The present study was undertaken to comparatively investigate the attachment capacities of Azospirillum brasilenseSp245 and its lipopolysaccharide-defective Omegon-Km mutants KM018 and KM252, as well as their activities with respect to the alteration of the morphology of wheat seedling root hairs. The adsorption dynamics of the parent Sp245 and mutant KM252 strains of azospirilla on the seedling roots of the soft spring wheat cv. Saratovskaya 29 were similar; however, the attachment capacity of the mutant KM252 was lower than that of the parent strain throughout the incubation period (15 min to 48 h). The mutation led to a considerable decrease in the hydrophobicity of the Azospirillumcell surface. The lipopolysaccharides extracted from the outer membrane of A. brasilenseSp245 and mutant cells with hot phenol and purified by chromatographic methods were found to induce the deformation of the wheat seedling root hairs, the lipopolysaccharide of the parent strain being the most active in this respect. The role of the carbohydrate moiety of lipopolysaccharides in the interaction of Azospirillumcells with plants is discussed.  相似文献

PGRP-S (Tag7) is an innate immunity protein involved in the antimicrobial defense systems, both in insects and in mammals. We have previously shown that Tag7 specifically interacts with several proteins, including Hsp70 and the calcium binding protein S100A4 (Mts1), providing a number of novel cellular functions. Here we show that Tag7–Mts1 complex causes chemotactic migration of lymphocytes, with NK cells being a preferred target. Cells of either innate immunity (neutrophils and monocytes) or acquired immunity (CD4+ and CD8+ lymphocytes) can produce this complex, which confirms the close connection between components of the 2 branches of immune response.  相似文献

By methods of difference and derivative spectroscopy it was shown that in etiolated leaves at 77 K three photoreactions of P650 protochlorophyllide take place which differ in their rates and positions of spectral maxima of the intermediates formed in the process: P650R668, P650R688, and P650R697. With an increase of temperature up to 233 K, in the dark, R688 and R697 are transformed into the known chlorophyllide forms C695/684 and C684/676, while R668 disappears with formation of a shorter wavelength form of protochlorophyllide with an absorption maximum at 643–644 nm.Along with these reactions, at 77 K phototransformations of the long-wave protochlorophyllide forms with absorption maxima at 658–711 nm into the main short-wave forms of protochlorophyllide are observed. At 233 K in the dark this reaction is partially reversible. The metabolism of glucose and lactose in Escherichia coli K-12 cells has been studied using a bioelectrochemical (BEC) approach. The magnitude and the duration of the response of a BEC anode were found to be functions of the composition of nutrient media and the concentration of bacterial cells. The amount of electricity that is generated enzymatically during the metabolism of a particular substrate depends on the activity of the relevant enzymes. This suggests that the BEC approach can be used for evaluating the activity of particular enzyme systems.  相似文献

Exopolysaccharides (EPS) synthesized by Paenibacillus polymyxa 1465 in the course of batch cultivation were proven to contain neutral and acidic fractions. EPS are heterogeneous polysaccharides, represented by a complex of macromolecules with molecular mass of 7 × 104 to 2 × 106 Da. The acidic component was shown to be predominant in EPS preparations isolated from bacteria cultivated on glucose, which corresponds to a higher viscosity of EPS water solutions. The exoglycans were shown to contain glucose, mannose, galactose, and uronic acids. Polyclonal rabbit antibodies against the isolated P. polymyxa 1465 EPS preparations were used in a comparative immunodiffusion analysis of a number of P. polymyxa strains.  相似文献

Statistical analysis of the low-frequency (1.0 sec-1 and lower) neuronal impulse activity (IA) meets a few fundamental difficulties. Among them, the most significant is the small number of measurements (interspike intervals) recorded within an acceptable analysis epoch. In our study, we examined the possibility of using the normalized (by its maximum value) informational entropy (Hn) for estimation of the significance of changes in the IA generated by low-frequency neurons of the rostral hypothalamus after electrical stimulation of the prefrontal cortex. We compared the efficiencies of using the U-test (Kolmogorov–Mann–Whitney) and Hn estimate for the analysis of the same samples of neuronal responses. The results allow us to conclude that Hn is a significantly more acceptable estimate for detection of stimulation-induced modifications of the IA generated by low-frequency neurons, as compared with the U-test. The direction of shifts in the Hn value makes it possible to estimate the pattern of neuronal response. This value reflects the state of the neuron and correlates with the type of neuronal responses.  相似文献

The phylogenetic position and generic composition of the moss family Plagiotheciaceae were explored using DNA sequence data from three genomes: plastid trnL‐F and rps4, mitochondrial nad5 intron and nuclear ITS1‐5.8S‐ITS2. Our phylogenetic analyses included 35 terminals from Plagiotheciaceae and 71 outgroup taxa from a representative set of hypnalean moss families. The family Plagiotheciaceae is resolved in the early‐diverging Hypnales grade, together with Fontinalaceae, Habrodontaceae and several genera which are mainly distributed in the area of the former Gondwanan supercontinent. However, monophyly of the family can only be attained if the three Southern Hemisphere genera, Acrocladium, Catagonium and Rhizofabronia, are excluded. Ancestral state reconstruction for eight morphological characters reveals that many characters used to delimit the family, such as a lack of pseudoparaphyllia and rhizoids inserted in the leaf axils, were already present in the ancestor of Hypnales. Dispersal–vicariance analysis suggests that Plagiotheciaceae and Fontinalaceae have their ancestral distributions in the area of the former Laurasian supercontinent. As the analyses also reveal a Gondwanan distribution for the ancestor of Hypnales in general, Plagiotheciaceae and Fontinalaceae represent the first diverging Laurasian lineages in the order. © 2013 The Linnean Society of London  相似文献
