BackgroundThe Gambia initiated a control programme for schistosomiasis in 2015. In light of this, recent and comprehensive data on schistosomiasis is required to effectively guide the control programme. This study aimed to evaluate the prevalence and associated risk factors of schistosomiasis among primary school children in The Gambia.MethodsWe utilised data from a previous study conducted in 2015 in 4 regions of The Gambia: North Bank Region (NBR), Lower River Region (LRR), Central River Region (CRR) and Upper River Region (URR). In the parent study, ten schools were selected randomly from each region. Urine and stool samples collected from 25 boys and 25 girls (7–14 years) in each school were examined for urinary schistosomiasis (Schistosoma haematobium infection) and intestinal schistosomiasis (Schistosoma mansoni infection) using urine filtration, dipstick and Kato-Katz methods.Principal findingsUrinary schistosomiasis had an overall prevalence of 10.2% while intestinal schistosomiasis had a prevalence of 0.3% among the sampled school children. Prevalence of urinary schistosomiasis was significantly different among regions (χ 2 = 279.958, df = 3, p < 0.001), with CRR (27.6%) being the most endemic region, followed by URR (12.0%), then LRR (0.6%), and NBR (0.0%). Prevalence of intestinal schistosomiasis was also significantly variable among regions, with 4 of the 5 positive cases detected in CRR and 1 case in URR. Every school sampled in CRR had at least one student infected with S. haematobium, 50% of schools in URR had S. haematobium infection, and just one school in LRR had S. haematobium infection. While S. haematobium infection was significantly higher in boys (χ 2 = 4.440, df = 1, p = 0.035), no significant difference in infection rate was observed among age groups (χ 2 = 0.882, df = 2, p = 0.643). Two of the 5 students infected with S. mansoni were boys and 3 were girls. Four of these 5 students were in the 10–12 years age group and 1 was in the 7–9 years age group. Macrohaematuria and microhaematuria were found to be statistically associated with presence of S. haematobium eggs in urine. Being a male was a risk factor of S. haematobium infection. Bathing, playing and swimming in water bodies were found to pose less risk for S. haematobium infection, indicating that the true water contact behaviour of children was possibly underrepresented.ConclusionThe findings of this study provide invaluable information on the prevalence of schistosomiasis in The Gambia. This was useful for the schistosomiasis control efforts of the country, as it guided mass drug administration campaigns in eligible districts in the study area. More studies on S. mansoni and its intermediate snail hosts are required to establish its true status in The Gambia. As children sometimes tend to provide responses that potentially please the research or their teacher, data collection frameworks and approaches that ensure true responses in studies involving children should be devised and used.  相似文献

Background

Chronic schistosomiasis is associated with T cell hypo-responsiveness and immunoregulatory mechanisms, including induction of regulatory T cells (Tregs). However, little is known about Treg functional capacity during human Schistosoma haematobium infection.

Methodology

CD4+CD25hiFOXP3+ cells were characterized by flow cytometry and their function assessed by analysing total and Treg-depleted PBMC responses to schistosomal adult worm antigen (AWA), soluable egg antigen (SEA) and Bacillus Calmette-Guérin (BCG) in S. haematobium-infected Gabonese children before and 6 weeks after anthelmintic treatment. Cytokines responses (IFN-γ, IL-5, IL-10, IL-13, IL-17 and TNF) were integrated using Principal Component Analysis (PCA). Proliferation was measured by CFSE.

Principal Findings

S. haematobium infection was associated with increased Treg frequencies, which decreased post-treatment. Cytokine responses clustered into two principal components reflecting regulatory and Th2-polarized (PC1) and pro-inflammatory and Th1-polarized (PC2) cytokine responses; both components increased post-treatment. Treg depletion resulted in increased PC1 and PC2 at both time-points. Proliferation on the other hand, showed no significant difference from pre- to post-treatment. Treg depletion resulted mostly in increased proliferative responses at the pre-treatment time-point only.

Conclusions

Schistosoma-associated CD4+CD25hiFOXP3+Tregs exert a suppressive effect on both proliferation and cytokine production. Although Treg frequency decreases after praziquantel treatment, their suppressive capacity remains unaltered when considering cytokine production whereas their influence on proliferation weakens with treatment.  相似文献

17α-substituted ethynylestradiols, derived from estrone, were converted to their corresponding 17α-(bromo- or iodo-propargyl)estrone intermediates. Nucleophilic substitution onto these moieties with malonate diester followed by hydrolysis and complexation with cis-Pt(Me2en)I2 (Me2en = N,N-dimethylethylenediamine) gave cis-Pt(Me2en)(2-(3-(17β-estradiol-17α-yl)-prop-2-ynyl)malonato) 7, thus demonstrating that these estrogen-derived compounds can be used to synthesize stable Pt(II) complexes. The 3-(17β-estradiol-17α-yl)-prop-2-ynyl-1-sulfanylethylthiol 23 was also prepared.  相似文献

Substituted 1-hydroxy-4,4-dialkyl-3-oxo-3,4-dihydronaphthalene benzothiadiazine derivatives were investigated as inhibitors of genotype 1 HCV polymerase. Background

Harvest index (HI), the ratio of grain yield to total biomass, is considered as a measure of biological success in partitioning assimilated photosynthate to the harvestable product. While crop production can be dramatically improved by increasing HI, the underlying molecular genetic mechanism of HI in rapeseed remains to be shown.

Results

In this study, we examined the genetic architecture of HI using 35,791 high-throughput single nucleotide polymorphisms (SNPs) genotyped by the Illumina BrassicaSNP60 Bead Chip in an association panel with 155 accessions. Five traits including plant height (PH), branch number (BN), biomass yield per plant (BY), harvest index (HI) and seed yield per plant (SY), were phenotyped in four environments. HI was found to be strongly positively correlated with SY, but negatively or not strongly correlated with PH. Model comparisons revealed that the A–D test (ADGWAS model) could perfectly balance false positives and statistical power for HI and associated traits. A total of nine SNPs on the C genome were identified to be significantly associated with HI, and five of them were identified to be simultaneously associated with HI and SY. These nine SNPs explained 3.42 % of the phenotypic variance in HI.

Conclusions

Our results showed that HI is a complex polygenic phenomenon that is strongly influenced by both environmental and genotype factors. The implications of these results are that HI can be increased by decreasing PH or reducing inefficient transport from pods to seeds in rapeseed. The results from this association mapping study can contribute to a better understanding of natural variations of HI, and facilitate marker-based breeding for HI.

The composition of plant communities everywhere now likely comprises alien as well as native species, and those aliens that become invasive have wide-ranging impacts on the structure and function of recipient ecosystems. These impacts include perturbations to soil carbon (C) cycling, but the direction and magnitude of impacts are species and climate dependent, making it difficult to generalize whether a specific invader will promote losses or gains in soil C stocks. Generalizations of a specific invader??s impacts are necessary; however, because the range of an invader can encompass thousands of square kilometers, meaning their effects can have broad, regional consequences. To quantify broad-scale and context-dependent impacts of a specific invader, multi-site investigations that capture and measure local and regional environmental heterogeneity are necessary. Using this approach, we show that a widespread grass invader of forest understories is associated with declines in soil C during infilling (spreading within the invaded range). Across the 36 study sites, total soil C stocks declined (P?=?0.113) by approximately 12% (estimated mean?±?SD, uninvaded: 2,429?±?512.9 vs. invaded: 2,140?±?520.7?g?C?m?2). The decline in total soil C is driven by a significant (P?=?0.047) reduction in the native-derived, mineral-associated soil C fraction. This fraction, whose mass and slow turnover makes it an important C store, is approximately 15% lower in invaded (estimated mean?±?SD: 1,560?±?400.4?g?C?m?2) than uninvaded plots (1,826?±?398.1?g?C?m?2). Notably, declines in this C fraction are only apparent at 21 of the sites, reflecting how environmental heterogeneity in other variables (specifically pH, soil moisture, and clay content) are important to quantify to determine invader impacts across a region. The 26% decline in microbial biomass with invasion (P?=?0.011; estimated mean?±?SD, uninvaded: 10.05?±?1.79 vs. invaded: 7.40?±?1.80?g?C?m?2) is also dependent on site characteristics (pH), and reductions are greater where the invader occurs at higher densities. Reductions in microbial biomass and soil C with invasion suggest that grass invasion will alter soil C cycling and decrease forest-C stores across the study region, although invader effects at a specific-site will be dependent on environmental context.  相似文献

Many primates now live in anthropogenic landscapes dominated by human activity such as agriculture. Conserving primates in such contexts requires detailed information about habitat use, including landscape features that may influence population viability. We studied Northeast Bornean orangutan (Pongo pygmaeus morio) habitat use in a forestry plantation in East Kalimantan, Indonesia. We conducted camera trapping and nest surveys at 13 locations across three habitat types in the plantation (planted acacia stands, planted eucalyptus stands, and secondary forest patches left uncut or allowed to regenerate) September 2012–March 2013, and calculated four measures of orangutan abundance for each location (independent photo captures/100 camera trap days, or RAI2; nest encounter rate; nest density; and orangutan density). Orangutans are relatively common in the plantation; they used all three habitat types and exhibited a higher RAI2 than 70% of other mammal species detected. A logistic regression found that proximity to natural forest areas best predicted orangutan abundance calculated using camera trap data (RAI2) but that habitat type combined with distance to natural forest best predicted orangutan abundance calculated using nest counts. This suggests that orangutans use planted areas for movement and feeding, but rely on patches of natural forest for resting and access to key resources. Our study and others indicate that orangutans can coexist with some human activities if provided with sufficient access to natural forest. However, we must conduct further research to facilitate effective conservation planning, including gathering additional details about habitat and resource use and possible long-term population impacts.  相似文献

Background

The protection afforded by human erythrocyte polymorphisms against the malaria parasite, Plasmodium falciparum, has been proposed to be due to reduced ability of the parasite to invade or develop in erythrocytes. If this were the case, variable levels of parasitaemia and rates of seroconversion to infected-erythrocyte variant surface antigens (VSA) should be seen in different host genotypes.

Methods

To test this hypothesis, P. falciparum parasitaemia and anti-VSA antibody levels were measured in a cohort of 555 asymptomatic children from an area of intense malaria transmission in Papua New Guinea. Linear mixed models were used to investigate the effect of α+-thalassaemia, complement receptor-1 and south-east Asian ovalocytosis, as well as glucose-6-phosphate dehydrogenase deficiency and ABO blood group on parasitaemia and age-specific seroconversion to VSA.

Results

No host polymorphism showed a significant association with both parasite prevalence/density and age-specific seroconversion to VSA.

Conclusion

Host erythrocyte polymorphisms commonly found in Papua New Guinea do not effect exposure to blood stage P. falciparum infection. This contrasts with data for sickle cell trait and highlights that the above-mentioned polymorphisms may confer protection against malaria via distinct mechanisms.  相似文献
