We present the first genome-wide study of recent evolution in Culex pipiens species complex focusing on the genomic extent, functional targets and likely causes of global and local adaptations. We resequenced pooled samples of six populations of C. pipiens and two populations of the outgroup Culex torrentium. We used principal component analysis to systematically study differential natural selection across populations and developed a phylogenetic scanning method to analyse admixture without haplotype data. We found evidence for the prominent role of geographical distribution in shaping population structure and specifying patterns of genomic selection. Multiple adaptive events, involving genes implicated with autogeny, diapause and insecticide resistance were limited to specific populations. We estimate that about 5–20% of the genes (including several histone genes) and almost half of the annotated pathways were undergoing selective sweeps in each population. The high occurrence of sweeps in non-genic regions and in chromatin remodelling genes indicated the adaptive importance of gene expression changes. We hypothesize that global adaptive processes in the C. pipiens complex are potentially associated with South to North range expansion, requiring adjustments in chromatin conformation. Strong local signature of adaptation and emergence of hybrid bridge vectors necessitate genomic assessment of populations before specifying control agents.  相似文献

We provide cytochrome c oxidase subunit 1 (COI) barcode sequences of fishes of the Nayband National Park, Persian Gulf, Iran. Industrial activities, ecological considerations and goals of The Fish Barcode of Life campaign make it crucial that fish species residing in the park be identified. To the best of our knowledge, this is the first report of barcoding data on fishes of the Persian Gulf. We examined 187 individuals representing 76 species, 56 genera and 32 families. The data flagged potentially cryptic species of Gerres filamentosus and Plectorhinchus schotaf. 16S rDNA data on these species are provided. Exclusion of these two potential cryptic species resulted in a mean COI intraspecific distance of 0.18%, and a mean inter- to intraspecific divergence ratio of 66.7. There was no overlap between maximum Kimura 2-parameter distances among conspecifics (1.66%) and minimum distance among congeneric species (6.19%). Barcodes shared among species were not observed. Neighbour-joining analysis showed that most species formed cohesive sequence units with little variation. Finally, the comparison of 16 selected species from this study with meta-data of conspecifics from Australia, India, China and South Africa revealed high interregion divergences and potential existence of six cryptic species. Pairwise interregional comparisons were more informative than global divergence assessments with regard to detection of cryptic variation. Our analysis exemplifies optimal use of the expanding barcode data now becoming available.  相似文献

The proteinase-activated receptors (PARs) are a novel family of G protein-coupled receptors, and their effects in neurodegenerative diseases remain uncertain. Alzheimer's disease (AD) is a neurodegenerative disorder defined by misfolded protein accumulation with concurrent neuroinflammation and neuronal death. We report suppression of proteinase-activated receptor-2 (PAR2) expression in neurons of brains from AD patients, whereas PAR2 expression was increased in proximate glial cells, together with up-regulation of proinflammatory cytokines and chemokines and reduced IL-4 expression (p < 0.05). Glial PAR2 activation increased expression of formyl peptide receptor-2 (p < 0.01), a cognate receptor for a fibrillar 42-aa form of beta-amyloid (Abeta(1-42)), enhanced microglia-mediated proinflammatory responses, and suppressed astrocytic IL-4 expression, resulting in neuronal death (p < 0.05). Conversely, neuronal PAR2 activation protected human neurons against the toxic effects of Abeta(1-42) (p < 0.05), a key component of AD neuropathogenesis. Amyloid precursor protein-transgenic mice, displayed glial fibrillary acidic protein and IL-4 induction (p < 0.05) in the absence of proinflammatory gene up-regulation and neuronal injury, whereas PAR2 was up-regulated at this early stage of disease progression. PAR2-deficient mice, after hippocampal Abeta(1-42) implantation, exhibited enhanced IL-4 induction and less neuroinflammation (p < 0.05), together with improved neurobehavioral outcomes (p < 0.05). Thus, PAR2 exerted protective properties in neurons, but its activation in glia was pathogenic with secretion of neurotoxic factors and suppression of astrocytic anti-inflammatory mechanisms contributing to Abeta(1-42)-mediated neurodegeneration.  相似文献

The dynamics of electric field-induced transient birefringence Deltan(t) and light scattering (detected as turbidity) of 190 nm diameter unilamellar vesicles of dioleoylphosphatidylcholine are investigated as a function of applied field strength E, length of the square pulse Deltat, lipid concentration, mean hydrodynamic diameter , ionic strength, and temperature. Generally, induced birefringence exclusively is observed at low lipid concentration and below certain threshold values of E and Deltat, whereas concomitant induced turbidity appears at high lipid concentration and above thresholds values of E and Deltat. Turbidity is monitored through the change in transmitted intensity DeltaS parallel(t) and DeltaS perpendicular(t) of light polarized parallel and perpendicular to the applied field E. The field-induced structural changes are reflected in double-exponential forward relaxation and triple-exponential reverse relaxation of the positive birefringence, and in non-exponential relaxations of DeltaS parallel (t) and DeltaS perpendicular(t). Under the field, the associated physical events are interpreted as elongation of the spherical bilayer shells in the direction of E, linear chain formation (pearling) of the induced dipolar liposomes parallel to E, and partial fusion of adjoining vesicles within the chains. Under conditions where electroporation can be detected, pore opening succeeds the elongation of the vesicles. After termination of the field, the vesicles return to their original time average spherical shape, the oriented chains randomize and disintegrate, and the fused structures are converted either to unilamellar or multilamellar vesicles.  相似文献

Hydatid cyst caused by Echinococcus granulosus is one of the most important parasitic diseases around the world and many countries in Asia, including Iran, are involved with this infection. This disease can cause high mortality in humans as well as economic losses in livestock. To date, several molecular methods have been used to determine the genetic diversity of E. granulosus. So far, identification of E. granulosus using real-time PCR fluorescence-based quantitative assays has not been studied worldwide, also in Iran. Therefore, the aim of this study was to investigate the genetic diversity of E. granulosus from center of Iran using real-time PCR method. A total of 71 hydatid cysts were collected from infected sheep, goat, and cattle slaughtered in Isfahan, Iran during 2013. DNA was extracted from protoscolices and/or germinal layers from each individual cyst and used as template to amplify the mitochondrial cytochrome c oxidase subunit 1 gene (cox1) (420 bp). Five cattle isolates out of 71 isolates were sterile and excluded from further investigation. Overall, of 66 isolates, partial sequences of the cox1 gene of E. granulosus indicated the presence of genotypes G1 in 49 isolates (74.2%), G3 in 15 isolates (22.7%), and G6 in 2 isolates (3.0%) in infected intermediate hosts. Sixteen sequences of G1 genotype had microgenetic variants, and they were compared to the original sequence of cox1. However, isolates identified as G3 and G6 genotypes were completely consistent with original sequences. G1 genotype in livestock was the dominant genotype in Isfahan region, Iran.  相似文献

The polar headgroup contribution to monolayer behavior of dipalmitoylglucosylglycerol has been examined through studies of 1,2-di-O-palmitoyl-3-O-(alpha-D-glucopyranosyl)-sn-glycerol (di-16:0-alpha GlcDG) and 1,2-di-O-palmitoyl-3-O-(beta-D-glucopyranosyl)-sn-glycerol (di-16:0-beta GlcDG) in which the sugar headgroup is linked via an alpha or beta linkage to the diacylglycerol moiety. The results indicate that the limiting areas per molecule of the resultant condensed states are smaller than those of the corresponding phosphatidylcholine (DPPC) but larger than those of dipalmitoylphosphatidylethanolmine (DPPE). In the expanded state, while the areas per molecule are similar to those of DPPC at low pressures, both glycolipids occupy smaller areas at higher pressures. The expanded-state areas of the glucolipids are also slightly greater than those of DPPE. The initial compressional phase transition pressure of the glucolipid liquid-expanded/liquid-condensed transition (pi t) is, however, less sensitive to temperature than are the pi t values of phospholipids. Both of these effects must relate to strong headgroup/water interactions, which, in turn, result in a stabilization of the liquid-expanded states. In the expanded states the alpha anomers are slightly less tightly packed than the beta anomers, as is indicated by the somewhat higher areas per molecule of the expanded states and the lower transition temperatures. These differences in chain-melting temperatures are slightly smaller than those observed in bilayers. While the areas per molecule of the dipalmitoyl glucolipids are greater than those of dipalmitoylphosphatidylethanolamine, they nevertheless exhibit a greater tendency to form nonbilayer structures. Such observations indicate that other factors besides geometric shape play a role in bilayer/nonbilayer transitions.  相似文献
