In this paper a modular model of the GnRH neuron is presented. For the aim of simplicity, the currents corresponding to fast time scales and action potential generation are described by an impulsive system, while the slower currents and calcium dynamics are described by usual ordinary differential equations (ODEs). The model is able to reproduce the depolarizing afterpotentials, afterhyperpolarization, periodic bursting behavior and the corresponding calcium transients observed in the case of GnRH neurons.  相似文献

Behavioral effects of somatostatin-14, and some of its fragments [somatostatin(3–8), somatostatin(9–14), somatostatin(7–10)] after intracerebroventricular (ICV) administration have been investigated in male rats. In a passive avoidance learning test, somatostatin-14 (0.6 nM) given immediately after the learning session increased the avoidance latency at 24 hr after the injection, when compared to a somatostatin(3–8) (0.6 nM)-treated group. However, compared to a saline-treated group, the peptides did not significantly influence the avoidance latency. Somatostatin-14 administered in higher dose (6.0 nM) decreased the avoidance latency compared to the saline-treated group, while its fragments did not influence it. In an open field behavioral test, immediately after the 24-hr passive avoidance test, 6 nM of somatostatin-14 decreased the rearing activity, while the fragments did not influence this behavior. Somatostatin-14 produced barrel rotation in a dose-related manner, but after the injection of a high dose of the peptide (12 nM) all of the animals died in cardiorespiratory failure (apnea, pulmonary oedema). One of the fundamental discoveries of W. Arnold was the detection of thermally stimulated light emission from preilluminated photosynthetic material (Arnold and Sherwood (1957) Proc Natl Acad Sci USA 43: 105–114). This phenomenon, called thermoluminescence (TL), is characteristic of a wide range of materials (semiconductors, minerals, inorganic and organic crystals, and complex biological systems such as the photosynthetic apparatus) which share the common ability of storing radiant energy in thermally stabilized trap states.The original discovery of TL in dried chloroplasts later proved to be a phenomenon common to all photosynthetic organisms: photosynthetic bacteria, cyanobacteria, algae and higher plants. Following the pioneering work of Arnold, considerable effort has been devoted to identification and characterization of photosynthetic TL components. This work has firmly established the participation of various redox states of the water-oxidizing complex and the quinone electron acceptors of Photosystem II in the generation of photosynthetic glow curves. Since TL characteristics are very sensitive to subtle changes in redox properties of the involved electron transport components, the TL method has become a powerful tool in probing a wide range of PS II redox reactions. Abstract: Calcium/calmodulin-stimulated protein kinase II (CaMPK II). a major kinase in brain, has been established to play an important role in neurotransmitter release and organization of postsynaptic receptors, and it is known to be involved in long-term potentiation and memory. Less is known about the function of this enzyme in nonneural cells. Here we report on the production, presence, and phosphorylation of the α-subunit of CaM-PK II in primary cultures of cerebral endothelial cells. These results raise the possibility that α-CaM-PK II can act as one of the key enzymes of calcium-mediated intracellular signaling in the cerebral endothelial cells and suggest that α-CaM-PK II may participate in such basic cellular processes as permeability in physiological and pathological conditions.  相似文献

Endolithic microorganisms (those living inside rocks) occur in hot and cold deserts and exist under extreme environmental conditions. These conditions are discussed on a comparative basis. Quantitative estimates of biomass are comparable in hot and cold deserts. Despite the obvious differences between the hot and cold desert environment, survival strategies show some common features. These endolithic organisms are able to switch rapidly their metabolic activities on an off in response to changes in the environment. Conditions in hot deserts impose a more severe environmental stress on the organisms than in the cold Antarctic desert. This is reflected in the composition of the microbial flora which in hot desert rocks consist entirely of prokaryotic microorganisms, while under cold desert conditions eukaryotes predominate.Proceedings of the Fourth College Park Colloquium on Chemical Evolution:Limits of Life, University of Maryland, College Park, 18–20 October 1978.  相似文献

The calcifying cave inhabitant atmophytic blue-green algaGeitleria calcarea is reported from new localities in Florida and in the Cook Islands.—G. floridana n. sp., is described from caves in Florida. The calcified sheath has the shape of a quadratic prism and is built of crystalline acicular subunits about 0.1 µm in diameter. The subunits mostly form a rhombic lattice pattern but in some cases, they are not distinguishable and then the surface of the sheath is smooth.This paper is dedicated with gratitude to my former teacher, Prof. Dr.Lothar Geitler, for his 80th birthday.  相似文献

Tumors often harbor orders of magnitude more mutations than healthy tissues. The increased number of mutations may be due to an elevated mutation rate or frequent cell death and correspondingly rapid cell turnover, or a combination of the two. It is difficult to disentangle these two mechanisms based on widely available bulk sequencing data, where sequences from individual cells are intermixed and, thus, the cell lineage tree of the tumor cannot be resolved. Here we present a method that can simultaneously estimate the cell turnover rate and the rate of mutations from bulk sequencing data. Our method works by simulating tumor growth and finding the parameters with which the observed data can be reproduced with maximum likelihood. Applying this method to a real tumor sample, we find that both the mutation rate and the frequency of death may be high.  相似文献

UPF3 is a key nonsense-mediated mRNA decay (NMD) factor required for mRNA surveillance and eukaryotic gene expression regulation. UPF3 exists as two paralogs (A and B) which are differentially expressed depending on cell type and developmental stage and believed to regulate NMD activity based on cellular requirements. UPF3B mutations cause intellectual disability. The underlying molecular mechanisms remain elusive, as many of the mutations lie in the poorly characterized middle-domain of UPF3B. Here, we show that UPF3A and UPF3B share structural and functional homology to paraspeckle proteins comprising an RNA-recognition motif-like domain (RRM-L), a NONA/paraspeckle-like domain (NOPS-L), and extended α-helical domain. These domains are essential for RNA/ribosome-binding, RNA-induced oligomerization and UPF2 interaction. Structures of UPF2′s third middle-domain of eukaryotic initiation factor 4G (MIF4GIII) in complex with either UPF3B or UPF3A reveal unexpectedly intimate binding interfaces. UPF3B’s disease-causing mutation Y160D in the NOPS-L domain displaces Y160 from a hydrophobic cleft in UPF2 reducing the binding affinity ∼40-fold compared to wildtype. UPF3A, which is upregulated in patients with the UPF3B-Y160D mutation, binds UPF2 with ∼10-fold higher affinity than UPF3B reliant mainly on NOPS-L residues. Our characterization of RNA- and UPF2-binding by UPF3′s middle-domain elucidates its essential role in NMD.  相似文献
