Deleted in Breast Cancer 1 (DBC1) is an important metabolic sensor. Previous studies have implicated DBC1 in various cellular functions, notably cell proliferation, apoptosis, histone modification, and adipogenesis. However, current reports about the role of DBC1 in tumorigenesis are controversial and designate DBC1 alternatively as a tumor suppressor or a tumor promoter. In the present study, we report that polyoma small T antigen (PyST) associates with DBC1 in mammalian cells, and this interaction leads to the posttranslational downregulation of DBC1 protein levels. When coexpressed, DBC1 overcomes PyST-induced mitotic arrest and promotes the exit of cells from mitosis. Using both transient and stable modes of PyST expression, we also show that cellular DBC1 is subjected to degradation by LKB1, a tumor suppressor and cellular energy sensor kinase, in an AMP kinase-independent manner. Moreover, LKB1 negatively regulates the phosphorylation as well as activity of the prosurvival kinase AKT1 through DBC1 and its downstream pseudokinase substrate, Tribbles 3 (TRB3). Using both transient transfection and stable cell line approaches as well as soft agar assay, we demonstrate that DBC1 has oncogenic potential. In conclusion, our study provides insight into a novel signaling axis that connects LKB1, DBC1, TRB3, and AKT1. We propose that the LKB1–DBC1–AKT1 signaling paradigm may have an important role in the regulation of cell cycle and apoptosis and consequently tumorigenesis.  相似文献

Anthemis cotula is a widespread invasive alien species in Kashmir Himalaya. Being a winter annual, the species reproduces entirely by achenes and synchrony between germination requirements of the species and the habitat conditions must be of critical importance in its invasiveness. To examine how the achenes of different ages respond to different environmental cues, two laboratory experiments were performed wherein effects of different nitrogen applications and growth hormones under continuous light and dark conditions were explored. Results show that the achenes are positively photoblastic and have requirement for after‐ripening. Nitrogen applied either as potassium nitrate or as thiourea significantly improved achene germination both under continuous light and dark conditions. Although kinetin (6‐furfurylaminopurine) did not influence achene germination, gibberellic acid (GA3) applied at 1.0 mM concentration had the most significant effect on the final percentage germination under both light and dark conditions. These results suggest that the achenes have an elaborate mechanism of sensing the habitat conditions that helps them to synchronise their germination with favourable environmental conditions – a strategy that aids the species in ensuring recruitment, survival and its spread in Kashmir Himalaya.  相似文献

Arbuscular Mycorrhizas: Drivers or Passengers of Alien Plant Invasion   总被引:2,自引:0,他引:2  
Observational and manipulative studies have revealed that alien plant invasions are an outcome of interplay between a myriad of biotic and abiotic factors operating at various spatio-temporal stages and scales. Despite the salient role of ubiquitous arbuscular mycorrhizal (AM) fungi in plant interactions, studies exploring the role of such symbionts in invasiveness of alien species and invasibility of communities are limited, in part because of difficult-culturablilty of AM fungi on artificial media and apparent complexities in manipulations of AM-plant interactions in field and laboratory experiments. Moreover, analysis of the AM-plant invasion studies conducted so far have yielded contradictory results with some indicating facilitation of invasion by AM fungi and others its inhibition. Other studies have indicated that arbuscular mycorrhizal symbiosis has no effect on invasiveness of alien plants. While arbuscular mycorrhizas may facilitate invasiveness of some alien plants, such plants may also potentially impact mycorrhizal community structure and functions in the invaded habitats in different ways. The present review addresses these paradoxically conflicting observations in the context of mutualism-commensalism-parasitism gradient that characterizes the relationship between AM fungi and their alien vs. native hosts and also discusses the influence of alien invasive plants on mycorrhizal community structure of invaded ecosystems. Through critical analysis of costs and benefits for invasive plants that associate with AM fungi in their introduced range, invasion-induced shifts in AM mutualism are evaluated in the context of their impact on native biodiversity. Underlining limitations of methodologies and experimental designs usually employed to understand AM-mediated plant invasiveness, we proposes herein some alternative frameworks and experimental approaches to overcome these limitations.  相似文献

Curcumin, a polyphenol, has pharmacological effects including antioxidant, anti-inflammatory and anti-cancer features. In this study, we have performed comparative in vivo evaluations of CDF (curcumin difluorinated) and curcumin in cisplatin-induced nephrotoxicity in rats. Male Wistar rats were divided into four groups: (1) Control; (2) Cisplatin (7 mg/kg body wt, intraperitoneal as a single dose); (3) Cisplatin and CDF (50 mg/rat/day; for 12 days); (4) Cisplatin and curcumin (50 mg/rat/day), for 12 days). Cisplatin treated rats exhibited kidney injury manifested by increased serum N-urea and creatinine (P?<?0.001). Kidney from cisplatin treated rats also exhibited significant increase in malondialdehyde (MDA) and 8-isoprostane levels (P?<?0.001). Treatment with CDF and curcumin prevented the rise in serum N-urea, creatinine, MDA and 8-isoprostane as compared to experimental control group in kidney (P?<?0.05). Compared to curcumin, CDF had greater potential in suppressing cisplatin-induced pro-inflammatory factors NF-κB and COX-2 as well as downstream markers Nrf2 and HO-1 (P?<?0.05) in kidney. The analysis on anion transport markers (OAT1 and OAT3) showed a similar trend (CDF?>?curcumin). CDF could reduce the expression of multi-drug resistance markers OCT1, OCT2, MRP2 and MRP4 to a much greater extent than curcumin (P?<?0.05). We also demonstrate that CDF influenced the expression of p-mTOR, p-p70S6K1, p-4E-BP1 and p-Akt. These data suggest that CDF can potentially be used to reduce the chemotherapy induced nephrotoxicity thereby enhancing the therapeutic window of cisplatin. The results also proved that compared to curcumin, CDF has superior protective effect in nephrotoxicity.  相似文献

Esophageal squamous cell carcinoma (ESCC) is one of the most prevalent cancer in Jammu and Kashmir region of India and has multi-factorial etiology involving dietary habits, genetic factors, and gene environmental interactions. Inactivation of the p16 gene expression by aberrant promoter methylation plays an important role in the progression of esophageal carcinoma. In the present investigation, we have studied the role of p16 promoter methylation in 69 histopathologically confirmed ESCC tissues and compared it with corresponding normal adjacent tissues for DNA methylation in the CpG island in the p16 promoter region by methylation-specific polymerase chain reaction (MSP) and p16 protein expression by immunoblotting. The results showed loss of p16 expression in 67% (46/69) of tumor tissues compared to only 3% in control tissues (2/69). Promoter methylation was observed in 52% (36/69) of tumor tissues and it gradually increased with the increasing severity of histological grades of the cancer (P = 0.0001). Loss of p16 expression with promoter methylation was observed in 26 of 36 cases (72%). Analysis of patients dietary habits revealed a strong association between promoter methylation and high consumption of hot salted tea (P < 0.05) which is a most favourite drink commonly consumed by Kashmiri people.  相似文献

The critically endangered and perennial alpine endemic angiosperms, namely, Aquilegia nivalis, Lagotis cashmeriana and Meconopsis latifolia inhabit such habitats in the Kashmir Himalaya that are characterised by short growing season and heavy snow cover for about 3–4 months during winter season. The seeds of these species under natural conditions experience a long period of pre-chilling during winter prior to their germination in following spring season. Taking cue from such a requirement, present study investigated the effect of chilling and exogenous application of growth hormones, NO3 and NH4 + on total percent germination of otherwise deep-dormant seeds of these species, under alternate light/dark and continuous dark light regimes. Prolonged pre-chilling followed by treatment of seeds with different doses of GA3 had a pronounced stimulatory effect on the total germination percentage in all the three species. In fact, highest germination percentage in A. nivalis was recorded only when pre-chilled seeds were treated with 1.5 mM GA3 under alternate light/dark conditions. Likewise, germination of pre-chilled seeds of L. cashmeriana with ruptured seed coats was improved when treated with various concentrations of GA3. Seed germination in M. latifolia was also favourably influenced by treatment of pre-chilled seeds with GA3 or nitrogen applied either as NO3 or as NH4 + under alternate light/dark conditions. Treatment of seeds with kinetin (6-furfuryl-aminopurine) had no significant influence on germination percentage in any of the three species. Thus, prolonged chilling of seeds followed by their treatment with GA3 under alternate light/dark conditions are the requirements necessary for seed germination in these species.  相似文献

Biodiversity inventories unravel insightful taxonomic and biogeographic patterns that provide valuable inputs in guiding conservation practices and policymaking. Here we present an updated inventory of the native and alien woody flora of the Kashmir Himalaya, India, together with analyses of the patterns of taxonomic composition, geographic distribution, and invasion status of the alien species. The data was assembled from the floristic literature of the last two centuries, supplemented with herbarium records and validated by field surveys over the past decade. In total, the woody flora comprised of 520 species; out of these 322 species were native and 198 alien, yielding a proportion of 38% aliens. Amongst the natives, 16 species are on the IUCN red list 2009. While 37 genera included both native and alien species indicating a potential for hybrid formation, only 15 families were represented by alien species exclusively. Regarding the invasion status, presently 37 alien woody species are naturalized and 7 are invasive in the region. The results show that the Kashmir Himalaya harbours a diverse woody flora. It is concluded that a distinction between the native and the alien flora, as adopted in the present study, is crucial for a meaningful analysis of any biodiversity patterns. We predict that in the near future many alien woody species, presently naturalized in the region, can become serious plant invaders, and therefore need further research efforts and timely management.  相似文献

Recent studies have established the controlling influence of rhizospheric biota, especially arbuscular mycorrhizal fungi (AMF), on colonization and spread of some alien plants in their introduced range. But how AMF from different geographical sources influence traits that contribute to invasiveness, particularly in presence of neighbouring plants of other species, has been rarely investigated. Thus, we compared the influence of some local (Kashmir Himalayan isolates) and non-local (isolates from Rajasthan, India) AMF isolates of Glomus moseae, G. fasciculatum and Gigaspora margarita on vegetative and reproductive attributes of Mayweed Chamomile (Anthemis cotula L.), a highly invasive species in the Kashmir Himalaya, India. We also examined whether or not the neighbouring plant species, namely Daucus carota L. (Apiaceae) alters the mutualistic interaction between the AMF and A. cotula. Pot experiments revealed greater positive impact of the local than the non-local AMF on vegetative as well as reproductive attributes of A. cotula. Experimental field studies showed that the incidence of highly prevalent Arum-type mycorrhizal colonization in natural populations of A. cotula was reduced in presence of D. carota. Besides, the local AMF significantly promoted growth of A. cotula more than D. carota under mixed-culture conditions. These results suggest that the facilitation of some alien plant invasions by AMF needs to be considered together with plant–plant interactions and invasion-induced changes in the soil microbial community.  相似文献
