Intraperitoneal administration of thymopentin, a thymopentin II-derived pentapeptide, had no stable and evident effect in the two anxiety models (elevated plus-maze and licking-conflict test) studied. However, in the elevated plus-maze test thymopentin antagonized the behavioral effects of DMCM, a beta-carboline derivative with anxiogenic properties. Further, it was demonstrated that the licking-conflict test procedure itself produced a significant elevation of plasma corticosterone levels, increased the number of [3H]flunitrazepam and decreased the number of [3H]muscimol binding sites in rat hippocampus. The forced-swimming stress similarly to the licking-conflict test also caused an increase in hippocampal [3H]flunitrazepam binding sites. Although ineffective behaviorally in the tests for anxiety, thymopentin pretreatment effectively reversed the changes in corticosterone levels caused by the licking-conflict test. Moreover, it normalized the changed number of benzodiazepine and GABA receptors after stressful stimuli. It is well known that not all anxiolytic drugs (i.e. buspirone) are equally active in behavioral tests for anxiety. According to our data we propose that thymopentin has stress-protective activity. As in vivo and in vitro thymopentin did not change [3H]-flunitrazepam and [3H]muscimol binding, the direct effect of this peptide on the GABA-benzodiazepine-Cl- ionophore receptor complex is unlikely. The action of this peptide on GABA release and/or metabolism can be suggested.  相似文献

The migratory direction in young passerine migrants is based on innate information, with the geomagnetic field and celestial rotation as references. To test whether the direction of celestial rotation is of importance, hand-raised pied flycatchers in Latvia were exposed during the premigratory period to a planetarium rotating in different directions. During autumn migration, when their orientation behavior was recorded in the local geomagnetic field in the absence of celestial cues, birds that had been exposed to a sky rotating in the natural direction showed a unimodal preference of their south-westerly migratory direction. Birds that had been exposed to a sky rotating in the reversed direction, in contrast, showed a bimodal preference of an axis south-west-north-east. Their behavior was similar to that of pied flycatchers that had been raised without access to celestial cues. In Latvia, the magnetic field alone allows only orientation along the migratory axis, and celestial rotation enables birds to select the correct end of this axis. Our findings show that the direction of rotation is of crucial importance: celestial rotation is effective only if the stars move in the natural direction.  相似文献

Nuclear hormone receptors, such as the ecdysone receptor, often display a large amount of induced fit to ligands. The size and shape of the binding pocket in the EcR subunit changes markedly on ligand binding, making modelling methods such as docking extremely challenging. It is, however, possible to generate excellent 3D QSAR models for a given type of ligand, suggesting that the receptor adopts a relatively restricted number of binding site configurations or ‘attractors’. We describe the synthesis, in vitro binding and selected in vivo toxicity data for γ-methylene γ-lactams, a new class of high-affinity ligands for ecdysone receptors from Bovicola ovis (Phthiraptera) and Lucilia cuprina (Diptera). The results of a 3D QSAR study of the binding of methylene lactams to recombinant ecdysone receptor protein suggest that this class of ligands is indeed recognised by a single conformation of the EcR binding pocket.  相似文献

Summary Positive and negative ion FAB mass spectrometry were found to be useful for the structural analysis of phosphorylated peptides containing multiple O-phosphoseryl residues. The positive ion FAB mass spectra obtained for Ac-Ser(P)-Ser(P)-NHMe and Ac-Ser(P)-Ser(P)-Ser(P)-NHMe showed that -eliminative loss of H3PO4 from the Ser(P)-residue was a major event in the fragmentation of the two phosphopeptides and that successive losses of H3PO4 from the [M+H]+ ion occurred when the Ser(P)-cluster was located at the N-terminus. In contrast, the FAB mass spectrum of Ac-Glu-Ser(P)-Leu-Ser(P)-Ser(P)-Ser(P)-Glu-Glu-NHMe showed only a single loss of H3PO4 from the [M+H]+ ion, with further losses of H3PO4 from internal Ser(P)-residues only occurring when fragmentation of the parent phosphopeptide generated daughter fragments that contained (part of) an N-terminal Ser(P)-residue. A series of novel 2-alkoxy- and 2-aryloxyiminoalkyl trifluoromethanesulfonanilide derivatives have shown significant in vitro parasiticidal activity against the ectoparasites Ctenocephalides felis and Rhipicephalus sanguineus. A number of these compounds also displayed significant in vitro endoparasite activity against the nematode Haemonchus contortus.  相似文献

Granulomatous inflammation typifying Crohn''s disease was centred within or confined to appendices in six patients, two of whom developed lesions attributable to Crohn''s disease elsewhere in the gut. The remaining four patients have remained symptom-free for periods varying from two to six years. Histological evidence of Crohn''s disease was also present in five of 46 appendices available for re-examination in a survey of 63 cases of Crohn''s enterocolitis. It is adduced that appendiceal involvement in Crohn''s disease is not uncommon.  相似文献

The influence of short protein fragments on immobilization stress-induced alterations in neuroendocrine and immune systems (catecholamine content in the striatum, hypothalamus and adrenals, serum corticosterone concentration, specific antibody producing activity) was investigated. Immunoglobulin G fragments--tuftsin, rigin, polar amino acid set--polarin and thymus hormone fragment--thymopoetin, as well as substance P (as reference drug) were administered intraperitoneally at doses of 100 and 500 micrograms/kg 30 min before exposure to stress. Rigin and thymopentin showed high stress-protective activity. It is suggested that similar protein fragments, being endogenously formed, may play a regulating role in neuroimmunological homeostasis during exposure to stress.  相似文献
