Although the conversion of lactate to glycogen (glyconeogenesis) in muscle was demonstrated a long time ago, the biochemical reactions responsible for this process are still a controversial matter. In the present study, advantage was taken from the specific inhibition induced by phenylalanine on muscle pyruvate kinase (PK) to investigate the role of reverse PK activity in muscle glyconeogenesis. Addition of phenylalanine to the incubation medium of a preparation of isolated, intact skeletal muscles that maintain metabolic activity for several hours reduced by 50% the rate of incorporation of [14C]lactate or [14C]bicarbonate into muscle glycogen. Muscle extracts presented high levels of maximal activity of PK in the reverse direction, which was completely blocked in the presence of phenylalanine. In contrast, mercaptopicolinic acid, an inhibitor of phosphoenolpyruvate carboxykinase (PEPCK), did not affect the incorporation of 14C from either lactate or bicarbonate into muscle glycogen. Maximal PEPCK activity was much lower in muscle extracts than in gluconeogenic or glyceroneogenic tissues and was suppressed in the presence of mercaptopicolinic acid. The data suggest that a reversal of the metabolic flux through the reaction catalyzed by PK contributes to the accumulation of lactate-derived glycogen that occurs in skeletal muscle under certain physiological conditions.  相似文献

The present Venezuelan population is the product of admixture of Amerindians, Europeans, and Africans, a process that was not homogeneous throughout the country. Blood groups, short tandem repeats (STRs), mtDNA, and Y-chromosome markers have been used successfully in admixture studies, but few such studies have been conducted in Venezuela. In this study we aim to estimate the admixture components of samples from two different socioeconomic levels from Caracas, Venezuela's capital city, compare their differences, and infer sexual asymmetry in the European Amerindian union patterns. Gene frequencies for blood groups ABO and Rh (CDE) and for the STRs VWA, F13A01, and FES/FPS and mtDNA and Y-chromosome haplogroups were studied in a sample of 60 individuals living in Caracas, taken from a private clinic (high socioeconomic level), and 50 individuals, also living in Caracas, drawn from a public maternity clinic (low socioeconomic level). The admixture analysis for the five autosomal markers gives a high European component (0.78) and an almost negligible African sub-Saharan component (0.06) for the high socioeconomic level, whereas for the low socioeconomic level the sub-Saharan, European, and Amerindian components were 0.21, 0.42, and 0.36, respectively. Estimates of admixture based on mtDNA and Y-chromosome markers reveal that the Amerindian contribution to these Caracas samples is almost entirely through females, because the Y-chromosome Amerindian and African sub-Saharan chromosomes found in this study were scarce. Our study reveals that the identification of the grandparents' geographic origin is an important methodological aspect to take into account in genetic studies related to the reconstruction of historical events.  相似文献

This paper presents a mathematical model for the propagation of errors in body segment kinematics to the location of the center of rotation. Three functional calibration techniques, usually employed for the gleno-humeral joint, are studied: the methods based on the pivot of the instantaneous helical axis (PIHA) or the finite helical axis (PFHA), and the “symmetrical center of rotation estimation” (SCoRE). A procedure for correcting the effect of soft tissue artifacts is also proposed, based on the equations of those techniques and a model of the artifact, like the one that can be obtained by double calibration. An experiment with a mechanical analog was performed to validate the procedure and compare the performance of each technique. The raw error (between 57 and 68 mm) was reduced by a proportion of between 1:6 and less than 1:15, depending on the artifact model and the mathematical method. The best corrections were obtained by the SCoRE method. Some recommendations about the experimental setup for functional calibration techniques and the choice of a mathematical method are derived from theoretical considerations about the formulas and the results of the experiment.  相似文献

Question: Will the predicted climate changes affect species distribution in the Iberian Peninsula? Location: Iberian Peninsula (Spain and Portugal). Methods: We modelled current and future tree distributions as a function of climate, using a computational framework that made use of one machine learning technique, the random forest (RF) algorithm. This algorithm provided good predictions of the current distribution of each species, as shown by the area under the corresponding receiver operating characteristics (ROC) curves. Species turnover, richness and the change in distributions over time to 2080 under four Intergovernmental panel on climate change (IPCC) scenarios were calculated using the species map outputs. Results and Conclusions: The results show a notable reduction in the potential distribution of the studied species under all the IPCC scenarios, particularly so for mountain conifer species such as Pinus sylvestris, P. uncinata and Abies alba. Temperate species, especially Fagus sylvatica and Quercus petraea, were also predicted to suffer a reduction in their range; also sub‐mediterranean species, especially Q. pyrenaica, were predicted to undergo notable decline. In contrast, typically Mediterranean species appeared to be generally more capable of migration, and are therefore likely to be less affected.  相似文献

The present work proposes new boundaries for the current submediterranean territories of the Iberian Peninsula, defining them at the smallest scale attempted to date. The boundaries proposed are not sharp divisions but somewhat ‘gradual’, reflecting the transitional nature of the territories they encompass. Climate change predictions were used to estimate how the distribution of these submediterranean regions might change in the near future. The maps constructed are based on the distribution of marcescent Quercus species—trees that characterise the submediterranean plant landscape where they form the main forest communities. To determine their climatic range, the distribution of different types of Iberian oak forest was represented in ‘climate diagrams’ (ordination diagrams derived from principal components analysis), both in terms of individual species and groups of species based on leaf ecophysiological type, i.e. marcescent (Submediterranean), sclerophyllous (Mediterranean), semideciduous (Mediterranean) and deciduous (Eurosiberian). The climate range of each type of forest was determined, and the means of representative climate variables are analysed by one way ANOVA. The variables differentiating the forest groups were also examined by discriminant analysis. The range of the climate variables found to be associated with the majority of marcescent forests was used to determine the distribution of territories throughout the Peninsula with the same conditions (i.e. whether marcescent forests were present or not), thus providing a map of the Iberian submediterranean territories. Predictions of climate change were used to investigate possible climate-induced modifications in the boundaries of these territories in the near future. The patterns obtained show dramatic reductions in the extension of the Iberian submediterranean environment. Submediterranean conditions will probably disappear from the areas where they currently reign, and it seems unlikely that any new, large submediterranean areas will form by displacement towards higher altitudes. The outlook for the unique submediterranean vegetation of the Iberian Peninsula is gloomy.

DNA maturation in bacteriophage λ is the process by which the concatemeric precursor DNA is cleaved at sites called cos to generate mature λ DNA molecules. These DNA molecules are then packaged into procapsids, the empty capsid precursors. The enzyme that catalyses these events is λ DNA terminase. It is composed of two subunits, made of 181 and 641 amino acids, the products of genes Nu1 and A, respectively. The product of the FI gene (gpFI ) stimulates the formation of an intermediate in capsid assembly called complex II, which contains a procapsid, terminase and DNA. The mechanism of stimulation remains unknown. It has been suggested that gpFI may also stimulate terminase-mediated cos cleavage, in the absence of procapsids, by increasing enzyme turnover. Mutants in FI fail to mature and package DNA but, in comparison with other capsid gene mutants, FI mutants are leaky. Second site mutants of FI phages, called ‘fin’ (for FI independence), bypass the necessity for gpFI. These mutants were originally localized to the region of Nu1 and A and are of two classes: finA includes those that induce the synthesis of fourfold more gene A product (gpA ) than wild-type phages, and finB includes those that produce normal amounts of gpA. Whereas all finA mutants analysed map to Nu1, finB mutants have been found both in E and in Nu1. The existence of E mutants able to bypass the necessity for gpFI in vivo shows that gpE and gpFI interact, directly or indirectly. Here we have analysed and sequenced two finA mutants and one finB mutant. All of these map in Nu1. Of the two finA mutants, one corresponds to an Ala163Ser change and the other is a silent mutation. It is likely that the finA mutations alter mRNA conformation in a manner that results in an increase in the efficiency of A mRNA translation. The fourfold increase in gpA synthesis translates into a 10-fold increase in terminase activity. These results show that terminase overproduction is sufficient to bypass the necessity for gpFI and that such an overproduction can be achieved by changes in the efficiency of translation of A due to subtle changes in the sequence upstream of the gene. The finBcs103 mutation is a His-87→Tyr change in Nu1. Therefore, an alternative way in which to bypass the requirement for gpFI involves an alteration in the structure of gpNu1. It is likely that the altered gpNu1 would increase cleavage and packaging efficiency directly or indirectly. We have determined that DNA cleavage in vivo does not occur in the absence of gpFI. Therefore it seems that gpFI somehow facilitates an otherwise latent capacity of terminase to autoactivate its nucleolytic activity.  相似文献

Bacteriophage lambda with mutations in genes that control prohead assembly and other head precursors cannot mature their DNA. In this paper we present evidence that the failure of these phage mutants to mature DNA is a reflection of a mechanism that modulates terminase nicking activity during normal phage development. We have constructed plasmids that contain the lambda-cohesive end site (cos) and the genes that code for DNA terminase, the enzyme that matures DNA by cutting at cos. The DNA terminase genes are under control of a thermosensitive cI repressor. These plasmids lack most of the genes involved in prohead morphogenesis and other head precursors. However, when repression is lifted by destruction of the thermosensitive repressor, the terminase synthesized is able to cut almost 100% of the plasmids. Therefore, these plasmids can mature in the absence of proheads and other head gene products. The plasmids are also able to complement mutants of lambda deficient in terminase and DNA maturation. However, in these complementation experiments, if the phage carry mutations in prohead genes E or B, not only is phage DNA maturation blocked, but the plasmid also fails to mature. These experiments show that, in the absence of proheads, phage lambda produces a trans-acting inhibitor of maturation. The genetic determinant of this inhibitor maps in a region extending from the middle of gene B to the end of gene C. A model is proposed in which the nicking activity of DNA-bound terminase is inhibited by the trans-acting inhibitor. Prohead (and other factors) binding to this complex would release the block to allow DNA cleavage and packaging.  相似文献

Hybrid Zones Between Two European Oaks: a Plant Community Approach   总被引:1,自引:1,他引:0  
Phenomena of hybridization can affect the ecology and evolution of the species involved in the process, as well as their communities. Although numerous papers focus upon the problem of taxonomy, few of these have attempted to study hybrid zones in relation to the analysis of their communities. On the Iberian Peninsula, hybridization phenomena among different oak species are frequent. It is, however, between Quercus faginea Lam. and Quercus pubescens Willd. where the most noteworthy hybridization phenomenon occurs. In this respect, we are familiar with the existence of different introgression levels but we are unaware of whether these hybrids are the transitory result of the interspecific genetic flow or whether these are maintained by means of extrinsic selection processes. Study of plant communities’ flora and environment might shed light upon this issue. Comparison between plant communities dominated by one of the parental species and those dominated by individuals of hybrid origin might enable us to establish the presence or absence of an environment that is potentially selective in favour of the hybrids. Thus the possible existence of extrinsic selection. Furthermore, this information will help us to understand plant community distribution in an area␣that is difficult to interpret. To this purpose, we used multivariate ordination techniques (DCA and CCA) based upon a total of 395 floristic releves covering the whole range of the parental species on the Iberian Peninsula and upon climatic and edaphic variables for each of these releves. We also compared the groups obtained in relation to floristic similarity (Jaccard index), richness and diversity (Shannon–Weaver index). Forests associated with Quercus pubescens are related to heavy summer precipitation, whereas Quercus faginea forests correspond to lower values of this variable and higher ones for continentality. Between both formations, there is a broad hybrid zone, with diffused borders that are related to an environmental gradient of Mediterranean influence. In this region, two types of forest communities were distinguished, which enabled us to divide the hybrid zones into two territories. Our results allowed us to locate the hybrid zone in an ecotone. The differentiation between habitats appears to indicate models of ecological selection. These models require, by definition, the presence of an environmental gradient between the parental zones. We are, however, aware of the need for future experiments in order to establish whether the hybrids are better adapted than the parental species. Only with availability of all this information can intrinsic selection be rejected.  相似文献

We propose to model planar movements between two human segments by means of rolling-without-slipping kinematic pairs. We compute the path traced by the instantaneous center of rotation (ICR) as seen from the proximal and distal segments, thus obtaining the fixed and moving centrodes, respectively. The joint motion is then represented by the rolling-without-slipping of one centrode on the other. The resulting joint kinematic model is based on the real movement and accounts for nonfixed axes of rotation; therefore it could improve current models based on revolute pairs in those cases where joint movement implies displacement of the ICR. Previous authors have used the ICR to characterize human joint motion, but they only considered the fixed centrode. Such an approach is not adequate for reproducing motion because the fixed centrode by itself does not convey information about body position. The combination of the fixed and moving centrodes gathers the kinematic information needed to reproduce the position and velocities of moving bodies. To illustrate our method, we applied it to the flexion-extension movement of the head relative to the thorax. The model provides a good estimation of motion both for position variables (mean R(pos)=0.995) and for velocities (mean R(vel)=0.958). This approach is more realistic than other models of neck motion based on revolute pairs, such as the dual-pivot model. The geometry of the centrodes can provide some information about the nature of the movement. For instance, the ascending and descending curves of the fixed centrode suggest a sequential movement of the cervical vertebrae.  相似文献
