The nuclear DNA content of 38 malignant and 25 benign bone tumours was measured by flow cytometry. The specimens were taken either from biopsies or from surgical specimens. Seventeen of 26 primary malignant bone tumours were aneuploid, 15 had a single aneuploid DNA content, and 2 had a biclonal abnormality. Thirteen of 15 osteosarcomas were aneuploid, but only 2 of 6 chondrosarcomas showed an aneuploid DNA content. Six of 12 metastatic malignant bone tumours were also aneuploid. All 25 benign tumours had a diploid DNA content. Cell cycle analysis showed that the proportion of cells in S- and G2M-phases was higher in the malignant compared to benign tumours, indicating a higher proliferative activity. The increase was statistically significant (p less than 0.05) both in diploid and in aneuploid tumours. Among five tumours studied after chemotherapy, four displayed a marked hyperdiploid abnormality. Aneuploidy and high proliferative activity both were highly associated with malignant bone tumours, suggesting that DNA flow cytometry may be an adjunct in the assessment of malignancy of bone tumours.  相似文献

The disposition of the enantiomers of hydroxychloroquine (HCQ) and its major metabolites in ocular tissues of rabbits has been studied. Both albino, New Zealand White (NZW), and pigmented animals were administered daily oral doses of rac-HCQ, (S)-HCQ or (R)-HCQ (20 mg/kg) over 1, 6, or 8 day periods or for 8 days followed by a 7-day washout period. At the end of the study periods, plasma and whole blood samples were collected and the rabbits were sacrificed. The eyes were collected, the aqueous humor removed with a syringe, and the eyes separated into the cornea, lens, vitreous body, iris, choroid-retina, sclera, and conjunctiva. The concentrations of (R)-HCQ, (S)-HCQ, and their respective metabolites were determined using a validated enantioselective liquid chromatographic assay. The data from these studies indicate that HCQ accumulated in both pigmented and nonpigmented ocular tissues. In the pigmented tissues, HCQ and its metabolites were bound to melanin and the binding was not enantiospecific. In the nonpigmented tissues and in the iris and retina-choroid of the NZW rabbits, the accumulation appeared to be the result of a reversible and enantioselective binding of HCQ and its metabolites to an unidentified biopolymer present in these ocular tissues. © 1994 Wiley-liss, Inc.  相似文献

The role of nitrogen as a factor controllingphytoplankton biomass was studied in nutrientenrichment incubations in the laboratory using waterfrom pelagic region of two mesotrophic lakes ineastern Finland, Lake Kallavesi (in year 1994) andLake Juurusvesi (in year 1995). We used differentcombinations of phosphorus and nitrogen additions ina total of eight experiments. Furthermore, we includedDaphnia grazing treatment to the experimentaldesign in Lake Juurusvesi experiments. The nitrogentreatments did not increase chlorophyll aconcentration in any of the experiments compared withthe controls. Chlorophyll a content was highestin those nutrient treatments where phosphorus wasadded with or without nitrogen. Daphnia grazingdecreased chlorophyll a concentration comparedwith non-grazed treatments. In some cases grazing alsocaused higher ammonium concentrations. Theseexperiments, as well as the nutrient ratio of the lakewater used, suggest that phosphorus is likely tocontrol the amount of phytoplankton biomass. Responding to different questions generated by biodiversity and ecosystem services policy or management requires different forms of knowledge (e.g. scientific, experiential) and knowledge synthesis. Additionally, synthesis methods need to be appropriate to policy context (e.g. question types, budget, timeframe, output type, required scientific rigour). In this paper we present a range of different methods that could potentially be used to conduct a knowledge synthesis in response to questions arising from knowledge needs of decision makers on biodiversity and ecosystem services policy and management. Through a series of workshops attended by natural and social scientists and decision makers we compiled a range of question types, different policy contexts and potential methodological approaches to knowledge synthesis. Methods are derived from both natural and social sciences fields and reflect the range of question and study types that may be relevant for syntheses. Knowledge can be available either in qualitative or quantitative form and in some cases also mixed. All methods have their strengths and weaknesses and we discuss a sample of these to illustrate the need for diversity and importance of appropriate selection. To summarize this collection, we present a table that identifies potential methods matched to different combinations of question types and policy contexts, aimed at assisting teams undertaking knowledge syntheses to select appropriate methods.  相似文献

Under the changing climate, asymmetric warming pattern would be more likely during day and night time, instead of symmetric one. Concurrently, the growth responses and water use of plants may be different compared with those estimated based on symmetric warming. In this work, it was compared with the effects of symmetric (ETs) and asymmetric (ETa) elevation of temperature alone, and in interaction with elevated carbon dioxide concentration (EC), on the grain yield (GY) and evapotranspiration in winter wheat (Triticum aestivum L.) based on pot experiment in the North China Plain (NCP). The experiment was carried out in six enclosed‐top chambers with following climate treatments: (1) ambient temperature and ambient CO2 (CON), (2) ambient temperature and elevated CO2 (EC), (3) elevated temperature and ambient CO2 (ETs; ETa), and (4) elevated temperature and elevated CO2 (ECETs, ECETa). In symmetric warming, temperature was increased by 3°C and in asymmetric one by 3.5°C during night and 2.5°C during daytime, respectively. As a result, GY was in ETa and ETs 15.6 (P < 0.05) and 10.3% (P < 0.05) lower than that in CON. In ECETs and ECETa treatments, GY was 14.9 (P < 0.05) and 9.1% (P < 0.05) higher than that in CON. Opposite to GY, evapotranspiration was 7.8 (P < 0.05) and 17.9% (P < 0.05) higher in ETa and ETs treatments and 7.2 (P < 0.05) and 2.1% (P > 0.05) lower in ECETs and ECETa treatments compared with CON. Thus, GY of wheat could be expected to increase under the changing climate with concurrent elevation of CO2 and temperature as a result of increased WUE under the elevated CO2. However, the gain would be lower under ETa than that estimated based on ETs due to higher evapotranspiration.  相似文献

Background  

During the course of a bacterial infection, the rapid identification of the causative agent(s) is necessary for the determination of effective treatment options. We have developed a method based on a modified broad-range PCR and an oligonucleotide microarray for the simultaneous detection and identification of 12 bacterial pathogens at the species level. The broad-range PCR primer mixture was designed using conserved regions of the bacterial topoisomerase genes gyrB and parE. The primer design allowed the use of a novel DNA amplification method, which produced labeled, single-stranded DNA suitable for microarray hybridization. The probes on the microarray were designed from the alignments of species- or genus-specific variable regions of the gyrB and parE genes flanked by the primers. We included mecA-specific primers and probes in the same assay to indicate the presence of methicillin resistance in the bacterial species. The feasibility of this assay in routine diagnostic testing was evaluated using 146 blood culture positive and 40 blood culture negative samples.  相似文献

The seabuckthorn carpenter moth, Holcocerus hippophaecolus, which has a generation time of four years, is recently becoming one of the major pests of the seabuckthorn (Hippophae rhamnoides) in Inner Mongolia, Liaoning, Shanxi, Ningxia and Shaanxi of China (Hua et al., 1990). The larvae of the H. hippophaecolus mainly damage the stems and roots of the seabuckthorn, and the mature larvae pupate in the soil. The spatial distribution of the pupae was analyzed by using biostatistics and geostatistics in order to effectively control the insect and further study the spatial distribution of the population. Results show that most of the pupae (90%) had an eclosion time span from early June to the end of July. The sex ratio of the pupae was nearly 1:1 in the woodland samples. In addition, 24.3% of the 971 trees investigated had pupae and it ranged from 0 to 4 per tree within a distance of 1.3 m from the base of the stem. 90% of the pupae were aggregated within a distance of 1 m from the base of the stem. The pupae show intense spatial aggregation in the sampled woodland which had an 11.1 m spatial dependence and a 90.7% intensity in the local spatial continuity. Moreover, the population presented an intensive spotted distribution and many aggregated spots were found in the woodlands. As for the relationship between grid size and variogram of the pupae, the variations in the range, the intensity of local spatial continuity and the sill were all very low or non-existent when the grid size was 5 m, 6 m or 7 m. Whereas, the value of the decisive coefficient was the biggest when the grid size was 5 m making it the ideal grid size.  相似文献

研究于 1998~ 2 0 0 3年在库区 10个区县 ,35个调查点进行。研究时 ,库区蝴蝶的生活环境被划分为 80个小生境 ,设置样带6 6 1条 ;调查期间获得 (Sampled)蝴蝶 331种 ,10 780只个体。报道三峡库区蝴蝶群落的部分参数 :2 5 3种蝴蝶的小生境占有率低于 10 % ,占有 4 0 %以上小生境的蝴蝶只有 7种 ;在 6 0条以上样带都获得的蝴蝶有 16种 ,而在 10条以下样带内获得的蝴蝶 2 19种 ,占总数的 6 6 .2 % ;优势种 2 1个 (6 .3% ) ,常见种 12 5个 (37.8% ) ,少见种 10 3个 (31.1% ) ,罕见种 82个 (2 4 .8% ) ;相对多度高于 1%的种类 2 2个 ,仅占总数的 6 .6 % ;个体数分布的规律接近对数级数法则。三峡库区蝶类现在的分布反映了库区生境破碎化的结果 ,意味着适宜的生境斑块周围分布不适宜生境 ,种群受到面积、隔离、边缘等多种效应的影响 ,形成了现有的物种多样性及其空间格局  相似文献

Internucleoside coupling by the title reagent, illustrated by synthesis of dinucleoside and trinucleoside thiophosphates and selenophosphates is accomplished by a new strategy involving the displacement of 4-nitrophenoxy group. Evapotranspiration (E) and CO2 flux (Fc) in the growing season of an unusual dry year were measured continuously over a Scots pine forest in eastern Finland, by eddy covariance techniques. The aims were to gain an understanding of their biological and environmental control processes. As a result, there were obvious diurnal and seasonal changes in E, Fc, surface conductance (gc), and decoupling coefficient (Ω), showing similar trends to those in radiation (PAR) and vapour pressure deficit (δ). The maximum mean daily values (24-h average) for E, Fc, gc, and Ω were 1.78 mmol m−2 s−1, −11.18 µmol m−2 s−1, 6.27 mm s−1, and 0.31, respectively, with seasonal averages of 0.71 mmol m−2 s−1, −4.61 µmol m−2 s−1, 3.3 mm s−1, and 0.16. E and Fc were controlled by combined biological and environmental variables. There was curvilinear dependence of E on gc and Fc on gc. Among the environmental variables, PAR was the most important factor having a positive linear relationship to E and curvilinear relationship to Fc, while vapour pressure deficit was the most important environmental factor affecting gc. Water use efficiency was slightly higher in the dry season, with mean monthly values ranging from 6.67 to 7.48 μmol CO2 (mmol H2O)−1 and a seasonal average of 7.06 μmol CO2 (μmol H2O)−1. Low Ω and its close positive relationship with gc indicate that evapotranspiration was sensitive to surface conductance. Mid summer drought reduced surface conductance and decoupling coefficient, suggesting a more biotic control of evapotranspiration and a physiological acclimation to dry air. Surface conductance remained low and constant under dry condition, supporting that a constant value of surface constant can be used for modelling transpiration under drought condition.  相似文献
