A rat model for human minimal change nephropathy was obtained by the intravenous injection of adriamycin (ADR) at 5 mg/kg. By using an in vivo electron paramagnetic resonance (EPR) spectrometer operating at 700 MHz, the temporal changes in signal intensities of a nitroxide radical, 4-hydroxyl-2,2,6,6-tetramethylpiperidine-1-oxyl (TEMPOL), in the kidneys of rats with ADR nephropathy were investigated. The decay rate of the EPR signal intensity obtained in the kidney is indicative of the renal reducing ability. It was found that the reducing ability in the kidney declined on the 7th day after ADR administration and recovered after the 14th day. Impairment of the reducing ability occurred before the appearance of continuous urinary protein. The in vitro EPR study showed that this impairment of in vivo renal reducing ability is related to impairment of the reducing ability in the mitochondria. 相似文献

In teleosts, the distribution of neurons in the preoptic-hypothalamic region and their associated neurohypophysial hormones, such as vasotocin (VT), appears to be different among species. This differential distribution is thought to reflect the social and/or sexual status of individuals within a species. In the present study, we analyzed the number, size and the distribution of vasotocin/isotocin (VT/IT) neurons in the brains of both male and female medaka (Oryzias latipes) using immunohistochemistry. VT/IT neurons were similarly located in an inverted L-shape in the nucleus preopticus in both gender, as has been already reported in salmonids. However, computer-assisted image analysis revealed sexual dimorphism in the number of VT/IT-immunoreactive (ir) neurons, with greater numbers found in males as compared to females. Further, in the female brain, the number of VT/IT-ir neurons decreased significantly after spawning. In pre-spawning compared to post-spawning females, the small-sized VT/IT-ir neurons dominated. Sexual differentiation of the medaka is fully dependent upon the steroid status during the early developmental stages and steroids are also known to trigger gender-specific behavior in the adult medaka. Our findings strongly suggest that VT and/or IT neurons may be functionally related to ovulation and/or the reproductive axes through connections to their steroidal status. 相似文献

Yeasts that ferment both hexose and pentose are important for cost-effective ethanol production. We found that the soil yeast strain NY7122 isolated from a blueberry field in Tsukuba (East Japan) could ferment both hexose and pentose (d-xylose and l-arabinose). NY7122 was closely related to Candida subhashii on the basis of the results of molecular identification using the sequence in the D1/D2 domains of 26S rDNA and 5.8S-internal transcribed spacer region. NY7122 produced at least 7.40 and 3.86 g l−1 ethanol from 20 g l−1 d-xylose and l-arabinose within 24 h. NY7122 could produce ethanol from pentose and hexose sugars at 37°C. The highest ethanol productivity of NY7122 was achieved under a low pH condition (pH 3.5). Fermentation of mixed sugars (50 g l−1 glucose, 20 g l−1 d-xylose, and 10 g l−1 l-arabinose) resulted in a maximum ethanol concentration of 27.3 g l−1 for the NY7122 strain versus 25.1 g l−1 for Scheffersomyces stipitis. Summary Intracellular localization of calmodulin was examined in the budding yeast,Saccharomyces cerevisiae. Distribution of calmodulin changes in a characteristic way during the cell cycle. Calmodulin localizes to a patch at the presumptive bud site of unbudded cells. It concentrates at the bud tip in small-budded cells, and later it diffuses throughout the entire bud. At cytokinesis, calmodulin is largely at the neck between the mother and daughter cells. Double staining experiments have shown that the location of some polarized actin dots is coincident with that of calmodulin dots. Polarized localization of actin dots is observed in cells depleted of calmodulin, suggesting that calmodulin is not required for localization of the actin dots. Thecdc24 mutant that has a defect in bud assembly at the restrictive temperature fails to exhibit polarized localization of calmodulin, indicating that theCDC24 gene product is responsible for controlling the polarity of calmodulin. 相似文献

Saccharomyces cerevisiae is a multifunctional molecular switch involved in establishment of cell morphogenesis. We systematically characterized isolated temperature-sensitive mutations in the RHO1 gene and identified two groups of rho1 mutations (rho1A and rho1B) possessing distinct functional defects. Biochemical and cytological analyses demonstrated that mutant cells of the rho1A and rho1B groups have defects in activation of the Rho1p effectors Pkc1p kinase and 1,3-beta-glucan synthase, respectively. Heteroallelic diploid strains with rho1A and rho1B mutations were able to grow even at the restrictive temperature of the corresponding homoallelic diploid strains, showing intragenic complementation. The ability to activate both of the essential Rho1p effector proteins was restored in the heteroallelic diploid. Thus, each of the complementing rho1 mutation groups abolishes a distinct function of Rho1p, activation of Pkc1p kinase or 1,3-beta-glucan synthase activity. 相似文献

Cultured human neuroblastoma (GOTO) cells were induced to differentiate by dibutyryl cyclic AMP (Bt2cAMP) and/or retinoic acid (RA). A combination of Bt2cAMP (1 mM) and RA (1 microM) yielded the most significant networks of neurites after 3 to 4 days, this being associated with the reduction of N-myc mRNA levels. Next, we examined several cellular genes that were possibly linked with changes in N-myc gene expression under these conditions. Among the genes examined, both nucleolin and a major heat-shock protein (hsp70) mRNAs showed changes concomitant with those in N-myc mRNA levels when induced by Bt2cAMP and RA. Dibutyryl cAMP alone induced several short cellular processes and caused a marked decrease in N-myc mRNA within 2 days. RA alone induced a few long and straight neurites along the longitudinal axis of individual cells and a significant decrease in growth rate but showed neither network formation nor a decrease in N-myc gene expression. These results indicate differential effects of Bt2cAMP and RA on the regulatory mechanisms of both cell proliferation and differentiation and also indicate a possible association of expression of N-myc gene with those of hsp70 and nucleolin genes. 相似文献

During meiosis, VDE (PI-SceI), a homing endonuclease in Saccharomyces cerevisiae, introduces a double-strand break (DSB) at its recognition sequence and induces homologous recombinational repair, called
homing. Meiosis-specific RecA homolog Dmc1p, as well as mitotic RecA homolog Rad51p, acts in the process of meiotic recombination,
being required for strand invasion and exchange. In this study, recruitment of Dmc1p and Rad51p to the VDE-induced DSB repair
site is investigated by chromatin immunoprecipitation assay. It is revealed that Dmc1p and Rad51p are loaded to the repair
site in an independent manner. Association of Rad51p requires other DSB repair proteins of Rad52p, Rad55p, and Rad57p, while
loading of Dmc1p is facilitated by the different protein, Sae3p. Absence of Tid1p, which can bind both RecA homologs, appears
specifically to cause an abnormal distribution of Dmc1p. Lack of Hop2, Mnd1p, and Sae1p does not impair recruitment of both
RecA homologs. These findings reveal the discrete functions of each strand invasion protein in VDE-initiated homing, confirm
the similarity between VDE-initiated homing and Spo11p-initiated meiotic recombination, and demonstrate the availability of

The identification of carpronium chloride and a metabolite in human urine has been performed by means of field desorption mass spectrometry using deuterium labelling. Essential points in this study are the simultaneous administration of a deuterium labelled drug ([2H9]carpronium chloride), a purification procedure by ion pair extraction with an iodine reagent, and the use of paper electrophoresis to examine the degree of clean-up. The field desorption mass spectra of the purified extracts obtained from sample urine gave a characteristic pattern resulting from the carpronium cation (m/z 160, m/z 169) and a metabolite of the N-(3-carbohydroxypropyl)trimethyl ammonium cation (m/z 146, m/z 155). 相似文献
