Paraquat (1,1'-dimethyl-4,4'-bipyridinium), a widely used non-selective herbicide, is a redox cycling agent with adverse effects on dopamine systems. Epidemiological data have shown that exposure to paraquat is one of the several risk factors for Parkinson's disease. We have already shown that cyclo(His-Pro), an endogenous cyclic dipeptide produced by the cleavage of the thyrotropin releasing hormone, has a cytoprotective effect through a mechanism involving Nrf2 activation that decreases production of reactive oxygen species and increases glutathione synthesis. Using primary neuronal cultures and PC12 cells as targets of paraquat neurotoxicity, we addressed whether and how cyclo(His-Pro) causes cellular protective response against paraquat-mediated cell death. We found that cyclo(His-Pro) attenuated reactive oxygen species production, and prevented glutathione depletion by up-regulating Nrf2 gene expression, triggering its nuclear accumulation and activating the expression of heme oxygenase1. These protective effects were abolished by RNA interference-mediated Nrf2 knock down whereas were unaffected by RNA interference-mediated Keap1 knock down. Inhibition of heme oxygenase activity decreased cyclo(His-Pro)-induced neuroprotection. These results suggest that cyclo(His-Pro), acting as a selective activator of the brain modulable Nrf2 pathway, may be a promising candidate as neuroprotective agent that act through induction of phase II genes. 相似文献

Pollen data from 18,000 14C yr bp were compiled in order to reconstruct biome distributions at the last glacial maximum in southern Europe and Africa. Biome reconstructions were made using the objective biomization method applied to pollen counts using a complete list of dryland taxa wherever possible. Consistent and major differences from present‐day biomes are shown. Forest and xerophytic woods/scrub were replaced by steppe, both in the Mediterranean region and in southern Africa, except in south‐western Cape Province where fynbos (xerophytic scrub) persisted. Sites in the tropical highlands, characterized today by evergreen forest, were dominated by steppe and/or xerophytic vegetation (cf. today’s Ericaceous belt and Afroalpine grassland) at the last glacial maximum. Available data from the tropical lowlands are sparse but suggest that the modern tropical rain forest was largely replaced by tropical seasonal forest while the modern seasonal or dry forests were encroached on by savanna or steppe. Swollen vesicles generally 40 micron in diameter were prepared from spinach chloroplasts. These vesicles appear to originate from thylakoids. The present study reports results obtained with individual vesicles using micromanipulative procedures. The electric potential across the membrane was measured with microelectrodes and the pH of the internal space was calculated from the fluorescence of the pH indicator pyranine. The individual vesicles photophosphorylate as measured with luciferin-luciferase. Impalement with microelectrodes did not affect the ability of individual vesicles to photophosphorylate. However, there was no significant membrane potential either with continuous illumination or light flashes. In contrast, we found a delta pH of 3.7 under photophosphorylative conditions and the incubation with the appropriate buffers blocked photophosphorylation presumably by preventing formation of a pH gradient. We propose that, in these vesicles, the membrane potential plays no role in photophosphorylation, whereas a pH gradient is obligatory. 相似文献

In the Western Highlands of Scotland there is a very high incidence of alimentary cancers in cattle. The carcinomas of the upper alimentary canal are found in association with virus-induced benign papillomas, and transformation of papillomas to carcinomas has been observed. Strong circumstantial evidence suggests that the progression to malignancy is due to the interplay between the virus, bovine papillomavirus type 4 (BPV-4), and carcinogen(s) present in bracken fern, which infests the marginal upland grazing grounds. The carcinomas are often accompanied by adenomas and adenocarcinomas of the lower bowels. To elucidate the role of the virus in the transformation process, we have analysed several malignancies of the alimentary canal, and have detected the viral genome in only one case of transforming papilloma of the oesophagus and one case of carcinoma of the tongue. We conclude that, although required for the induction of papillomas, the presence of the BPV-4 DNA is not necessary for the progression to, or the maintenance of, the transformed state. 相似文献

Abstract A strain of Leptospira interrogans was isolated from a patient suffering from leptospirosis and was typed by the Gross Agglutination Absorption test using monoclonal antibodies prepared against different serovars of the Australis serogroup. This newly isolated strain belonged to serovar bratislava . It is the first reported isolation from man, in Italy, of Leptospira bratislava , thus supporting the emerging role of this serovar in human leptospirosis. 相似文献

Abstract The expression of nitrite reductase has been tested in a wild-type strain of Pseudomonas aeruginosa (Pao1) as a function of nitrate concentration under anaerobic and aerobic conditions. Very low levels of basal expression are shown under non-denitrifying conditions (i.e. absence of nitrate, in both aerobic and anaerobic conditions); anaerobiosis is not required for high levels of enzyme production in the presence of nitrate. A Pseudomonas aeruginosa strain, mutated in the nitrite reductase gene, has been obtained by gene replacement. This mutant, the first of this species described up to now, is unable to grow under anaerobic conditions in the presence of nitrate. The anaerobic growth can be restored by complementation with the wild-type gene. 相似文献

The permissive action of glucose (or glyceraldelhyde) is necessary to the glucagon induced insulin release. By collecting every 15 seconds the venous effluent of the perfused and isolated rat pancreas it was observed that the insulin response to glucagon (2 microgram/ml) was immediate if the pancreas was preperfused with low concentration of glucose (5 mM) or glyceraldelhyde (2,5 mM). On the other hand glucagon alone elicited no response, and the insulin discharge occurred 60 to 90 seconds after the addition of glucose (5 mM) or glyceraldelhyde (2,5 mM) this time being probably allowed to the metabolism of the sugar. The pancreatic response to 15 mM glucose occurred also 60 to 90 seconds after the stimulus. On the other hand when the medium contained a low concentration of glucose (or glyceraldelhyde) increasing the glucose concentration by 10 mM provoked an immediate insulin release. This suggests that glucose has two actions differing by their lag phase. One, "permissive", apparent after some delay, mimicked by glyceraldelhyde, necessary for glucagon induced insulin release, is mediated probably by the metabolic products of the sugar. The second, "triggering", initiates instantaneously the insulin release but appears dependent on the first action. 相似文献

The rate of α-methylglucoside efflux from wild-type cells of Escherichia coli K-12 is enhanced by different substrates, as long as they are readily respired. A similar enhancement takes place in strains with impaired oxidative phosphorylation (unc mutants), regardless of their being able (strains AN120, N144, and AN382) or unable (strain NR70) to energize the membrane through respiratory electron flow. The uncouplers carbonylcyanide-m-chlorophenylhydrazone and tetrachlorosalicylanilide do not diminish the efflux acceleration in wild-type strains or unc mutants. However, the stimulation of α-methylglucoside efflux does not occur in the mutant AN59 which cannot perform a normal respiratory electron transport due to a defective synthesis of ubiquinone. The failure to stimulate the efflux is observed with succinate, which is a typical substrate of respiration, as well as with substrates which can yield ATP both at respiratory and substrate levels such as gluconate or glycerol. Moreover, potassium cyanide nullifies the acceleration of α-methylglucoside efflux caused in any type of strain and by any substrate. These results show that neither ATP nor an energized state of the membrane appears to be needed for respiration to accelerate α-methylglucoside release from E. coli cells, and question the existence of any energy-requiring reaction for αMG exit, previously proposed by other authors. 相似文献

Tandem integration of complete and defective SV40 genomes in mouse-human somatic cell hybrids. 总被引:14,自引:0,他引:14
We have analyzed the arrangement of SV40 DNA sequences integrated in human chromosome 7 in two lines of mouse-human somatic cell hybrids: one containing only one human chromosome 7 per cell and the other containing an average of about three. We found that the integration site differs in both the viral and host sequences in the two clones. However, the sites of integration into the several copies of human chromosome 7 of one clone are identical. Each chromosome 7 in both clones carries approximately six viral genomes tandemly linked. Some of these genomes lack about 20% of the DNA from the late region, including the Eco RI site. 相似文献
