐rich taiga floors could not survive in this habitat.  相似文献

As a part of multidisciplinary anthropological research of population structure on Croatian island of Rab, finger and palm prints from 548 inhabitants (225 males and 323 females) of six villages on Rab were collected and their qualitative dermatoglyphic traits analysed. We calculated frequencies of patterns (whorls, loops and arches) on fingers, frequencies of patterns in the interdigital areas, Thenar and Hypothenar of the palms and frequencies of position of the axial triradius “t” (t, t′, t″ and other t) for male and female population of each village. Statistical analysis was performed by using Chi-square test, and Hiernaux Δg biological distances between pairs of populations were determined. The results of this study show existence of statistically significant diversity among the village populations in qualitative dermatoglyphic traits both for males and females. This variability found in the population from the island of Rab can be explained by influence of the microevolutionary factors through endogamy and the way of migrational flow on the island in course of history. Subpopulation of Barbat in the eastern part of Rab, which is the closest to the mainland, is probably different because of a high migration rate from the mainland during the past centuries. Subpopulations of Lopar, Supetarska Draga and Kampor were the most isolated subpopulations, situated in three different valleys in the west of Rab, and therefore genetic drift could have been more influential there and responsible for noticed differences.  相似文献

There is growing concern that pollution from petrochemical and oil refinery installations in Pancevo (Serbia) has increased the incidence of various diseases including cancer. The aim of our study was to investigate cancer incidence in Pancevo and to compare it with the region of central Serbia. Cancer incidence data were obtained from the corresponding Serbian Cancer Registries. Systematic local monitoring of benzene, toluene and methyl mercaptane in the atmosphere within Pancevo's industrial area indicated that the average monthly and yearly concentrations often exceeded permitted levels proposed by EU standards (5 microgm(-3)). Cancer incidence was lower in Pancevo than in central Serbia (the standardised incidence for all types of cancers in Pancevo was 218.3 and in central Serbia it was 241.7 per 100,000 inhabitants). The available data do not allow us to correlate air pollution in Pancevo with increased cancer incidence.  相似文献
