Pseudoalteromonas sp. 1020R, isolated from the Pacific coast of Japan, produces prodigiosin family pigments. Structural analysis indicated that these are prodigiosin (2-methyl-3-pentyl-prodiginine) and three other prodigiosin congeners which differ only in the lengths of the alkyl side chains. These compounds exhibited different extents of cytotoxicity against U937 leukemia cells, and cell death was accompanied by typical features of apoptosis.  相似文献

The effects of abscisic acid (ABA) on the size of the apertureof stomata on epidermal strips of Vicia faba were studied inincubation media with different pH values. The osmotic potentialof guard cells, as determined by the limiting plasmolysis method,was higher at pH 4.0 than at pH 6.0, although the size of thestomatal apertures was almost identical at both pH values. AtpH 4.0, ABA effectively caused stomatal closure but had onlya small effect on the osmotic potential, whereas, at pH 6.0,ABA significantly increased the osmotic potential. ABA promotedthe efflux of Cl and malate from epidermal strips intothe incubation medium, an effect which was more marked at pH6.0, with a concomitant efflux of K+ to balance the charge onthe exported anions. Polylactic acid (PLA) is a biodegradable polymer, currently used in pharmaceutical and surgical devices. There is a concern that cyclic polylactic acid (CPLA), which is a by-product of PLA synthesis, may be introduced into the human body as an undesirable contaminant. We carried out a quantitation investigation of the CPLA heptamer (CPLA-7) by liquid chromatography mass spectrometry (LC-MS). We found that CPLA-7 binds strongly with serum proteins and that only 62% of CPLA-7 was recovered after routine deproteination; therefore, we directly injected serum into the LC-MS/MS system after passage through a bovine serum albumin (BSA)-coated chromatographic column and found the recovery of CPLA-7 was improved to 84%, and that the detection (S/N=3) and quantitation limit (S/N=10 and below 15% relative standard deviation) were 1.5 and 2.5 ng/mL, respectively. We conclude that direct injection LC-MS/MS, using a BSA column, is a simple and effective quantitative analysis method for CPLA in serum.  相似文献

The production of violacein by Pseudoalteromonas sp. 520P1 has many features of quorum sensing. Signaling molecules were extracted from bacterial culture and subsequently identified as N-(3-oxooctanoyl)-homoserine lactone and N-tetradecanoyl-homoserine lactone. The former but not the latter induced the production of violacein in strain 520P1. We conclude that N-(3-oxooctanoyl)-homoserine lactone is a signaling molecule involved in the production of violacein.  相似文献

In rice, alday, wheat and tobacco (Nicotiana tabacum L. Samsunand Samsun NN) plants which contained large amounts of ABA,the transpiration rate decreased rapidly with 2 ppm SO2 fumigationand reached 20 to 65% of the initial level after 5- to 30-minexposure depending on their ABA contents. In the cases of broadbean and tobacco (N. glutinosa L.) with low ABA contents, therate slightly increased for 20 and 40 min, respectively, afterthe start of the fumigation and then decreased gradually. Thetranspiration rates of corn and sorghum, in spite of their extremelylow ABA contents, pronouncedly decreased with SO2 fumigationand reached 65 and 50%, respectively, of the initial levelsafter 40-min exposure. Foliar application of 0.04 N HC1 to N.tabacum L. Samsun NN leaves remarkably depressed the transpirationrate, while the application of 0.04 M Na2SO3 decreased the rateonly to the same level as water treatment. Foliar applicationof either HCl or Na2SO3 to N. glutinosa L. leaves exerted littlechange in the transpiration rate. When 10–4M ABA was appliedto broad bean leaves prior to HCl and Na2SO3 treatment, theirtranspiration rate was decreased by HCl, but not by Na2SO3 application.In sonicated epidermal strips peeled from broad bean leaves,Na2SO3 produced a slight increase in the stomatal aperture sizein the absence of ABA, but showed no effect in the presenceof ABA. The aperture size was identical in the pH range of 3.0to 7.0 in the incubation medium. In the presence of ABA in themedium, the aperture size was small in the acidic region ofpH with a minimal value at pH 4.0. ABA decreased the aperturesize at concentrations above 10–9 M at pH 4.0 and 10–6M at pH 7.0 in the medium. [2–14C] ABA uptake by epidermalstrips was large in the acidic region, especially at pH 4.0. Thirteen strains of novel marine bacteria producing a purple pigment were isolated from the Pacific coast of Japan. They were divided into two groups based on their 16S ribosomal RNA gene sequences, and both groups of bacteria belonged to the genus Pseudoalteromonas. The UV-visible spectrum of the pigment was identical to those of violacein, a pigment produced by several species of bacteria including Chromobacterium violaceum, an opportunistic pathogen. Further analysis of the chemical structure of the pigment by mass spectroscopy and nuclear magnetic resonance spectroscopy showed that the pigment was violacein. The high purity of violacein in the crude extract enabled us to employ simple and nonpolluting procedures to purify the pigment. Isolated bacteria may be useful as a C. violaceum substitute for the safe production of violacein.  相似文献

Bioluminescence (BL) imaging based on d-luciferin (d-luc)–luciferase reaction allows noninvasive and real-time monitoring of luciferase-expressing cells. Because BL intensity depends on photons generated through the d-luc–luciferase reaction, an approach to increase intracellular levels of d-luc could improve the detection sensitivity. In the present study, we showed that organic anion transporter 1 (OAT1) is useful, as a d-luc transporter, in boosting the BL intensity in luciferase-expressing cells. Functional screening of several transporters showed that the expression of OAT1 in HEK293?cells stably expressing Pyrearinus termitilluminans luciferase (HEK293/eLuc) markedly enhanced BL intensity in the presence of d-luc. When OAT1 was transiently expressed in HEK293?cells, intracellular accumulation of d-luc was higher than that in control cells, and the specific d-luc uptake mediated by OAT1 was saturable with a Michaelis constant (Km) of 0.23?μM. The interaction between OAT1 and d-luc was verified using 6-carboxyfluorescein, a typical substrate of OAT1, which showed that d-luc inhibited the uptake of 6-carboxyfluorescein mediated by OAT1. BL intensity was concentration-dependent at steady states in HEK293/eLuc cells stably expressing OAT1, and followed Michaelis–Menten kinetics with an apparent Km of 0.36?μM. In addition, the enhanced BL was significantly inhibited by OAT1-specific inhibitors. Thus, OAT1-mediated transport of d-luc could be a rate-limiting step in the d-luc–luciferase reaction. Furthermore, we found that expressing OAT1 in HEK293/eLuc cells implanted subcutaneously in mice also significantly increased the BL after intraperitoneal injection of d-luc. Our findings suggest that because OAT1 is capable of transporting d-luc, it can also be used to improve visualization and monitoring of luciferase-expressing cells.  相似文献
