Oxidative damage and inflammation are closely associated with the pathogenesis of acute lung injury (ALI). Thus, we explored the protective effect of isovitexin (IV), a glycosylflavonoid, in the context of ALI. To accomplish this, we created in vitro and in vivo models by respectively exposing macrophages to lipopolysaccharide (LPS) and using LPS to induce ALI in mice. In vitro, our results showed that IV treatment reduced LPS-induced pro-inflammatory cytokine secretion, iNOS and COX-2 expression and decreased the generation of ROS. Consistent findings were obtained in vivo. Additionally, IV inhibited H2O2-induced cytotoxicity and apoptosis. However, these effects were partially reversed following the use of an HO-1 inhibitor in vitro. Further studies revealed that IV significantly inhibited MAPK phosphorylation, reduced NF-κB nuclear translocation, and upregulated nuclear factor erythroid 2-related factor 2 (Nrf2) and heme oxygenase 1 (HO-1) expression in RAW 264.7 cells. In vivo, pretreatment with IV attenuated histopathological changes, infiltration of polymorphonuclear granulocytes and endothelial activation, decreased the expression of ICAM-1 and VCAM-1, reduced the levels of MPO and MDA, and increased the content of GSH and SOD in ALI. Furthermore, IV treatment effectively increased Nrf2 and HO-1 expression in lung tissues. Therefore, IV may offer a protective role against LPS-induced ALI by inhibiting MAPK and NF-κB and activating HO-1/Nrf2 pathways.  相似文献

Induction of p21 (WAF1/CIP1/SDI1), a physiological mediator of cell cycle arrest, inhibits multiple genes involved in cell division. We have investigated the determinants of p21- mediated inhibition of two of these genes, polo-like kinase 1 (PLK1) and topoisomerase IIalpha (TOPO IIalpha) p21 expression from an inducible promoter in human HT1080 cells rapidly decreases cellular levels of PLK1 and TOPO IIalpha promoters in transient and stable transfection assays. Promoter mutagenesis studies show that inhibition of the PLK1 promoter by p21 is mediated in part by tandem sequences CDE (cell cycle-dependent element) and CHR (cell cycle genes homology region). p21 response of the TOPO IIalpha promoter inhibition and the effects of promoter mutations differ under the conditions of growth arrest produced by p21 induction or by mimosine, a cell cycle inhibitor that increases p21 RNA but not protein expression in HT1080 cells. These results indicate that inhibition of cell division-associated genes by p21 is mediated by different but overlapping mechanisms, which are not a general con-sequence of cell cycle arrest.  相似文献

siRNA (small interfering RNA) and shRNA (small hairpin RNA) are powerful and commonly used tools in biomedical research. Currently, siRNAs are generally designed as two 21 nt strands of RNA that include a 19 nt completely complementary part and a 2 nt overhang. However, since the si/shRNAs use the endogenous miRNA machinery for gene silencing and the miRNAs are generally 22 nt in length and contain multiple internal mismatches, we tested if the functionality can be increased by designing the si/shRNAs to mimic a miRNA structure. We systematically investigated the effect of single or multiple mismatches introduced in the passenger strand at different positions on siRNA functionality. Mismatches at certain positions could significantly increase the functionality of siRNAs and also, in some cases decreased the unwanted passenger strand functionality. The same strategy could also be used to design shRNAs. Finally, we showed that both si and miRNA structured oligos (siRNA with or without mismatches in the passenger strand) can repress targets in all individual Ago containing cells, suggesting that the Ago proteins do not differentiate between si/miRNA-based structure for silencing activity.  相似文献

A bacterial strain Pseudomonas sp. a3 capable of degrading nitrobenzene, phenol, aniline, and other aromatics was isolated and characterized. When nitrobenzene was degraded, the release of NH(4) (+) was detected, but not of NO(2) (-). This result implied that nitrobenzene might have a partial reductive metabolic pathway in strain a3. However, aniline appeared as one of the metabolites during the aerobic degradation of nitrobenzene. Moreover, the appearance of 2-aminophenol during aniline degradation by strain a3 indicated that novel initial reactions existed during the degradation of nitrobenzene and aniline by strain a3. Strain a3 was immobilized in the mixed carrier of polyvinyl alcohol and sodium alginate to improve its degrading efficiency. The optimal concentrations of polyvinyl alcohol and sodium alginate in the mixed carrier were 9 and 3 %, respectively. The immobilized cells had stable degradation activity and good mechanical properties in the recycling tests. The immobilized cells also exhibited higher tolerances in acidic (pH 4-5) and highly saline (10 % NaCl) environments than those of free cells. The biodegradation of nitrobenzene mixed with aniline and phenol using immobilized cells of Pseudomonas sp. a3 was also greatly improved compared with those of free cells. The immobilized cells could completely degrade 300 mg L(-1) nitrobenzene within 10 h with 150 mg L(-1) aniline and 150 mg L(-1) phenol. This result revealed that the immobilized cells of Pseudomonas sp. a3 could be a potential candidate for treating nitrobenzene wastewater mixed with other aromatics.  相似文献

In our previous work, supramolecular films composed of hydrophilic cellulose and hydrophobic polyaniline (PANI) dissolved in NaOH/urea aqueous solution at low temperature through rearrangement of hydrogen bonds have been constructed. To further understand the miscibility and processability of the complex solution, the dynamic rheological behaviors of the PANI/cellulose complex solution were investigated, for the first time, in the present work. Transmission electron microscope (TEM) results demonstrated that the inclusion complexes consisted of PANI and cellulose, existed in the aqueous solution, showing a good miscibility. Time-temperatures superposition (tTs) results indicated that the PANI/cellulose solution exhibited a homogeneous system, and the complex solution was more stable than the cellulose solution in the temperature range from 5 to 25 °C. Winter-Chambon theory was proved to be capable of describing the gelation behavior of the PANI/cellulose complex solution. The relaxation exponent at the gel point was calculated to be 0.74, lower than the cellulose solution, indicating strong interactions between PANI and cellulose chains. Relatively larger flow activation energy of the PANI/cellulose solution suggested the formation and rupture of linkages in "junction zones" during the gelation processes. Furthermore, PANI/cellulose gels could form at elevated temperature as a result of the physical cross-linking and chain entanglement, and it was a thermoirreversible process. Moreover, the PANI/cellulose solution remained a liquid state for a long time at the temperature range from 0 to 8 °C, which is important for the industry process.  相似文献

Different proxies have been used to investigate Quaternary paleoclimate change. Here, we used weathering of calcium nodules in paleosols on China’s Loess Plateau as a proxy for Quaternary paleoclimate changes to provide an alternative indicator of these changes. Paleosol and carbonate nodules were collected from Luochuan and Lantian counties in Shaanxi Province, China. We found that this approach allowed quantitative reconstruction of temperature, rainfall, soil mineral composition, and the effects of weathering and leaching. The changes in carbonate content in the loess and paleosol sequences were controlled by alternating dry and wet climatic conditions. Nodule formation conditions were directly affected by the leaching and migration of elements. The loess and paleosol sequences developed calcium nodules, and their formation was closely related to the rainfall and leaching characteristics of the paleoclimate. The paleoclimate and soil minerals affected the vegetation types and directly influenced changes in the soil. During formation of the calcium nodules, the surface vegetation evolved slowly, and the number of species and quantity of vegetation both decreased.  相似文献
